    Notes: A direct method was developed for the determination of seafood amines. The method utilizes the principle of ion-moderated partition high performance liquid chromatography (HPLC).The method involves the extraction of muscle tissue in perchloric acid (PCA) and the subsequent chromatography of the neutralized PCA extract using dilute pH 11.0 sodium hydroxide as elution solvent, the entire procedure requiring approximately 30 min to complete. The biogenic amines (histamine, putrescine and cadaverine) were separated and the method compared with an existing HPLC procedure. Trimethylamine and dimethylamine were also determined. by this method. Results compared favorably with data obtained by existing calorimetric procedures used for the determination of these compounds.

    Notes: The MHC restriction criteria for T cells activating macrophages in vivo and mediating antimicrobiol resistance to Listeria monocytogenes were determined. Antimicribiol resistance could be transferred by T cell in order of decreasing efficiency from syngeneic. RT1A. compatible, RTI.B compatible and RT1 incompatible donors. Alloreactive T cells responding to either A locus or B locus encoded antigens in a graft-versus-host reaction were also able to activate macrophages. Approximately five times as many MLC-reactive precursors responded to B locus alloantigens as to A locus alloantigens, but A-resticted Listeria-specific T cells wre considerably more numerous (or more efficient) in Listeriainfected hosts than were B-restricted, Listeria-specific T cells. This was unexpected, since A-restricted, Listeria-specific T cells failed to transfer delayed hypersensitivity (DTH) to soluble bacterial antigens.

    Notes: Among 290 F2 progeny of an r10 x ACP cross were two recombinants which allowed the loci for glyoxalase-1 and neuraminidase-1 to be mapped relative to the RT1.A and dw-3 loci in the major histocompatibility complex (MHC) of the rat. In 673 progeny of the same cross there was a recombinant between ft and dw-3, and in 403 progeny of the backcross BY1 x (BY1 x BDIX)F1 there was another recombinant between ft and dw-3. These data, combined with those from previous studies, provide the information for constructing a detailed map of the rat major histocompatibility complex: the gene order and size in the rat are very similar to those in the mouse and different from those in man and in the other species that have been studied. Comparison of the structures of the MHC in the various species leads to a hypothesis about the evolution of the MHC which involves sequential duplications of the genes coding for class I and class II loci and an inversion in the prototypic muridae which placed the class II loci between the class I loci.

    Notes: Polymorphism of serum acid phosphatase was detected by electrofocusing in acrylamide gel. The locus has two alleles Apsa and Apsb that segregated in the backcross progeny (A x B) Fl x B in a 1:1 ratio. The phenotype APS-A is dominant in the F1 generation. The gene for serum acid phosphatase segregated independently of the major histocompatibility complex and of the sex of the rats.

    Notes: Haemagglutination inhibition studies using isolated membrane components from rat red blood cells showed that the activity of the major antigenic system RT2 was contained exclusively in the glycoprotein fraction and confirmed immunochemically the existence of two alleles, RT2a and RT2b. The SDS-PAGE analysis of the membrane showed two interconvertible glycoproteins of apparent molecular weight 72,000 and 36,000, and the higher molecular weight component appeared to be a dimer of the lower molecular weight one. Analysis of the various membrane preparations after 125I-labelling showed that the glycoproteins existed in at least two molecular species on the surface of the red cells and that one species was more readily radio-iodinated and dimerized than the other. The RT2 antigenic specificities were preferentially associated with this component.

    Notes: Morphological studies of metaphase chromosomes were done with rats derived from the BIL/1 strain, which has genes affecting growth and reproduction linked to the major histocompatibility complex by conventional Giemsa-trypsin staining and the results were compared to rat strains not carrying these defects. A subterminal-terminal centromeric polymorphism was detected in chromosome 3 [del 3 (pter → cent)] among the strains studied. Comparison of the G-banded karotypes of the rats carrying the defects with the karotypes of the otsher strains did not reveal any gross chromosomal abnormalities.

    Notes: Five monoclonal antibodies reacting with class I MHC antigens were produced by fusing lymphocytes from WF (RT1u) rats immunized against DA (RT1a) rats with P3-X63-Ag8-653 myeloma cells. Sequential immunoprecipitation studies with the mAb and the WF anti-DA alloantiserum demonstrated the presence of four different class I molecules: all four molecules were reactive with the alloantiserum; three of them contained the determinant for mAb 155; two of the latter three molecules shared the determinants for mAb 3, 56 and 60, and one of these two molecules also contained the determinant for mAb 118. The four molecules could be isolated from the antigen preparation by sequential immunodepletion first with 118, next with 3, then with 155 and finally with the alloantiserum or by sequential absorption with affinity columns of Sepharose 4B coupled to the antibodies. The three antigens which were sequentially isolated with the mAb 118, 3, and 155, respectively, were analysed by SDS-PAGE after digestion with Staphylococcus aureus V8 protease, and they showed differences in peptide patterns. The relative amounts of the antigens expressed on red blood cells and on lymphocytes were different based on the results of sequential isolation and indirect cellular radioimmunoassay: the antigen which reacted with both mAb 3 (and 56 or 60) and 155 was the major class I antigen on red blood cells, and the antigen which reacted with mAb 118, 3 (and 56 or 60) and 155 was the major class I antigen on lymphocytes.

    Notes: The genetic basis of allogeneic restriction in the transfer of delayed type hypersensitivity (DTH) to soluble Listeria monocytogenes antigens (LMA) in rats was analysed using a set of congenic strains. The DTH parameter assayed was the localization of radiolabelled donor lymphoblasts in subcutaneous antigen stimulation sites in the ear. Sharing of the RT1.B region between the donor and host was essential for the transfer of DTH to LMA, which suggests that the RT1.B region codes for restriction elements controlling antigen recognition by TDTH cells. The recombinant haplotype RT1r1 was exceptional in that transfer of DTH to A-region matched recipients was, at least in part, possible. Measurement of donor lymphoblast localization in DTH sites afforded an opportunity to quantify allogeneic effects influencing localization of sensitized donor cells in DTH sites borne by alien recipients. Both RT1.B and RT1.A region differences could induce allogeneic effects, but they were an order of magnitude lower than those based on restricted recognition by donor T cells.

    Notes: The classification of the RT1.A phenotypes of a large number of inbred and congenic strains of rats, including those prototypic for all of the known RT1.A phenotypes, was done by examining the reactivity of a variety of unabsorbed antisera with cells of the different strains. The data were analysed by the techniques of numerical taxonomy without reference to the previous classification by the classical serological methods. The most effective way of analysing the data was the use of the Jaccard correlation coefficient and the average linkage method of statistical analysis. Four conclusions were drawn from this study. First, the RT2 antigens must be matched in the donor and recipient pair used to raise the anti-RT1 (MHC) antiserum or spurious typing results can occur due to the presence of anti-RT2 antibodies. Second, using anti-RT1 antisera raised in RT2 compatible pairs of rats, the classification of inbred and congenic strains into the RT1.A phenotypes on the basis of classical serological studies was confirmed. Third, two groups of closely related RT1.Ac phenytopes were found (a, f, d and b, m). Fourth, the AUG and PVG strains, both of which carry the the RT1.Ac phenotype, are quite distantly related on the basis of this taxonomic analysis and the RTl.Ag and RT1.An phenotypes are the most distantly related compared to all of the other rat strains and to each other. These data provide further evidence that the RT1 polymorphism of the rat is very limited, especially in respect of man and mouse.
