    Notes: The free radical cation of 1,4-diaminobenzene (p-phenylenediamine, PPD+.) has been generated in aqueous solution, wherein it is short-lived, by oxidation of the parent compound with bromine. The same PPD+. radical has also been produced by UV photolysis of the parent at 77 K in an ethanolic glassy matrix. Resonance Raman spectra of the radical and its parent are reported and analysed in detail with the aid of normal coordinate and molecular orbital calculations. Evidence is presented for (PPD+.)2 dimer formation in aqueous solution at room temperature.

    Notes: Doubly charged ion mass spectra were obtained for 46 low molecular weight oxygen containing compounds. A double focusing Hitachi RMU-7L mass spectrometer, operated at 3.2 kV accelerating voltage, was used to measure spectra for aliphatic alcohol, ketone, ether, aldehyde, ester and acid molecules, as well as several aromatic oxygen containing compounds. In general, the spectra were dominated by fragment ions which resulted from extensive H loss and C—C bond rupture as well as O elimination from the doubly charged molecular ions. Total product ion intensities from doubly charged ion spectra of aliphatic oxygen containing compounds were approximately 1% of the corresponding total ion intensity in the benzene doubly charged ion spectrum. Appearance energies for forming prominent doubly charged molecular and fragment ions were determined. Measured values ranged from 26 to 45 eV. A geometry optimized quantum mechanical SCF treatment was used to compute the energies, charge densities and structures for several of these oxygen containing doubly charged ions.

    Notes: The polyimide resin LARC-160 was prepared from diethyl-3,3′,4,4′-benzophenone tetracarboxylate (BTDE), ethyl-5-norbornene-2,3-dicarboxylate (NE), and Jeffamine AP-22. The imidization reactions of NE and BTDE were studied by HPLC, 13C-NMR, and IR. NE imidizes slowly at 12°C; BTDE imidizes when the resin is heated above 100°C. Both imidization reactions proceed directly to the imide. Neither amic acid is present in significant quantities at any stage of the imidization reactions. The monomer mixture has been stored at 12°C for periods up to 14 months. The effects of resin aging at this temperature on the chemical composition of the resin monomer mixture and the imidized polymer formed on curing were investigated. Aging the resin monomer mixture has the effect of partially advancing the imidization reaction. Aging also results in the formation of slightly higher-molecular-weight polyimide chains after curing of the resin at 140 and 180°C. Bisnadimide (BNI) is observed as a major reaction product, regardless of resin age.

    Notes: Poly(vinyl chloride) film shows substantial changes in its photoacoustic absorption spectra (PAS) on either irradiation or heating in the presence of O2 or N2. These spectral changes are due to the conjugation structures and carbonyl (group formation), formed during photo and thermal degradation. In this paper interpretations of the PAS and of the decomposition mechanism of PVC are presented.

    Notes: Matrix vesicles, extracellular microstructures known to eb involved in endochondral calcification, are rich in alkaline phosphatase and have been shown to contain actin. The mechanism of matrix vesicle formation in chondrocytes in not well understood. Chondrocytes from the epiphyseal growth plate, when grown in primary culture, elaborate alkaline phosphatase-rich vesciles. We examined the distribution of the cytoskeletal proteins actin, myosin, tubulin, and vinculin at various time-points during culture using indirect immunofluorescent labeling. Concomitantly, the production of alkaline phosphatase-containing matrix vesicles was also followed. Cell morphology changed noticeably at two distinct stages during the 22-day culture period: Immediately after release from the growth plate the cells were founded, but after 4 days of cultre they began to spread out and acquire irregular shapes with distinct filopodia. By 13 datsm as tge cekks attaubed confluency, they reacquired a rounded, polygonal appearance. At all time-point, tubulin was seen as a dense network of microtubules radiating from the perinuclear region throughout the cytoplasm toward the cell periphery. Initially actin was seen in filamentous from, but displayed a punctate distribution focused at contact points during the cell-spreading stage of culture. After confluency, actin was concentrated at cell-cell junctions. Initially, vinculin was diffusely distributed, but became focused in multiple adhesion plaques and at the termini of filpodia during the cell-spreading stage of culture. Following confluency vinculin became concentrated at cell-cell junctions. Myosin was observed at all time-points in small, intensely localized focal points in the cytoplasmic region of the cells and was consistently absent from the nuclear and peripheral regions. The amount of myosin in the cells increased steadily with time in culture. Elaboration of alkaline phosphatase-rich vesicles, which corresponded closely with the rounded morphology of early and late stages of culture, may be correlated with contact inhibition.

    Notes: Five commercial varieties of uncoated fused silica capillary tubing used in high resolution gas chromatography were subjected to cobalt-60 gamma radiation. The advantages and superior performance of fused silica open tubular columns coated with OV-1 and subsequently immobilized or crosslinked in situ by radiation is reported in a previous paper in this Journal; however, in this investigation a loss in flexibility of irradiated capillaries was also noted. Quantitative measurements on flexibility were performed on specimens of irradiated fused silica capillaries, indicating that changes do indeed occur upon irradiation. Scanning electron micrographs also illustrate slight deterioration of the outer protective polyimide coating. Most surprisingly, it was also established that a variation does exist in the flexibility of the commercially available raw material studied.

    Notes: Avian mycloblastosis virus (AMV) is a replication-defective acute leukemia virus, requiring a helper virus to provide the viral proteins essential for synthesis of new infectious virus. The genome of the AMV has undergone a sequence substitution in which a portion of the region normally coding for the “env” protein has been replaced by chicken cellular sequences. These latter sequences are essential for the transforming activity of the virus. We have determined the complete nucleotide sequence of this region. Examination of the AMV oncogenic sequence revealed an open reading frame starting with the initiation codon ATG within the acquired cellular sequences and terminating with the triplet TAG at a point 33 nucleotides into helper viral sequences to the right of helper-viral-cellular junction. The stretch of 795 nucleotides would code for a protein of 265 amino acids with a molecular weight of 30,000 daltons. The eleven amino acids at the carboxy terminus of such a protein would be derived from the env gene of helper virus.

    Notes: The conformational properties of a series of gastrin-related peptides in aqueous solution and in 2,2,2-trifluoroethanol (TFE) have been investigated by CD measurements. In aqueous solution the peptides Leu32-HG-34 (human big gastrin), Nle15-HG-17 (human little gastrin), and Nle11-HG-13 assume a random-coil structure in the pH range 3-7. In TFE the three hormones fold into partially ordered structures, consisting of mixtures of α-helix, β-form and random coil. Comparison with the CD properties of the shorter gastrin peptides HG-4 (tetragastrin), Nα-Boc-HG-5 (pentagastrin), and HG-7 (heptagastrin) indicates that the biologically important C-terminal sequence Trp-Met-Asp-Phe-NH2 in TFE does not maintain the same geometry upon elongation of the chain at the N-terminus from 4 to 34 residues. Thus, the various conformations in solution of the gastrin peptides examined do not provide a structural explanation for their very similar biological activity. Therefore, we hypothesize that the C-terminal tetrapeptide amide folds into an “active” structure only upon interaction with the receptor.

    Notes: The water absorbed by poly(vinyl acetate), PVAc, at 23°C was found in two states. The first, which can account for up to 4 weight percent, was bound to the polymer. The second was in a freezable or clustered form. The latter type of water had no effect on PVAc's glass temperature, whereas, the former kind plasticized Tg. In annealing studies, the enthalpic and dielectric response of PVAc when held at a fixed temperature increment, ΔT, below Tg, was observed to be independent of the amount of bound water. The time dependence of the shift in the dielectric relaxation spectrum and the recovery of the enthalpy towards its equilibrium value as PVAc approached its equilibrium glassy state from a lower temperature as compared to a higher temperature was initially slower. This delayed response to expansion was of the order of the polymer's average relaxation time at the lower temperature. A model was proposed to explain this asymmetric behavior based upon changes in the polymer's free volume as well as its occupied volume.
