    Notes: Low-angle light scattering results reported previously demonstrated that measurements on high molecular weight native DNA must be made at angles below 30° in order to obtain correct molecular weights. Earlier light-scattering data obtained on denaturated DNA at angles above 30° showed no change in molecular weight upon denaturation, even though other techniques clearly showed that strand separation occurred. This paper reports low-angle measurements on solutions of calf thymus and T7 DNA denatured under acidic conditions. The results demonstrate that a halving of molecular weight consistent with strand separation is detected by light scattering only when low-angle data are used to obtain correct molecular weights for native material. As expected from theoretical considerations, the scattering from denatured DNA is a linear function of sin2(θ/2), where θ is the angle of observation. This result shows that anticipated experimental artifacts have no significant effect on the low-angle measurements and demonstrates that the curvature in the scattering envelope observed for native DNA below 30° is an inherent property of the native molecule.

    Notes: A scheme of qualitative analysis is presented for the identification of the constituents of polyurethane foam products, or indeed polyurethane-type materials in general. Elastomeric and cellular polyurethanes are analyzed by hydrolysis to simpler compounds which are then separated and identified. The methods of identification employed include infrared and nuclear magnetic resonance spectroscopy, gas chromatography, and thin-layer chromatography.

    Notes: A simple inexpensive apparatus with a working volume of 10 liters of culture is described. Details of construction and procedures for operation of the cyclone column vessel are given. The vessel is self-contained, so that experimental parameters of temperature and aeration are individually controlled; homogeneous mixing and representative sampling of the culture, besides control of foam without need for antifoam, are obtained. The vessel may be used in single or multistage systems for aerobic or anaerobic cultivation of organisms in batch, chemostat, or phased cultures.

    Notes: An inexpensive scale-up of the cyclone column apparatus (Can. J. Microbiol., 11, 893 (1965)) is described which is suitable for pilot plant studies on the 10 liter scale. Details of its construction and operation are given.Experimental with phased cultures of C. utilis growing in the equipment are reported during which studies of dissolved oxygen and oxygen uptake were made over the cell cycle. Changes in both oxygen parameters occurred during the cycle and were useful as monitors of the growth. Cell of C. utilis were found to be very sensitive to changes in oxygen supply during the early stages of the cell cycle.

    Notes: The self-diffusion coefficient of methane has been measured from 150° to 350°K. and from 200 to 5,000 lb./sq. in. abs. At constant temperature, the density-diffusivity product is constant up to neighborhood of critical density and decreases sharply above that density. The temperature dependence of the low density data agrees with Chapman-Enskog theory. The Lennard-Jones (6-12) parameters determined from the low density data (∊/k = 130°K., σ = 3.85Å.) are in good agreement with those determined by other methods. A correlation for the self diffusion coefficient for methane has been developed which may provide predictions of other spherical nonpolar gases.

    Notes: Low-angle light scattering, sedimentation velocity, and intrinsic viscosity measurements have been made on circular and linear forms of lambda (λ) bacteriophage DNA. Available equations, used to relate hydrodynamic parameters of both forms to the molecular weight, give relatively consistent values of particle weights which essentially agree with the light-scattering results. An average molecular weight of (34 ± 3) × 106 for λ DNA was obtained in good agreement with literature values of (31-33) × 106. The linear λ DNA has a larger root-mean-square radius than the circular molecule, when determined by light scattering, but the difference does not appear to be us large as expected from hydrodynamic data. The two forms also show significantly different angular distrbutions of scattered light intensities which agree only qualitatively with those derived from existing theory. The light-scattering results suggest that further experiments and modifications of available theories should be undertaken.

    Notes: Efforts have been made to establish the chemical constitution of water-blown polyurethane foams. Specific reagents have been used to indicate the presence of biuret or allophanate linkages. Partial hydrolysis techniques have been used to degrade foams selectively and hence isolate parts of the chain network. These methods yield information which is not satisfactorily obtained from the examination of undegraded polymer and which is not available after total degradation. It has been shown that polyureatype linkages are present in the chain network of both polyester and polyether foams.

    Notes: Crystals of poor optical quality resist attempts to obtain polarised Raman spectra and produce high background levels of scattered radiation which make low frequency studies impossible. It is shown that the Brewster angle scattering technique, usually reserved for opaque or nearly opaque materials, can overcome these obstacles and yield spectra of acceptable signal-to-noise, low background radiation levels and with good polarisation data.
