SYNOPSIS. Inhibition analyses and reversal studies were made on approximately 300 compounds with the B12-dependent Euglena gracilis. Examples of competitive B12 antagonism were found among varied structures, including purines—notably 6-mercaptopurine—pteridines, and nicotinamides. Such compounds bear no structural analogy to the molecule of vitamin B12 and are designated simple B12 antimetabolites, in contrast to the general concept of antimetabolites, which implies close structural analogy between antimetabolite and metabolite. These studies suggest that such simple antimetabolites may act indirectly by interfering with cofactors closely concerned with the utilization of B12, presuming that such cofactors contain purine, pteridine or nicotinamide moieties.
