    Notizen: The electronic absorption and circular dichroism spectra of the DNA-acridine orange complex have been measured over a range of ionic strength, pH, and DNA phosphate to dye (P/D) ratios. Three circular dichroism bands associated with the long wavelength absorption band of acridine orange are induced on complex formation with DNA. Two of the dichroism bands, due mainly to dimeric dye molecules, are favored by low ionic strength, low pH (3.2), and a low P/D ratio (∼3), while the third, deriving primarily from monomeric dye, is optimum at high ionic strength, neutral pH, and a larger P/D ratio (9). The data suggest that monomeric acridine orange binds to DNA in the form of a left-handed helical array with four dye molecules per turn, while the bound dimer has a skewed sandwich conformation which is itself dissymmetric. The stereochemical relations between the bound monomer dye and the DNA are consistent with a modified intercalation model for the DNA-acridine complex.

    Notizen: The electronic absorption and circular dichroism spectra of the complex formed by acridine orange with poly-α,L-glutamic acid in the α-helix conformation have been measured in aqueous solution over a range of glutamate residue-to-dye ratios. Three Cotton effects (circular dichroism bands) associated with the long wavelength absorption band of acridine orange at 4950 A. are induced by complex formation between the dye and the polypeptide, and further circular dichroism bands are observed in the ultraviolet region associated with the 2700 A., but not with the 2950 A. absorption band of the dye. The induced optical activity is found to be relatively insensitive to the glutamate residue-to-dye ratio and to be more dependent upon the ionic strength of the solution. By Measuring the circular dichroism spectrum of the complex in aqueous solution under streaming conditions with the light propagated along the direction of flow the observed circular dichroism bands are assigned to electronic transitions polarized parallel or perpendicular to the axis of the polypeptide α-helix. From the spectroscopic data it is inferred that the dye aggregate in the L-PGA-AO complex has the form of a left-handed superhelix bound to the core of the right-handed α-helix of poly-α,L-glutamic acid. It is shown that the longer and the shorter of the in-plane axes of the dye molecule are probably orientated respectively at a small angle, and radially, with respect to the axis of the α-helix in the complex.

    Notizen: Longitudinal liquid mixing in fluidized and fixed beds was studied using sinusoidal and pulse response techniques. The tracer used was light emissive fluorescein dye. A systematical study of liquid phase dispersion by varying particle size, fluid velocity, fraction voids, and particle density was conducted. A generalized correlation applicable to both fixed bed and fluidized bed was obtained. The application of the correlation in predicting the effect of the dispersion on reactor performance was discussed.

    Notizen: The ability to express laboratory melt flow data in generalized terms serves both the fabricator and the resin producer: the fabricator can reduce the relations to the particular conditions of his process; the resin producer can assess the effect of polymer composition and structure on melt flow properties. An empirical method of correlating and generalizing laboratory capillary viscometer data has been developed which allows prediction of pressure-output relations for any cylindrical orifice. It is based on the Rabinowitsch and Bagley methods of calculating the correct shear rate and shear stress. The pressure drop across a die is made up of two terms, the entry pressure and the land pressure. Each is a function of the shear rate, but only the latter is a function of the die dimensions. Six key rheological parameters are developed, using polypropylene as an illustrative example. The effect of molecular weight and molecular weight distribution of polypropylene on the parameters is shown. In addition, some examples of practical uses of the generalized formulations are given. In the examples, the shear rate range of 10-1000 sec.-1 is covered, although the method could be applied to any experimentally attainable shear rates.

    Notizen: The ionization potentials of methyl-, t-butyl- and trimethylsilyl-substituted pyridines have been determined by high-resolution photoelectron spectroscopy. The results are discussed in terms of destabilizing inductive and stabilizing conjugative effects of the substituents, and have been parametrized on the basis of a HMO-type perturbation treatment. Our data are in accord with the idea that the topmost occupied orbital in pyridine is a ‘lone pair’ orbital.

    Notizen: The success of material implants in the vascular system has been limited by the fact that the intial events of blood coagulation on foreign material surfaces are not understood. Many materials have been evaluated in the search for a suitable implant material in this system. Recent success of porous ceramic materials in bone-ceramic compatibility studies stimulated the feeling that these materials should be tested in other body tissues such as the blood.Ceramic disks of Al2O3 and TiO2 were evaluated using a contact angle measurement technique to determine the characteristics of these material surfaces before and after exposure to a blood environment. This technique involved measuring the contact angle of sessile drops of various liquids on these surfaces with a microscope fitted with a goniometer eyepiece. By plotting these contact angles according to the procedure of Zisman, the characteristics of the surfaces could be evaluated.The clean ceramic disks were found to be extremely wettable even after exposure to a physiological saline environment. On exposure of these disks to a blood environment, it was observed that some type of film was adsorbed from the blood onto the ceramic surface during the initial minutes of exposure. This film had wetting characteristics consistent with layers of fibrinogen.

    Notizen: In this investigation, ceramics were studied to determine their role as rigid, abrasive implants in soft, living tissue. Discs and tubes of three ceramics, CaO·Al2O3, CaO·TiO2, and CaO·ZrO2, were introduced as porous and non-porous structures into muscle and connective tissue sites in rabbits. The animals were observed grossly to determine the duration of redness and swelling following surgery, and samples were retrieved at 1 week, 3 months, 6 months, and 9 months after implantation. A mild, acute inflammatory response immediately followed the implantation of all three materials in both the porous and non-porous forms. Histological sections of the ceramics and surrounding tissue, cut and stained for light microscopy, demonstrated the absence of inflammatory cells and revealed the normal morphology and organization of the cells present around all types of implants tested. Tissue around discs of porous ceramics healed faster and exhibited thinner fibrous encapsulations than with impervious discs of the same material. Healthy fibrous connective tissue with an ample blood supply occupied those implants with pores of 45-100 mμ, and even more rapidly filled the samples with a 100- to 150-μ pore size. The tissue ingrowth and tight adherence to the porous samples was believed responsible for the more moderate response to porous implants. No adverse responses of any kind were observed, except in a very few, atypical specimens.
