    Notes: A fermentor system with an external filtration loop has been developed to control the growth and sporulation of yeast in a single vessel. Excess growth medium, instead of being removed by centrifugation, is removed by filtration and replaced with acetate sporulation medium. The technique did give 80% sporulation after 20 hr, greatly improving the rate and degree of synchrony of sporulation and it also eliminated the contamination hazard of the conventional harvest technique, centrifugation, and resuspension of vegetative cells in sporulation medium. Furthermore it permits proper control of the environmental conditions throughout the growth, exchange, and sporulation phase. In this technique 100% recycle of biomass is achieved without any packing of the cells on the filter. This technique has wide application in the study of industrial fermentation that involves microbial differentiation such as the production of ergot alkaloids, bacitracin, and cephalosporin.

    Notes: The black yeast-like fungus Rhinocladiella mansonii NRRL Y-6272 elaborates an extracellular polysaccharide composed of N-acetyl D-glucosamine and N-acetyl D-glucosaminuronic acid residues in a 2:1 molar ratio. Production of this polymer was studied in 300 ml flasks and 20 liter fermentors. On the basis of glucose utilized, 13% conversion to polymer was obtained in four days at 25°C in medium containing yeast autolysate paste, K2HPO4, MgSO4, ZnSO4, L-asparagine or urea, and D-glucose. Nitrogen sources, aeration, agitation, temperature, and salts affected yields.The polymer in diluted fermentation broths was precipitated with ethanol in the presence of an electrolyte and collected by centrifugation. The viscosities of aqueous resolutions containing 1 and 1.5% of the polymer were 9400 cP and 21,500 cP, respectively.

    Notes: We have measured the specific viscosity of closed circular PM2 DNA in the presence of concentrations of ethidium bromide ranging up to 5 mg/ml. Both native viral PM2 DNA I and enzymatically prepared relaxed, closed circular PM2 DNA I0 exhibit a complex dependence of the specific viscosity upon the extent of supercoiling. As the number of superhelical turns is increased in the positive sense from zero, the viscosity first decreases to a minimum, then passes through a secondary maximum, and eventually again increases as the dye-induced duplex unwinding proceeds. In the case of DNA I, a corresponding behavior is mirrored in the negative sense as dye is removed from the principal viscometric maximum (complete relaxation of the DNA by dye). The shape of the curve relating specific viscosity to extent to supercoiling is similar for superhelical DNAs of either handedness, a result which we interpret to mean that the influence of any regions of special secondary structure (such as denatured loops) upon the viscosity is minimal. At very high dye concentrations the specific viscosity decreases dramatically. This effect might arise either from intermolecular aggregation or from a dye-induced collapse in the DNA secondary structure.

    Notes: Two black yeast polysaccharides show similar non-Newtonian behavior when the effects of polysaccharide concentration, shear rate, pH, salt, and temperature on the viscosity of dispersions are examined. Both polysaccharides readily form viscous aqueous dispersions and can be cast into flexible films, and both stabilize oil-water emulsions. Each of these two polysaccharides occurs extracellularly in the culture fluids of two different black yeast-like fungi - Rhinocladiella elatior Mangenot (strain NRRL YB-4163) and Rhinocladiella mansonii (Castellani) Schol-Schwarz (strain NRRL Y-6272). This report appears to be the first on the rheology of an extracellular polysaccharide which contains the unusual sugar N-acetyl-D-glucosaminuronic acid.

    Description: The characteristics of a thick hypersonic boundary layer turbulent for a length of 175 cm on a 4 deg sharp wedge were measured. The resulting boundary layer was free from transverse curvature effects and only mildly affected by upstream history effects caused by pressure and wall temperature gradients. Heat-transfer distributions were used to locate regions of laminar, transitional, and turbulent flow at an edge unit Reynolds number of 470,000 cm at wall-to-total temperature ratios from about 0.3 to 1. Wall cooling had little effect on the location of the transition region. Pitot and total temperature profiles and skin-friction measurements were obtained at several locations along the model longitudinal centerline. Mixing length and turbulent Prandtl number distributions were derived from the fully turbulent mean profiles.

    Description: The investigation reported is concerned with questions regarding a possible Mach number influence on skin friction reduction caused by injection. The investigation shows that data considered by Danberg (1967) for the no-blowing skin friction coefficient are in error. Accurate profiles and local skin friction coefficient values are obtained when the influence of low Reynolds number amplification in the outer region of the boundary layer is included in a calculation method.

    Description: Data are presented from the Ariel-V All-Sky Monitor which indicate a 17-day periodicity of the X-ray spectra from Cyg X-3 (3U2030+40). The data are consistent with data from the Ariel V Sky Survey Experiment. Assuming that the high intensity state of Cyg X-3 is well fitted by a structureless black body (Serlemitsos et al., 1975), and that the 17-day maxima correspond to increased electron scattering in the source, it is suggested that the 17-day effect is analogous to the 35-day variation in Her X-1. The possible explanation of this phenomenon as a result of the 17-day orbital period of the binary system containing Cyg X-3 is severely constrained by the stability of the well-known 4.8-hour variation.
