    Notes: Several techniques for investigating the multiplicity and stability of open isothermal enzymatic reactors are discussed and some of the pitfalls in previous thinking pointed out. The example which is used to illustrate these methods exhibits several interesting features. Among these is the existence of a stable oscillatory state which surrounds a unique steady state which is asymptotically stable to certain finite disturbances.

    Notes: Gram-sized quantities of purified arginine, formylmethionine, glutamic acid, and phenylalanine-2 tRNAs have been prepared from pools of E. coli K-12 MO7 mixed tRNAs by reversed-phase chromatography after preliminary fractionation on DEAE-cellulose. Purified formylmethionine tRNA and partially purified arginine tRNA and glutamie acid tRNA were obtained from large-scale RPC-3 runs (4 × 36 in. column). The arginine tRNA was further purified by rechromatography on RPC-4 columns, and the gluatmic acid tRNA by rechromatography on an RPC-3 column. Two phenylalanine tRNAs were resolved on large-scale (2 × 96 in. column) RPC-3 runs; only the second phenylalanine tRNA reached a satisfactory degree of activity. About 0.88 g of arginine tRNA, 70% activity; 3.32 g of formylmethionine tRNA, 97% activity; 0.80 g of glutamic acid tRNA, 83% activity and O.92 g of phenylalanine-2 tRNA, 78% activity, were produced. The processing steps employed are reliable and reproducible and the procedure is amenable to routine production of these tRNAs.

    Notes: A method is described for the large-scale preparation of erythropoietin from anemic sheep plasma. DEAE-cellulose and carboxymethylcellulose column chromatography was used to prepare Step II erythropoietin. A total of 168 sheep yielded 499 liters of plasma from which 323,000 IU of Step II erythropoietin was obtained.

Enzyme activity yields range between 30 and 80% of the activity of the untreated preparations. Even fragile enzyme systems, for instance those using mobile cofactors, can be efficiently immobilized.     Notes: Candida utilis was grown in batch culture with and without oxygen control. The concentrations of A-, B-, and C-type cytochromes were found to vary with the initial glucose concentration, with the dissolved oxygen concentration, and with time. A-type was the most sensitive. After glucose was essentially exhausted, the yeast catabolized ethanol, if it had been growing in a relatively low initial glucose concentration, or non-glucose carbohydrate, including some of that previously accumulated within the cell, if it had been growing in a high initial glucose concentration. This difference in metabolic pattern could explain why cytochrome derepression was initiated soon after glucose uptake ceased only if initial glucose had been relatively low. The effects of glucose and dissolved oxygen concentrations on yeast cytochromes and respiratory activity are discussed.

    Notes: A high molecular weight intracellular enzyme of Bacillus brevis ATCC 9999 is released when the organism is disrupted by sonication of homogenization. However, both processes also degrade the enzyme. Assays for protein release and specific enzymatic activity of the released protein indicate that both release and degradation can be represented by first-order kinetic models. Utilization of the difference between the kinetics of release and degradation allows optimization in the recovery of this enzyme for both the sonication and homogenization processes.

    Notes: This study reports some findings on the death of mammalian cells occurring from fluid dynamic effects when they are pumped through capillaries. The cell strains used were human HeLa S3 and mouse L929, grown in monolayer culture. Cells were harvested and suspensions were pumped through various lengths and diameters of stainless steel capillary tubing. Viability of the cells was assayed by the dye exclusion test. Cell deaths occurred and could be correlated with either average wall shear or power dissipation within the capillary tube. L929 cells were found to be more sensitive than HeLa S3 cells at all shear rates tested.

    Notes: A flowsheet has been developed which leads to the purification of 10 transfer ribonucleic acids (tRNAs) from E. coli K-12 MO. Crude tRNAs were recovered by phenol extraction and two ethanol precipitation steps. The initial separation of the crude tRNA mixture was achieved by RPC-3 reversed-phase chromatography. Following rechromatography under other conditions, the following tRNAs were recovered at a purity of 70 to 100%: arginine, aspartic acid, glutamic acid, lysine, formylmethionine-1, formylmethionine-3, normal methionine, phcnylalanine-1, phenylalaninc-2, and valine. Recoveries for these tRNAs ranged from 15 to 60%. The flowsheet was demonstrated with chromatogaphic runs ranging from 1,300 to 250,000 A260 units per run. The chromatographic steps were simple and readily reproducible.

    Notes: Changes in the catabolic pattern of Saccharomyces cerevisiae, growing in continuous culture, were effected by altering the glucose feed rate or the dissolved oxygen concentration. The cytochrome concentrations and the adenosine phosphate pool level of the yeast in a series of steady states and during three transitions were measured and compared with the glucose uptake rate (QG), the respiration rate (QO2), and the rate of ethanolic fermentation (QE).Respiration was decreased at high glucose feed rates only if oxygen was low but cytochromes were glucose repressible at both high and low oxygen concentrations. In the main, QE and the levels of ATP, ADP, and AMP were decreased and cytochrome concentration were elevated at low QG values. No consistent relationship between any of the adenosine phosphate parameters and QO2 was discernible.Evidence is presented for the concept that the QG directly controls the adenosine phosphate pool level and that a relationship between the concentration of adenosine phosphate anhydride bonds and the adenosine phosphate level is constantly maintained.

    Notes: A rate equation has been derived to describe the hydrolysis of benzylpenicillin to 6-aminopenicillanic acid by penicillin amidase. The integrated from of the rate equation has been shown to predict satisfactorily the progress of the reaction in a batch reactor using either soluble or immobilized penicillin amidase. The rate equation was also used to predict the performance of a continuous feed stirred tank reactor containing immobilized enzyme. There was good agreement with experimental measurements.
