    Notes: Summary An autoradiographical study of the distribution of fast transported material in the cerebro-visceral connective of Anodonta cygnea (L.) following the injection of 3H-5-hydroxytryptophan into the cerebral ganglion has been carried out at both the light- and electronmicroscope levels. Statistical analysis of the results at the electron microscope level revealed that the distribution of grains within the tissues of the cerebro-visceral connective was highly non-random. Both the “circle analysis” and the “hypothetical grain density analysis” methods showed that most of the 3H-activity was associated with dense core vesicles (1000 Å diameter). These dense core vesicles were most frequently seen in the neurosecretory axons found near the periphery of the connective, but a number of labelled vesicles were also present in the small axons. The significance of these findings in relation to the distribution and the fast axonal transport of 5-hydroxytryptamine in the cerebro-visceral connective is discussed.

    Notes: Summary The nervous system of juvenile and adult Ceriantheopsis americanus has been examined with the electron microscope. The nervous system is exclusively ectodermal, forming a plexus of fibres lying between the epithelium and the muscle layer. The plexus consists of three types of nerve fibre that vary in diameter from 0.1 to 20 μ, and a fourth type of fibre of uncertain nature. The reticulum, previously thought to be the ectodermal nervous system, is redescribed as the peduncles of epithelial supporting cells. Intraneural and neuromuscular synapses are described. About 97% of the interneural are polarized. The unpolarized synapses are a variety not previously described, consisting of a series of polarized zones, for which the name multipolarized synapse is proposed. Structurally, the interneural synapses are more complex than those previously seen in the Cnidaria. Presynaptic projections, periodic cleft densities, and a postsynaptic web are described. By contrast, the neuromuscular synapses bear no membrane specializations. On the basis of observed synaptic interactions of its components a preliminary model of the functional organization of the nervous system is proposed.

    Notes: Summary Electron microscopy of the cerebro-visceral connectives of Anodonta cygnea shows numerous long rows of dense core vesicles within the smaller axons of the connectives. These vesicles are often linked together by bridges of electron-dense material reinforced by one or more “arms”. The use of the tilting stage on the electron microscope, which allows the bridges to be examined from different angles, confirms the presence of these vesicular connections. Sub-cellular fractionation of the cerebro-visceral connectives was carried out to study the nature of these bridged dense core vesicles. In the negatively stained vesicle fraction two or more dense core vesicles are joined in a row by connections similar to the vesicular bridges of the intact connective. In the fixed vesicle fraction, many of the dense core vesicles are connected by bridges but long rows of bridged vesicles are not seen. The functional significance of the vesicular bridges is discussed. Preliminary findings of this work have been reported previously (McLaughlin and Howes, 1973).

    Notes: Summary The radial nerve cords of a number of different species of echinoids and asteroids have been examined with the electron microscope. They have been shown to consist of tracts of small naked axons with discrete regions of neuropile. Synapses within these areas of neuropile have been described and their significance discussed. A careful examination has failed to reveal any connections between the ectoneural nerve and the mesodermal nerves and muscles. Synapses across the connective tissue sheet separating these regions have been proposed to account for known functional connection. A hypothetical model of the nervous system has been proposed to account for known structure and function.

    Notes: Summary The avascular ventral nerve cord of the moth, Manduca sexta, possesses an extensive dorsal mass of connective tissue in which lie fibroblasts that produce a collagen-like protein. The lateral and ventral surfaces of the nerve cord are ensheathed by an acellular neural lamella. Beneath this lies a layer of microtubule-laden perineurial cells which are separated from one another at their peripheral borders by lacunae containing electron-opaque material to which the cells are attached by hemi-desmosomes. Beyond these spaces, narrow intercellular clefts occur between the interdigitating perineurial plasma membranes; these are then connected by both gap and tight junctions. The axons beneath are surrounded by glia which also contain many microtubules and which are linked to one another by desmosomes and tight junctions. When intact nerve cords are incubated in horseradish peroxidase, reaction product is subsequently found within the neural lamella as well as in the lacunae and clefts between perineurial cells, but not beyond this level. Desheathed preparations, however, contain peroxidase within the cytoplasm of the exposed glial cells. Lanthanum penetrates the neural lamella and the lacunae, clefts and gap junctions between adjacent perineurial cells, but no further. It therefore appears that the tight junctions in the perineurium may be the site of restriction to the entry of ions and molecules, the existence of which has been suggested previously by electrophysiological investigations.

    Notes: Summary By combining the Golgi and the electronmicroscope techniques it has been possible to identify accurately the system of centrifugal fibers which arborizes in the lamina of muscoid flies forming the so-called nervous bags. Each of them originates from a single fiber entering the lamina at the site in which the second order and the long visual fibers leave it. This single fiber represents the peripheral portion of a T-shaped trunk stemming from a small neuronal body located in the external region of the medulla. The central branch terminates within the first synaptic field of this visual center. After entering the lamina the centrifugal fiber ramifies profusely and its branches can be seen climbing and synapsing on the surface of the photoreceptor axon endings. The synaptic loci show characteristic synaptic ribbons located within the nervous bag fibers. This fact suggests that direction of conduction is from the medulla to the lamina. This study has also revealed that the intramedullar terminals of the centrifugal fibers establish intimate contacts with one of the two second order fiber endings.

    Notes: Summary Basal cells of the hard palate epithelium have been examined electronmicroscopically. Secretory granules, morphologically similar to those of the Merkel cells, occur in the groups of basal cells. A comparison concerning the cytoplasmic qualities of the two cell types is given, and the possibility of a cytogenetical or physiological relationship between the Merkel cells and the basal cells is discussed.

    Notes: Summary The morphology of the neuro-epithelial relations in the snout of the pig was studied with the light microscope and with the transmission electron microscope. The epithelial cells form lamellae which surround the intraepithelial nerves. These lamellae resemble the lamellae of the subepithelial bulbous corpuscles. Membrane contacts occur between the basal cells and the subepithelial cell profiles. Extrusions of the epithelial basal lamina are described. These morphological observations are discussed in relation to earlier studies. It is assumed that the epithelial cells may play a role in the formation of the subepithelial receptor organs.

    Notes: Summary Nerve fibres, running longitudinally as well as circularly between the core lamellae in the Herbst corpuscle are described. These fibres are morphologically different from the central afferent axon. They are most frequently observed in the outer part of the core, and contain inter alia numerous agranular vesicles measuring approx. 450 Å in diameter, dense core vesicles with a diameter approx. 800 Å and microtubuli (250 Å). Occasional specialized junctions are seen between the nerves and the neighbouring lamellae.
