    Notes: Conclusions Comparison of the Freundlich isotherms for adsorption and desorption of Aroclor 1254 indicate that adsorbed PCB does not desorb readily. The Freundlich exponent and K value for desorption, based on the organic carbon content of the sediment, may be of practical use in predicting PCB concentrations in the pore water of settled dredge spoil, provided that the replacement rate of pore water is slow enough to allow establishment of equilibrium conditions. It is assumed that these coefficients may apply to other sediments with different combinations of organic and inorganic components. However, further study to check this and whether the Freundlich isotherms apply at lower concentrations of PCB is required.

    Notes: Summary An acute reduction in the synaptic availability of serotonin (5HT) by p-chlorophenlalanine (PCPA) nullifies the decrease in the density of cortical beta adrenoceptors caused by desipramine (DMI) but does not appreciably alter the attenuation of the norepinephrine (NE) sensitive adenylate cyclase. The analysis of competition-binding curves of [3H]-dihydroalprenolol shows that the affinity of the agonist (−)-isoproterenol for cortical beta adrenoceptors is profoundly reduced following PCPA. This reduction in agonist affinity is enhanced by DMI. Resupplying 5HT by by-passing trptophan hydroxylase inhibition, by administering 5-hydroxytryptophan, converts a DMI non-responsive to a DMI responsive beta adrenoceptor population and shifts the markedly decreased agonist affinity towards the affinity values found in control preparations. The results demonstrate the pivotal role of 5HT in the regulation of the density and agonist affinity characteristics of cortical beta adrenoceptors and contribute to the scientific basis of the ‘serotonin-norepinephrine link hypothesis’ of affective disorders.

    Notes: Summary Δ 1-Tetrahydrocannabinol (Δ 1-THC) has been quantified directly in erythrocyte membranes from drug-treated mice using gas chromatography/mass spectrometry. Concentrations of approximately 6 ngΔ 1-THC/mg membrane protein (10−5 M) were found when effects of the drug on behavior were prevalent. At these concentrations the drug produced a decrease in membrane order as measured by ESR.

    Notes: Summary A new technique which removes all membranes, cytoskeletal elements, organelles, but preserves intact metaphase, anaphase and telophase configurations is combined with scanning electron microscopy (SEM) as an approach for direct visualization of chromosomal behavior in late mitosis. With this approach we are able to confirm the presence of a centromeric ring which stabilizes the centromeres during the cell cycle and present evidence for a lattice-like sheet of interchromatidic fibers in late mitosis.

    Notes: Abstract A comparison of chiasma distribution patterns between two chromosomal taxa, Moreton and Torresian, and their F1 hybrids demonstrates highly significant differences between all chromosomes analysed. In chromosomes 1, 2, 4, 5, 6 and 8 these differences can be directly attributed to pericentric heterozygosity in the F1 hybrid. In chromosomes 7 and 8 where there is no pericentric heterozygosity these differences may be due to heterozygosity for interstitial and terminal bands of hetero-chromatin or possibly undetected paracentric rearrangements. The F1 hybrids also have a significantly lower mean cell chiasma frequency. The Moreton and Torresian taxa differ significantly in chiasma distribution pattern in chromosomes 1, 2, 4, 5, 6 and 8 and both Moreton populations analysed have a significantly lower mean cell chiasma frequency than the Torresian population. In addition the two Moreton populations, (MMX) and (MAX), differ significantly in the chiasma distribution pattern in chromosomes 1 and 2 and the chromosomally more polymorphic population (MMX) has a significantly lower mean cell chiasma frequency. There is some evidence that the differences in both chiasma distribution and frequency between these two populations may be due to genetic differences in addition to the effects caused by chromosomal polymorphism. It has been shown that in general there is a substantial reduction in recombination in the intersitial regions of most chromosomes in the Moreton and particularly the Torresian taxon because of a proximal-distal localisation of chiasmata. In the F1 hybrid, however, nearly all recombination events are located within these interstitial regions. This provides support for the hypothesis that the frequent placement of chiasmata in regions of normally low recombination may disrupt the internal coadapted genetic environment of both chromosomal forms resulting in non-functional recombinant progeny in the next generation. The recombination data in this study also provide a basis for an empirical test of this hypothesis.

    Notes: Summary We report here the results observed when tubulin fluorescence immunohistochemistry is performed upon dissociated cultures of nervous tissue, principally of chick embryo spinal cord. When fixation includes nonpolar solvents or detergents, a uniform fluorescence is seen in neuron perikarya (with the exception of their nuclei), and the processes to which they give rise. Fixation with formaldehyde or glutaraldehyde alone, however, results in a discontinuous staining of neurites, and a less regular staining of their perikarya. The former pattern of response can be elicited if aldehyde fixation is followed by exposure to non-polar solvents. Such results are obtained both in thinly spread regions of the cultures, where neurons and their processes can easily be seen, and in the cell aggregates that also characterise them. Possible interpretations of these results are discussed.

    Notes: Summary This paper contains a comparison between four experimental methods for the detection of trace metals in biological tissues, e.g. autoradiography, dithizone-, Timmstaining and microPIXE (Particle Induced X-ray Emission). —As research object the hippocampus of the rat was chosen and mainly the Zn-distribution in the fascia dentata has been examined. The comparison was judged on four criteria: spatial resolution, specificity, quantification and influence of the protein binding of the metal on the outcome of each method. Some additional Timm and microPIXE experiments have been performed to compare the metal concentrations for both normal and Zn-deficient rats. The Zn/Cu ratio for both kinds of rats were calculated and some striking differences in the concentrations were obtained. The Zn/Cu ratio for a normal rat was found to be about 40% higher than for a deficient one.
