    Notes: Various biochemical properties of mitochondria isolated from primary monolayer cultures of mammary epithelial cells from mid-pregnant or hormonally stimulated mice were examined daily for seven or eight days. When compared with mitochondria from mammary glands of mid-pregnant animals, the specific activities of several mitochondrial enzymes were greatly reduced in cells after seven days in culture. There was a 5- to 6-fold reduction in the specific activities of cytochrome oxidase, succinate dehydrogenase and α glycerophosphate oxidase while malate dehydrogenase and adenylate kinase activities were 2- to 3-fold lower. The reduction in mitochondrial enzyme activities was gradual and related to the length of time the cells were in culture. Progressive changes were also seen in the electrophoresis profiles of mitochondrial proteins in SDS-urea polyacrylamide gels. Mitochondria isolated from 1-, 2-, 3- and 8-day cell cultures showed a continuous reduction in the relative amounts of several mitochondrial polypeptides in the gel profiles. Addition of 35S-methionine to cell cultures for short and long periods indicated that mitochondrial protein synthesis continued throughout the 8-day culture period. However, the synthesis of several particular polypeptides was reduced progressively during the culture period. These studies indicate that mouse mammary epithelial cells have a lowered capacity for respiratory metabolism as a result of specific mitochondrial alterations which might be associated with the general loss of differentiated morphology by those cells during monolayer culture.

    Notes: We have studied the effects of theophylline treatment on pigmentation characteristics and growth of two B16 melanoma cell lines, HFH-18 and P/140. Cell counts of control and theophylline-treated cultures confirmed that the drug inhibits cell growth. Light and electron microscope cytochemistry with the L-dopa reaction indicated that the two cell lines differ in their ability to transfer Golgi-associated tyrosinase to developing premelanosomes. The results of these experiments, considered with results of electrophoretic analyses and activity measurements by the Pomerantz method, also provide evidence that increased tyrosinase synthesis occurs in response to theophylline treatment. In addition, results indicate that theophylline induces changes in the rate of synthetic or degradative posttranslational modification of tyrosinase. Measurements of intracellular cyclic AMP levels by radioimmunoassay in control cultures and in theophylline- and α-MSH-treated cultures were made. Although the hormone induced spectacular increases in cyclic AMP levels, theophylline produced no detectable change. These results indicate that theophylline differs from α-MSH because theophylline-induced changes in pigmentation may not require the participation of intracellular cyclic AMP.

    Description: Techniques to determine the environmental effects from the space shuttle SRB (Solid Rocket Booster) exhaust effluents are used to perform a preliminary climatological assessment. The exhaust effluent chemistry study was performed and the exhaust effluent species were determined. A reasonable exhaust particle size distribution is constructed for use in nozzle analyses and for the deposition model. The preliminary assessment is used to identify problems that are associated with the full-scale assessment; therefore, these preliminary air quality results are used with caution in drawing conclusion regarding the environmental effects of the space shuttle exhaust effluents.

    Description: An experimental study was conducted using a plate test bed having a turbulent boundary layer to determine the momentum transfer to the faces of step/cavity combinations on the plate. Experimental data were obtained from configurations including an isolated configuration and an array of blocks in tile patterns. A momentum transfer correlation model of pressure forces on an isolated step/cavity was developed with experimental results to relate flow and geometry parameters. Results of the experiments reveal that isolated step/cavity excrecences do not have a unique and unifying parameter group due in part to cavity depth effects and in part to width parameter scale effects. Drag predictions for tile patterns by a kinetic pressure empirical method predict experimental results well. Trends were not, however, predicted by a method of variable roughness density phenomenology.

    Description: A test facility operating at substantially reduced pressures is described and shown to be suitable when used with an open-circuit turbulent boundary layer wind tunnel. The facility is capable of producing flows with momentum Reynolds number in the range 200-10,000 by varying the ambient pressure. At low-Reynolds-number flows the pressure is low enough to allow the tunnel to operate with large values of freestream speed. The tunnel's reliability is verified by examining flows where momentum Reynolds number is greater than 1200. The results skin friction coefficients, wake parameters, and basic velocity profiles - consistently agreed with well-established data. For momentum Reynolds number from 600 through 1200 the resulting data indicate that the 'universal velocity profile' is accurate and correctly describes the mean flow properties. For Reynolds number from 425 through 600, the viscous sublayer in this range can no longer maintain its characteristic size.

    Description: Experimental values of shock shapes (alpha = 0 degrees and 10 degrees) and static aerodynamic coefficients (alpha = -4 degrees to 12 degrees) for sharp and spherically blunted cones having cone half-angles of 30, 45, 60, and 70 degrees and nose bluntness ratios of 0, 0.25, and 0.50 are presented. Shock shapes were also measured at 0 degree angle of attack by using a flat-faced cylinder (90 degree cone) and a hemispherically blunted cylinder (sphere). All tests were conducted in helium (gamma = 5/3) at a free-stream Mach number of 20.3 and a unit free-stream Reynolds number of 22,400,000 per meter. Comparisons between measured values and predicted values were made by using several numerical and simple engineering methods.
