    Notes: A novel method is described for the preparation of stable glass capillary columns (glass open tubular columns), including the etching and formation of a polymer film on the inner glass capillary surfaces. The approach used here is based on low-temperature plasma etching and polymerization. Under the influence of a field of radio frequency discharge, low pressure gases of fluoric compounds, introduced into the glass capillary tube, generate excited fluorine radicals which etch the inner surface. The plasma of organosilicone monomer in the glass capillary yields a uniform polymerized film on the inner surface. The resultant material functions as a good stationary phase for glass capillary gas chromatography (GC2). The inner surfaces treated with such a plasma, can be studied by means of a scanning electron microscope (SEM). The flexibility of this method permits the use of various stationary phases and surface modification.

    Notes: Group-specific polysaccharides isolated by means of a cetavlon procedure are immunogenic in man and induce protective immunity against meningococcal meningitis. Minute quantities of the polymers in solution can act as vaccines. We now report the first characterization of a fractionated (C-1) group C polysaccharide in 0.4KM KCl and 0.05M sodium acetate by means of light-scattering spectroscopy. Independent measurements of refractive index increments, absolute scattered intensities, angular scattering intensities and line widths as a function of scattering angles and delay times at different concentrations using incident wavelengths of 632.8 nm from a He-Ne laser and of 488 nm from an argon-ion laser yield information on aggregation properties, molecular weight (Mr), radius of gyration 〈r0g〉1/2z, translational diffusion coefficient 〈D〉0z, and second virial coefficients A2 and B2 of C-1 polysaccharide.At relatively high ionic strength (0.04M KCl + 0.05M sodium acetate), we obtain for the C-1 polysaccharide in solution Mr = 5.15 × 105, 〈r2g〉1/2z = 345 Å, A2 = 1.25 × 10-4 ml/g, 〈D〉0z = 1.16 × 10-7 cm2/sec with a corresponding Stokes radius of 240 Å and B2 = 4.4 ml/g. A2 and B2 are the second virial coefficients from intensity- and diffusion-coefficient measurements. The C-1 polysaccharide aggregates in solution and behaves hydrodynamically like random coils. Viscosity and sedimentation studies further confirm our conclusions that the fractioned C-1 polysaccharide aggregates in solution and EDTA can partially break up those aggregates. However, the system remains polydisperse even after adding an excess amount of EDTA. The weight-average molecular weight of the C-1 polysaccharide in solution depends upon ionic strength and exhibits a minimum at ∼0.2M KCl. Finally, viscosity, light-scattering, and sedimentation results all show that the aggregated macromolecular system behaves like random-coiled polymers with no measurable shape factors.

    Notes: Partially purified glucose isomerase from a Streptomyces species was immobilized on porous glass particles and studied for various characteristics concerning its use as an industrial catalyst. The activities were investigated in relation to the reaction parameters and the enzyme deactivation was studied systematically under various reaction conditions. The half-life of the immobilized enzyme was found to exceed 200 days at 50°C. The rate equation of the reversible glucose ⇄ fructose reaction was derived and the kinetic constants were determined. The rate equation was found to be in good agreement with experimental data for both forward and reverse reactions. The degree of diffusional effects was experimentally measured and theoretically analyzed.

    Notes: Tower cycling fermentors for the production of single-cell protein from volatile substrates were studied. The mass transfer, mixing and circulation patterns, and residence time distribution (RTD) curves were investigated in these vessels. This study suggests that the tower cycling fermentors for volatile substrates fermentation may improve product yields and at the same time reduce the power consumption, thereby resulting in a significant increase in operating cost savings and capital profits. The results of this research further indicate a future potential for commercial scale tower cycling fermentors.

    Notes: α-Chymotrypsin was light sensitized by acylating with cis-cinnamoyl ester, a substrate interconvertible to the trans form by ultraviolet (UV) light. The degree of acylation by this method was complete leaving no residual activity of the enzyme. Upon UV irradiation the inhibited enzyme regained about 70% of its original activity, thereby adding light-sensitiveness to the proteolytic enzyme. In seeking a photographic application of the light-sensitized enzyme, a pigmenting enzyme was incorporated with it. The coupled enzyme system was shown to exhibit a light signal in the form of dark pigment slurry.

    Notes: The reliability of the process of Ramanathan and Gaudy (Biotechnol Bioeng., 13, 125 (1971)) for the completely mixed activated-sludge process holding the recycle cell concentration, XR, as a system constant with respect to step changes in hydraulic retention time was investigated. The experiments were run at initial dilution rates of ⅛, ⅙, ¼, and ½ hr-1 treating a soft drink bottling wastewater. The influent substrate concentration was maintained at 1000 mg/liter chemical oxygen demand and the hydraulic recycle ratio at 0.3. The recycle sludge concentration was maintained at about 7000 mg/liter.It was found that the system could accommodate hydraulic shock loads up to 200% positive changes and down to 50%negative changes without disruption of the effluent quality. Shorter retention time of the range studied, from 2 to 8 hr, has the advantage of shorter response time with respect to the response of the concentration of biological solids in the reactor.

    Notes: The gas-liquid oxygen transfer rate is a key step in the production of antibiotics in submerged fermentation. If the gas-liquid oxygen mass transfer rate is not equal to the required liquid-solid oxygen mass transfer rate at a particular cell concentration, then productivity of the particular fermentation operation will not be the maximum possible value.One way to increase the productivity of a given fermentation tank installation is to increase the cell concentration and to increase the oxygen transfer by changing the mixer and air supply to match the new requirements. In order to evaluate the cost of making this change to the larger mixing equipment, a typical cost example is given which can easily be modified for other combinations of production cost and mixer cost. As an example, it is seen that a considerable savings can result from a given installation by primarily changing the oxygen transfer ability of the equipment to utilize a given fermentor more efficiently. Production cost savings of 8 to 25% are shown in the example cited.

    Notes: The interaction energies (X1) between the polymers polypropylene, poly(vinylidene chloride), sulphonyldiphenyl formaldehyde resin and poly(vinyl pyrrolidone) and the solvents toluene, ethylbenzene, isopropylbenzene, ortho, meta, and para-xylene are determined by gas chromatography. As shown in earlier publications, this technique for the determination of X1, can be extended to similar systems of theoretical as well as of practical importance, although the selection of the present systems was primarly made on the fact that these systems are of industrial interest and have not so far been investigated by gas-liquid chromatography. The studies were carried out at two temperatures and the results obtained show a decrease of X1 with an increase of temperature.

    Notes: A thorough survey of the past and current available literature shows that the diffusion of the solvents toluene, ethylbenzene, ortho-, meta-, and para-xylenes in polypropylene has not been studied so far, although such studies find practical applications in various industrial problems. Diffusitives and the activation energies of the above mentioned solvents in polypropylene were determined in the temperature range of 45-120°C by gas chromatography. The values obtained in this communication compare fairly with those determined by conventional techniques for analogous systems, confirming the reliability of the gas chromatography as a powerful tool for diffusion measurement and the authenticity of the data presented herein.

    Notes: The glass transition temperatures, Tg, of polystyrene, poly (vinyl chloride) and poly(methyI methacrylate) have been determined from gas chromatographic measurements using n-hexane, n-heptane, meta-xylene and para-xylene solvents. The glass transition temperatures were detected on the z-shaped retention diagrams which were produced from the plot of the logarithm of the specific retention volumes of the above-mentioned solvents against the reciprocal of temperature, i.e. log Vgº vs. 1/T. The glass transition temperature is specified by the temperature where the slope of log Vgº vs. 1/T changes abruptly. The observed glass transition temperature of polystyrene produced by this technique was found to be in good agreement with those produced by other techniques such as the differential scanning colorimeter.The industrial importance of the glass transition temperature, Tg, might be due to the dramatic changes in the physical properties of the polymer, such as hardness and elasticity, which take place in the vicinity of this temperature. However, perfectly crystalline polymers do not exhibit glass transitions, because their chains are incorporated in regions of three-dimensional order, called crystallites. Completely amorphous polymers and semi-crystalline polymers usually exhibit both glass transition and melting.
