Notizen: Survey data collected during a grassland-recording investigation showed a strong positive correlation between nitrogen input and the output of utilized starch equivalent per acre. At the levels of output covered, sward type was not very important in relation to total yield. Herbage species were important for other reasons and the choice of seeds mixtures should be determined by factors such as earliness and lateness of growth and resistance to disease and drought. The survey showed that well-managed ley and permanent pasture are both capable of producing at a high level. A disturbing feature of the results was the low production of most spring resowings, primarily as a result of poor management. Although the records were obtained from an above-average group of farms, the results have general application. Potential responses to fertilizers may be even greater on less intensive farms, especially with regard to phosphate and potash. However, the level of production of a field will always depend finally on the level of management exercised by the farmer himself.

Notizen: SUMMARY. Temperature and current discharge regulated phytoplanktonic concentration, chlorophyll-a concentration, the light-saturated rate of photosynthesis (Pmax), and photosynthetic capacity (Pcap) in the Mississippi River at Prairie Island, Minnesota. The chlorophyll-a maximum was 48 mg m−3 in 1975, a wet year with a high current discharge, and 190 mg m−3 in 1976, a relatively dry year. The highest values of Pmax were 0.37 (mgO2 I−1h−1) in 1975 and 1.60 in 1976. Pcap varied from 3 to 21 (gO2 per g chlorophyll-a h−1) both years, and its value was highly correlated with temperature. The temperature optimum shifted from 16°C for Pcap in the spring, to greater than 28°C in the summer. Multiple regression analysis indicated a second-order relationship of Pcap in the spring to temperature. Other independent variables explained only negligible variation of Pcap.

Notizen: SYNOPSIS. Optimal assay conditions are described for 8 hydrolases of Euglena gracilis var. bacillaris, SM-L1 (streptomycinbleached) strain, 7 of which have an acid pH-optimum. Acid phosphatase, β-galactosidase, β-glucosidase, β-fucosidase, cathepsin D, RNase, DNase, and an esterase are active in cell homogenates. Amylase has very low activity, and β-glucuronidase, arylsulfatase, β, N-acetyl-glucosaminidase, α-fucosidase, and α- and β-mannosidase are inactive.Hydrolase activity increases as a culture proceeds from the midexponential to the late stationary-phase of growth, being most pronounced in the case of β-glucosidase. In cultures deprived of a utilizable carbon source, the specific activities of the hydrolases (per mg total protein or dry weight) increase. When expressed on a per cell basis, however, the activities of DNase decrease while those of β-galactosidase, cathepsin D, and RNase increase. The hydrolases appear to be involved in the adaptation of Euglena to the metabolic demands imposed by different conditions of growth.

Notizen: SYNOPSIS. Heme, intrinsically required by Trypanosomatidae, is unstable, especially in conventional alkaline (pH 7.2–8.0) media. Low solubility of heme in a pH 6.5 basal medium (developed to assay biopterin with Crithidia fasciculata) posed a problem: in media acidified during growth because of glycolysis, heme precipitated, perhaps contributed to acid-limited growth and interfered with densitometric estimation of growth. The remedy was to: replace glucose with less rapidly metabolized mannitol; distribute media in thin layers to promote oxidation of acetate, fumarate, and malate (presumably leaving an alkaline residue); and buffer heavily with histidine + Good zwitterionic buffers, and superimpcse physiological buffering by arginine + asparagine whose catabolism appeared to yield an excess of NH+4 over acid. Thereupon, Fe and Cu deficiencies sharply limited growth in the medium whose main chelators were: (a) 2,3–dihydroxybenzoic + 5-sulfosalicylic acids (which preferentially bind transitional elements at their higher valences; (b) malic and gluconic acids; and (c) histidine. With unconventionally heightened concentrations of Fe, Cu, and Mo (the latter serving as Cu buffer as well as nutrient per se), the hemin concentration could be lowered, widening the margin of safety for heme solubility. Growth then reached 1.4 × 108 cell/ml. This medium may serve to screen for ligands promoting uptake or release of Fe and Cu. The increased growth is a step towards improving the assay medium for biopterin and practical use of Crithidia to assay several B vitamins and essential amino acids for metazoa.

Notizen: SYNOPSIS. Earlier experiments suggested that the species of Haemoproteus which infects English wood-pigeons (Columba p. palumbus) and is transmitted by the hippoboscid dipteran fly Ornithomya avicularia, was unable to infect Columba livia domestica, a host of H. columbae. This has now been confirmed by the injection of sporozoites into two pairs of pigeons, one of each species; in both experiments the C. palubus developed parasitemia while the C. livia did not. It has also been shown that this species of Haemoproteus can complete sporogony in a small proportion (2/73) of Pseudolynchla canariensis, the vector of H. columbae.

Notizen: Swards based on a mixture of SI 70 tall fescue and S215 meadow fescue were very early in spring, gave high yields for conservation in mid-season and, rested from mid-August, gave good grazing in November-December. Under this treatment, the tall fescue assumed dominance, and the swards were persistent and remained productive. The digestibility of the tall fescue swards was always higher than that of swards based on cocksfoot with which they were compared. During spring and autumn the former sward was better grazed by stock, but the position was reversed in mid-summer. Cattle grazing tall-fescue-dominant swards made better liveweight gains than those grazing cocksfoot swards during November-December.

Notizen: The value of varieties of Italian, hybrid and Westerwold ryegrass for early-bite production was compared at 32 centres in different environments. The relative performance of Italian ryegrass varieties was fairly consistent from centre to centre over a period of 3 years. Danish and Leda Daehnfeldt Italians produced the most, and S22 the least, early spring production. Good early-bite and total annual yields were obtained from hybrid ryegrasses. Swards sown in the previous autumn were more winter-hardy than spring-sown swards and appeared to be as early in the following year.
