    Notes: Abstract Two loci account for all genetic variation resulting in difference in electrophoretic mobility in three hemoglobins (Hb1, Hb2, and HbX) in the hemolymph of the brine shrimp. Four α alleles and nine β alleles have been studied. In shrimps of all genotypes and in electrophoresis in media with varying degrees of molecular sieving, Hb2 is approximately equidistant from Hb1 and HbX. A shrimp heterozygous at both loci has a three-banded Hb1, a four-banded Hb2, and a three-banded HbX. We conclude that Hb2 contains n α-polypeptides and n β-polypeptides. Hb1 contains 2n α-polypeptides. HbX contains 2n β-polypeptides. During electrophoresis, the three native hemoglobins undergo reversible dissociation to n subunits. Subunits with the same charge reassemble to migrate as molecules of the same size as the native molecules. Although there is no evidence for an additional polypeptide in the three hemoglobins, we cannot exclude such a possibility. If it exists, it is under three constraints: (1) it must be present in equal amounts in each of the three hemoglobins; (2) it must have the same molecular weight as the α- and β-polypeptides; and (3) it must be free of genetic variation (detectable by electrophoresis).

    Notes: Abstract Electrophoretic mobilities of three hemoglobins (Hb1, Hb2, and Hb3) were studied in 15 populations of brine shrimps. Genetic segregation data support the model that Hb2 contains n α-polypeptides and n β-polypeptides; Hb1 contains 2n α-polypeptides. Hb3 contains neither α- nor β-polypeptides. There is no evidence of linkage of α and β loci with each other or with the locus (or loci) which governs Hb3 or with the nonhomologous portion of the sex chromosomes. Hemoglobins of different populations may be hybridized in vitro by incubation at high temperature. Reversible dissociation to subunits which contain only one (α or β) polypeptide occurs at 40 C (for Hb1) and at 50 C (for Hb2).

    Notes: Summary The foot epithelium of the slug Agriolimax reticulatus (M) can function as a digestive epithelium and possesses a vacuolar system in which heterophagic and autophagic material is hydrolysed. Using ferritin and peroxidase as markers, the process of endocytosis has been demonstrated at the level of the electron microscope. The mechanisms involved with the uptake of each marker differed in that ferritin was confined to fairly large vacuoles, while peroxidase was present in smaller pinocytotic vesicles which ultimately fused to form larger vacuoles or multivesicular bodies. Cytochemical tests for acid phosphatase revealed that the latter bodies acquired hydrolytic enzyme to become secondary lysosomes. Acid phosphatase was also localized in the Golgi apparatus of the epithelial cells. In addition to the endocytotic activity, evidence for autophagy was found in the epithelial cells and this process was apparently enhanced during starvation.

Die im Übergangszustand einer Dissoziation vorhandene Überschußenergie („non fixed energy“) ist mit der von Reaktionen in Lösung vergleichbar. Allgemein gilt, daß nur die energetisch günstigsten Zerfallswege miteinander konkurrieren können. Die Analyse von Übergangssignalen der unimolekularen Zerfälle metastabiler Ionen liefert Informationen über die bei der Reaktion freigesetzte kinetische Energie („kinetic energy release“), T; bei Experimenten in Lösung sind diese äußerst wertvollen Daten aufgrund von Stößen nicht zugänglich. Die Chemie von Ionen kann bequem studiert werden, wenn man die Ionen in einem Massenspektrometer erzeugt und die Zerfälle metastabiler Ionen analysiert.

    Notes: Organic ions decompose in the field-free regions of a mass spectrometer after a relatively well-defined lifetime (t ≈ 10-5s). This lifetime is very long compared with the time-scale of molecular bond vibrations and consequently the excess internal energy (“non fixed” energy) present in the transition state for dissociation is comparable with that found in solution experiments. In general, only the energetically most favorable decay routes are able to compete. Moreover, information concerning kinetic energy release (T) accompanying unimolecular decomposition may be obtained from a consideration of the metastable peak for the process; these extremely valuable data are inaccessible to solution experiments because of the effects of collisions. The chemistry of ions may therefore be investigated conveniently by generating the ions of interest in a mass spectrometer and analyzing the reactions which occur in metastable transitions.
