    Notes: The role of adenosine 3′,5′-monophosphate (cyclic AMP) in the regulation of mouse melanoma cell growth and differentiation was investigated. A variant melanoma (Cloudman S91-F) which displays a greater degree of transformation than the parental cell (Cloudman S91) was isolated. A correlation between cyclic AMP metabolism and transformation was made. Dibutyryl cyclic AMP depressed cell growth and increased pigmentation in both parental and variant cell lines. The parental cell line, however, was more responsive to melanocyte-stimulating hormone (MSH) which was found to affect cell growth and pigmentation by increasing cyclic AMP levels. The more transformed S91-F cell line contained lower levels of cyclic AMP than the parental cell line, and this fact correlated well with the higher degree of growth and lesser degree of pigmentation in the variant. Enzymatic analysis revealed that the hydrolysis of cyclic AMP in both cell lines was similar, while the adenylate cyclase activity of the variant cell line was lower than that of the parental cell line. Lineweaver-Burk plots demonstrated that the Km′s for the enzymes in the two cell lines were the same but that the Vmax of the S91-F cell line was significantly less than that of the S91 cell line. Thus, the lesion in the S91-F cell which is responsible for its more transformed characteristics seems to be one which affects adenylate cyclase at the level of the cell membrane.

    Notes: The alkali cation content of HeLa cells is independent of culture density and of whether the cells are grown in suspension or attached to the culture vessel. With a cell doubling time of 28 hours, the cell K content turns over approximately once per hour. Following partial blockade of the alkali-cation transport system with ouabain, two distinct but interrelated mechanisms operate in the cellular response: (a) an increase in intracellular Na stimulates the pump so that the short-term alteration in electrolyte composition is less than would be expected from the fraction of pump sites inhibited, and (b) there is a cyclo-heximide-sensitive recovery in transport capacity reflecting a restoration of functional transport sites to their normal density on the cell surface. Experimental manipulations that mimic the effect of ouabain lead to a stimulation of transport, but they do not result in an increase in the number of ouabain-binding sites on the surface. The data are consistent with a four-to-six-hour turnover of transport sites at the surface, but there is no evidence for a specific induction of the transport system within this short-term recovery period.

    Description: A multiple pulse sequence (8-pulse sequence) used for high-resolution solid state NMR is analyzed with regard to the information available from each of the four wide sampling windows. It is demonstrated that full quadrature phase information can be obtained using only a single phase detector and that, for the commonly encountered situation where the spectral width is much less than the folding frequency, the signals from the various windows can be combined easily using standard complex Fourier transform software. An improvement in the signal-to-noise ratio equal to the square root of 3 is obtained over either standard single or quadrature phase detection schemes. Procedures for correcting spectral distortions are presented.

    Description: A brief overview is provided of the coherent Doppler lidar work at the Marshall Space Flight Center, taking into account the status and planned development of the system to be used for severe storms research. The considered system is a CW Doppler lidar operating at a wavelength of 10.6 micrometers. This lidar uses a 20 watt carbon dioxide laser. It has a range which is continuously variable from 60 m to approximately 600 m. Range variation is achieved through changing the focus of a 30 cm f/2 Cassegrainian telescope. Successful measurements with the CW lidar have included two and three dimensional wind profiles, wing tip vortex tracks and profiles, dust devil tracks and profiles, and transverse as well as line-of-sight wind velocities. The pulsed lidar in recent ground tests has succeeded in measuring clear air returns to a range of 13 km and has provided substantial information on velocities associated with the gust fronts of thunderstorms.

    Description: The results of the Workshop on the Need for Lightning Observations from Space held February 13-15, 1979, at the University of Tennessee Space Institute, Tullahoma, Tennessee are presented. The interest and active involvement by the engineering, operational, and scientific participants in the workshop demonstrated that lightning observations from space is a goal well worth pursuing. The unique contributions, measurement requirements, and supportive research investigations were defined for a number of important applications. Lightning has a significant role in atmospheric processes and needs to be systematically investigated. Satellite instrumentation specifically designed for indicating the characteristics of lightning are of value in severe storms research, in engineering and operational problem areas, and in providing information on atmospheric electricity and its role in meteorological processes.
