    Notizen: Atypical eukaryotic RNA polymerase activity was demonstrated in nuclei of Crypthecodinium cohnii, a eukaryote devoid of histones. Nuclei were isolated from growing cultures of this dinoflagellate and assayed for endogenous RNA polymerase (EC 2.7.7.6) activity. There was a biphasic response to Mg2+ with optima at ˜ 0.01 and 0.02 M MgCl2, but in contrast to other eukaryotic RNA polymerases, this enzyme activity was inhibited by low MnCl2 concentrations. In the presence of 0.01 M MgCl2 the optimum (NH4)2SO4 concentration was 0.025 M, a concentration at which the nuclei were lysed. Incorporation of [3H]UMP into RNA was inhibited by actinomycin D and dependent on the presence of undegraded DNA, and the reaction product was sensitive to ribonuclease and KOH digestion. Omission of one or more ribonucleoside triphosphates greatly reduced the incorporation. Only a slight enhancement of RNA polymerase activity resulted from the addition of various amounts of native and denatured calf thymus DNA. Spermine caused a marked inhibition while spermidine had little effect on RNA synthesis in the nuclei. Under the optimum conditions described in the present paper the nuclei incorporated ˜ 3 pmoles of [3H]UMP/muml; DNA at 25 C for 15 min, and ˜ 80% of this activity was inhibited by the eukaryotic RNA polymerase II inhibitor, α-amanitin (20 m̈/ml). A unique situation therefore exists in C. cohnii nuclei, in which absence of histones (a prokaryotic trait) is combined with α-amanitin-sensitive RNA polymerase activity (a eukaryotic trait).

    Notizen: : A visual selection technique can be utilized to locate potential well field sites prior to on-site testing. The technique is based upon an understanding of the regional hydrology and an appreciation for development goals such as the identification of high-yield and low-impact locations. Although the uniqueness of Pasco, Pinellas, and Hillsborough Counties in Western Florida has been incorporated into the current application, the methodology, with modifications, is transferable to other geographic regions. Overlays of each criterion used in site selection are combined to form a regional composite showing favorable site locations and further serve a useful communications role as visual aids during public presentation. Implications of direct and indirect cost savings to the public are obtained when the methodology is applied to the location of municipal well fields.

    Notizen: Abstract In this revised formulation of guard cell mechanics, the material properties of the walls are re-examined. The observed elastic anisotropy of guard cell walls can be explained by non-random orientation of the cellulose micellae in the unstrained state. This micellar network is assumed to be loosely embedded in the wall matrix causing a two phase elongation process. In the first phase, the micellar network is ‘loose’ resulting in the walls behaving as an isotropic polymer when stretched. As the volume of the cell expands beyond some threshold, the network becomes ‘tightened’ and a second phase of elongation is initiated. During this anisotropic phase of cell expansion, wall elasticity reflectes changes in the orientation of the network to reduce its load.Using the above theoretical analysis, a turgor-pressure versus lumen volume relationship is simulated for Vicia faba. The relationship between aperture and water potential for this species is also established. The simulated results agree with the experimental evidence reported for Vicia faba. The estimated shear modulus of elasticity for guard cell walls is 2 MPa (20 bars) which is well within the limits of reported values for other biological tissues.

    Notizen: Abstract. The determination of guard cell lumen volume in relation to its geometric characteristic dimensions is presented. Stomatal movements can be divided into two stages: Spannungsphase and motorphase, with a transition point between them. During the Spannungsphase movement, the lumen volume increases as a result of the change in its geometric shape. At transition, the lumen volume is approximated by a portion of a circular tube with a rounded cross-section. The volume increase during the motorphase comes from three different sources: expansion by wall stretching, increasing inner cross-section of a guard cell by wall thinning, and polar expansion. The relationship between the aperture and the lumen volume is also established. The results set forth in this geometric model are essential to studies of the pressure interaction between a guard cell and its surrounding epidermal cells.
