Notizen: Chick embryo fibroblasts were maintained at confluency for up to 35 days in medium containing 0.5% or 0.75% fetal bovine serum or 2.5% or 5.0% horse serum. At weekly intervals cells were subcultured and serially propagated in medium containing 10% FBS until their replicative lifespans were completed. The results showed that the replicative lifespan of embryonic chick fibroblasts was dependent on the cumulative number of population doublings undergone by the culture and was not related to the calendar time cells were in culture. Further characterization of 0.75% FBS maintained chick cells returned to 10% FBS medium showed that cells had protein contents and incorporated 3H-thymidine into DNA at a rate that resembled that of young cells, despite an advanced chronological age.

Notizen: Chick embryo fibroblasts serially propagated in media containing division ratelimiting amounts of fetal bovine serum underwent premature culture senescence as illustrated by accelerated declines in the number of cells incorporating 3H-thymidine, increased population doubling times, reduced cell densities at subcultivation, and reduced replicative life-spans compared to cells grown in medium containing non-rate-limiting amounts of serum. Low serum serially propagated “senescent” cultures returned to 10% serum containing medium had proliferative rates, incorporated 3H-thymidine, and attained saturation densities at confluency similar to younger cells. “Senescent” cells serially propagated in low serum and returned to 10% serum achieved life-spans similar to cells continuously grown in the presence of 10% serum. The results of these and other studies show that cells serially propagated in the presence of division rate-limiting amounts of fetal bovine serum, or at high inoculation densities, accumulate a substantial number of cells in the population during exponential growth conditions that are not senescent but are prevented from entering DNA synthesis becuase of mitogen limitations. Our results indicate that the amount of serum mitogen in the growth medium affects only the rate at which cells express their genetically predetermined replicative potential and not the replicative lifespan per se. These results are discussed in relation to the techniques that should be employed for studying cellular aging and the mechanism of senescent cell formation.

Beschreibung: A search is made for anisotropic X-ray bremsstrahlung photon production from relativistic electrons by studying the heliocentric angular dependence of 53 flares detected at energies above 300 keV. No evidence is found for a higher rate of detectable flares near the limb at the 80 percent confidence level. This result implies that the X-ray directivity as defined by the ratio of photon intensity at 75 deg and 0 deg of heliocentric angle is less than 1.5 above 300 keV and strongly rejects any flare model predicting X-ray production from a radial 'beam' of energetic electrons.

Beschreibung: The Gamma Ray Spectrometer on the Solar Maximum Mission satellite has observed energetic solar neutrons (greater than 50 MeV) at the earth following a solar flare that occurred on the west limb on June 21, 1980 at 01:18:20 UT. Impulsive photon emission from 10 keV to greater than 65 MeV lasting over a period of about 66 s was followed by a transient flux of 50-600 MeV neutrons incident over a 17 minute period. The peak counting rate corresponds to an average flux at the earth of (3.8 + or - 0.6) x 10 to the -2nd neutrons/sq cm s at 130 MeV. These observations indicate the emission of 3 x 10 to the 28th neutrons/sr with energies greater than 50 MeV, requiring the rapid acceleration (much less than 60 s) of protons to GeV energies during the impulsive phase of the flare.

Beschreibung: Gamma-ray continuum emission from 0.3 to 1 MeV was observed with the gamma-ray spectrometer on the Solar Maximum Mission satellite during two impulsive solar flares on 1980 March 29, from active region 2363 at 0918 UT and from active region 2357 at 0955 UT. Evidence is presented for a hardening of the spectrum during the impulsive phase of the flares. The photon intensity greater than 100 keV appears to decay at a slower rate than that at lower energies. Time-integrated photon spectra for both flares are incompatible with a single-temperature thermal-bremsstrahlung model. Upper limits for prompt and delayed gamma-ray lines are presented.

Beschreibung: Results are presented for the flare of July 1, 1980, which started at approximately 1627 UT and in which simultaneous measurements were made of X-ray, gamma-ray, and optical continuum emission for the entire duration of the flare. The X-ray and gamma-ray observations were made by the Gamma-Ray Spectrometer on the Solar Maximum Mission satellite. The optical measurements were taken at the Sacramento Peak Observatory and the Big Bear Solar Observatory (Zirin and Neidig, 1981). It is found that the major white-light emission that occurs in the late phase of the flare could not have been due to heating by electron or ion precipitation. This conclusion derives from the fact that the X-ray and gamma-ray flux peaks approximately 1 minute before the maximum of the optical continuum mission emission. It is also found that approximately 73 percent of the optical continuum emission, representing a spatially and temporally distinct bright point, follows this maximum with little or no X-ray or gamma-ray emission in the same period.
