    Notes: Abstract— The fatigue crack closure response was investigated for a surface crack in BS4360 50B structural steel, subjected to (1) constant amplitude loading and (2) constant amplitude loading interrupted by a single peak overload. A variety of compliance techniques was employed to determine closure behaviour. The crack mouth gauge measured the bulk, plane strain closure load, while the near tip strain gauge indicated the surface, plane stress closure response. For constant amplitude loading it was found that the surface regions of a surface crack are closed for a greater portion of the load cycle than the maximum depth point. A single peak overload caused different closure and growth rate transients at the surface of the thumbnail crack and at the maximum depth point. For growth rates above 10-6 mm/cycle, such behaviour agrees with the response of a through crack when subjected to constant amplitude loading, and a single peak overload.

    Notes: Abstract— Accurate knowledge of defect sizes and crack shape development from initiating defects is essential for a realistic application of fracture mechanic? to the fatigue life prediction of welded joints. Defects at the toes of high quality longitudinal non-load-carrying fillet welded joints have been examined using two methods, and compared with those observed in plain unwelded plate. The defects observed at the weld toe were, on average, four times deeper than the largest observed in the unwelded plate but had similar root radii. For this particular geometry the defects could be considered continuous at the expected initiation site. A simple estimate of the stress concentrating effect of a single composite defect, coupled with experimental evidence, led to the conclusion that the proportion of total life spent in initiation is negligible for all stress levels which cause fatigue failure. Subsequent growth and crack shape development was observed to depend on the initial defect distribution and the complex stress field at the weld toe.

    Notes: MINDO/3 calculations have been carried out for a series of branched chain alkanes in order to assess effects of branching on calculated geometries and heats of formation (ΔHf). With vicinal branching, MINDO/3 calculates the central C—C bond to be too long. Bond angles are also found to be distorted. Errors in calculated heats of formation are large when geminal branching is present and significant with vicinal branching. Branching error corrections for ΔHf have been derived and applied to a separate series of branched acyclic and cyclic compounds. For the test sample, application of the branching error corrections gave calculated structures of acyclic branched hydrocarbons with heats of formation having an average absolute error of 1.3 kcal/mole rather than 17.3 kcal/mole before correction. Cyclic branched hydrocarbons are shown to be less well corrected. Calculations of heats of reaction have also been carried out for some isomerization and cyclization reactions using the MINDO/3 and MNDO methods. It is clear from the comparisons that MNDO calculations give less severe errors for highly branched compounds but the errors are still substantial. For prediction of heats of reaction, the error-corrected calculations are shown to be superior to the “raw” calculations obtained by MINDO/3 or MNDO.

    Notes: . Sarcocystis falcatula Stiles, 1893 is re-described. Intermediate hosts of the parasite which was earlier described as Sarcocystis debonei Vogelsang, 1929 are species of passeriform, psittaciform, and columbiform birds. In these birds, muscle zoites are 6.88 × 2.19 (4.8-8.4 × 1.2-3.6) μm and are enclosed in a cyst wall with regular protrusions, 1-5 μm long. The convoluted primary wall has multiple thin areas in the osmiophilic layer. Microtubules originate in the ground substance and extend to the tips of the protrusions. The only known definitive host is the opossum, Didelphis virginiana; rats, cats, a dog, and a ferret could not be infected from muscle cysts. Sporocysts from opossums infected from five different infected avian sources measure 11.2 × 7.4 (9.6−12.0 × 6.0-8.4)μm.

Opossums (Didelphis marsupialis), act as intermediate hosts for Besnoitia darlingi and could be infected orally with sporozoites (oocysts) and bradyzoites (tissue cysts), or intraperitoneally (i.p.) with tachyzoites. Infections could presumably be transmitted through cannibalism. Cats (Felis catus), the definitive host, could be infected only with bradyzoites but not sporozoites. Oocysts shed by cats measure about 12 × 12 μm, resemble similarly sized oocysts of Toxoplasma gondii and Hammondia hammondi, and must be differentiated by the appearance of tissue cysts after experimental infection of intermediate hosts. Cats did not form tissue cysts of B. darlingi. Tachyzoites from the related B. jellisoni could be used in the Sabin-Feldman dye test to determine the development of antibody to B. darlingi in opossums after infection.

Various ions and treatments known to alter the availability of free Ca2+ were examined with respect to their effects on the cytopharyngeal pouch, a large prey receptacle found in the potentially carnivorous macrostomal form of Tetrahymena vorax. Addition of Ca2+, Ba2+, or Sr2+ induced the pouch to separate from the region of the cytostome, forming a large empty vacuole. Na+, alone, had no effect, but lowered the concentration of Ca2+ required to produce maximum vacuolar formation in populations of cells. Vacuolar induction was also initiated by the cation ionophore A23187 or by subjecting macrostomal cells to an electric current. In the presence of divalent cation chelators EDTA and EGTA, the cytopharyngeal pouch collapsed and was resorbed. Taken together, these results suggest that Ca2+ plays an important role during phagocytosis in this cell type.

    Notes: Members of the family Sarcocystidae, as defined by Frenkel, have had a complicated history, principally due to the existence of both coccidial and cystic stages. This formerly clandestine relationship resulted in dual or partial designations of nomenclature for individual species. The problem was further compounded by the obligatory heteroxeny of several of the genera, making it impossible to transmit the parasites from one individual to another of a single host. As a result, oocysts similar in appearance, though from hosts separated taxonomically up to the familial level, were sometimes considered to be identical. Discoveries within the last decade have generated much interest and some understanding. Current studies of these and other coccidia should emphasize complete cyclical transmissions with cognizance of potential heteroxeny with the production of tissue cysts in intermediate hosts.

    Notes: The surface proteins and glycoproteins on red cells from normal and Babesia bovis-infected calf blood have been compared. Several radiolabeling probes were used to label specifically external membrane molecules which were then separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and identified by autoradiography or fluorography. No differences were observed among the Coomassie Blue-stained membrane proteins of erythrocytes from individual uninfected calves. Comparison of red cells from these animals also indicated no qualitative differences in the surface proteins with accessible tyrosyl residues labeled by lactoperoxidase-catalyzed radioiodnation, although some quantitative variation in the uptake of radioactivity into particular proteins was observed. the major radioiodinated bands on normal bovine erythrocytes had Mr of 165, 130, 90, and 45 kiloDaltons. However, labeling of surface glycoproteins by the periodate/[3H]NaBH4 and galactose oxidase (± neuraminidase)/[3H]NaBH4 methods showed significant differences in the surface proteins of red cells from individual uninfected calves. of 14 animals tested, 5 had major labeled glycoproteins of unique Mr. No changes were observed in radioiodinated surface proteins of total red cell samples from infected calves with 0.5-6% parasitemia. Radioiodination of concentrated infected red cells from the same samples (concentrated by selective hypotonic lysis of uninfected erythrocytes in KC1) resulted in the labeling of 3 new surface proteins, with Mr of 118, 115, and 60 kiloDaltons. the same new 125I-labeled bands were identified on infected cells from 3 avirulent strains of B. bovis used in vaccine production. Furthermore, in concentrated infected cells there was very poor radiolabeling of major bands strongly labeled on uninfected cells (Mr 165, 130, and 90 kiloDaltons), suggesting parasite-induced loss of these proteins. Although there were some differences in 3H-labeled surface glycoproteins of red cells from normal and. B. bovis -infected blood, they were restricted to minor labeled bands and were not seen consistently. the labeled surface glycoproteins of concentrated infected cells were very similar to those of the uninfected red blood cells from infected blood.

    Notes: The potential utility of protein arrays visualized by SDS-PAGE in taxonomic studies of protozoa has been investigated within the genus Tetrahymena. Matching coefficients have been obtained from one-dimensional separations of cytoskeletal proteins in a study involving 40 strains of Tetrahymena. In separate studies, the method was estimated to be accurate to within 10%, and the limit of intraspecific variation within the genus was estimated at ≅ 30%. Accordingly, it is suggested that strains showing matching coefficients 0.6 in these preparations probably represent different species. Further tests using strains identified to species by breeding criteria have shown that the 0.6 rule is asymmetric, i.e. although matching coefficients lower than this indicate separate species, values higher than this do not prove identity. Protein comparisons included in the present study indicate that strain Tur, previously identified as a strain of T. vorax, is distinct from all other macrostome-forming species. It is here designated Tetrahymena leucophrys n. sp.
