Summary Rod outer segments of the frog,Rana esculenta, were isolated in Ringer solution or double distilled water, spread on grids, sometimes degraded enzymatically, and negatively stained. Thin sections from embedments of isolated outer segments were examined in an electron microscope fitted with a goniometer stage without further preliminary treatment. It was concluded that the rod outer segment contains not only membranes, but also fibrils and tubules. No undisputable statement can as yet be made concerning the substructure of the membranes. It could be ascertained that fibrils are still extant in the outer segment even after treatment with pronase or phospholipase.
