    Notes: Summary The hormonal metabolite of vitamin D3, 1,25-dihydroxyvitamin D3 [1,25(OH)2D3], exerts its biological effects by binding to a cytosolic receptor protein. Such a protein has been demonstrated in vitamin D3 target organs including fetal rat calvariae and more recently in rat osteogenic sarcoma cells. In this study we have compared the binding of 25-hydroxyvitamin D3 [25(OH)D3] and 24,25-dihydroxyvitamin D3 [24,25(OH)2D3] to that of 1,25-(OH)2D3 in fetal rat calvariae and osteogenic sarcoma (OS) cells. Sucrose density sedimentation, DNA-cellulose chromatography, and intracellular uptake studies have been employed to evaluate these interactions. In cytosol preparations from calvariae, [3H]-1,25(OH)2D3 bound to a 3.3S macromolecule and to a much greater extent to a 5.8S macromolecule while both [3H]25(OH)D3 and [3H]24,25(OH)2D3 bound to the 5.8S macromolecule. By incubating intact calvariae and OS cells with labeled metabolites and thus establishing binding intracellularly prior to cell disruption, we have found that the 3.3S protein which has high specificity for 1,25(OH)2D3 occurs inside the cells; the 5.8S protein, however, does not occur inside the cells but is generated after cell disruption. The [3H]-1,25(OH)2D3-receptor complex adsorbed to DNA-cellulose and was eluted from this affinity resin at 0.28M KCl. In contrast, [3H]25(OH)D3 and [3H]-24,25(OH)2D3 binding activity did not adsorb to DNA-cellulose. We conclude that, in contrast to the 3.3S protein, the 5.8S macromolecule does not fulfill receptor criteria but is rather generated by the experimental manipulation of the bone cells. Our data suggest that the vitamin D3 actions on bone are mediated only via the 3.3S receptor, and hence quantitative but not qualitative differences of the effects of the various metabolites are feasible.

    Notes: Summary Bone Gla protein (BGP) was measured in the plasma by radioimmunoassay (RIA) during treatment of 59 patients with bone diseases including Paget's disease (N=9), primary hyperparathyroidism (N=25), chronic renal failure (N=20), and cancer involving bone (N=5). Plasma BGP was increased above normal in all patients. BGP decreased in the patients with Paget's disease following the acute and chronic administration of salmon calcitonin. Plasma BGP was higher in women then in men with primary hyperparathyroidism. Following parathyroidectomy, BGP decreased in both sexes but the decrease was significant in women only. Plasma BGP was increased in patients with renal osteodystrophy and did not change after hemodialysis. In the patients with bone cancer, plasma BGP decreased during treatment of the attendant hypercalcemia with salmon calcitonin. Although plasma BGP and serum alkaline phosphatase (AP) levels were generally correlated in these studies, there were examples of dissociation between the two. The measurement of plasma BGP appears to provide a specific index of bone metabolism that may in some circumstances be more sensitive than serum alkaline phosphatase measurement. However, further studies are necessary to establish the clinical value of plasma BGP measurement by RIA in the management of patients with bone diseases.

    Notes: The kinetics of the anionic polymerization of ∊-caprolactone, including the kinetics of macrocyclization and the kinetics of macrocycles propagation in THF solution, initiated with (CH3)3SiONa, were investigated. Rate constants of propagation (kp(n)) and of back-biting (kd(n)) involving monomer and cyclic oligomers (macrocycles) with n = 2, 3, … monomeric units were determined for n ≤ 7. It was found that starting from the tetramer kd(n) ∼ n-1,4±0,1 and kp(n) ∼ n0,7±0,15 (in Jacobson-Stockmayer's theory kp(n) ∼ n). This discrepancy is explained in terms of “conformational hindrance”, increasing for small cycles with the increase of the ring size until a certain ring size is reached. This hindrance decreases the availability of some of the ester groups in the macrocyclics for attack of the growing species, lowering in this way the exponent in the dependence of kp(n) on n from one to a lower value.

    Notes: A dimensionless dispersion number was developed to characterize liquid-liquid dispersions created in turbulent Couette flow with respect to their subsequent coalescence in a settling zone. For a given solvent pair and continuous phase, the dispersion number is relatively constant over a wide range of operating and dimensional parameters.     Notes: Fourier transform infrared spectroscopy (FT-IR) and torsional braid analysis (TBA) were used to study the reaction of an epoxy resin system cured with a sterically hindered amine. Isothermal torsional braid analysis showed the apparent activation energy to be approximately 42 kJ/mol. The reaction kinetics were also considered from the unreacted freshly mixed condition and a partially reacted “B-staged” condition using infrared spectroscopy, The B-staged condition is unreactive and stable at room temperature due to the quenching of the primary amine reaction by the glassy structure and the steric hindrance of the secondary amine reaction. Apparent activation energies for these two conditions were found to be 49.4 kJ/mol and 49.0 kJ/mol respectively. The storage life at room temperature of the B-staged resin system is predicted to be at least three months based on extrapolation of the experimental kinetic data.

    Notes: Carbon analysis was used as a method for measuring the quantity of a cationic vinyl benzyl silane (CVBS) adsorbed onto the surface of mica. The results obtained indicate that CVBS can be applied from aqueous solution over a wide pH range with best treatment in neutral and acidic media. The amount adsorbed depends on the concentration of the treating solution in the lower concentration range while in the higher range, the adsorption is less dependent on concentration. The relationship, silane adsorption vs. time of treatment, shows a steplike dependence. It is suggested that each step corresponds to a monolayer coverage. The data indicate that CVBS on mica is a highly ordered and closely packed multilayer phase with the silane molecules oriented normal to the surface. The multilayer adsorption of silane onto silane in this system may be compared in some respects to polymer crystal growth.
