    Notizen: Denatured histones H1 and H5 can be readily refolded on salt addition. Their digestion by trypsin leads to limit peptides of about 80 residues having the same nmr and CD spectra as those of the intact parent histones. Scanning microcalorimetry shows that (1) the folded structures of H1 and H5 are located entirely in their limit peptides; (2) both have values of the specific denaturation enthalpy typical for small globular proteins; and that (3) both exhibit a classic “2-state” transition (ΔHdcal = ΔHdvan't Hoff). The heat-denaturation profiles of H5 measured using intrinsic and extrinsic Cotton effect and side-chain nmr peaks do not coincide at all. Only the intrinsic Cotton effects give a Tm and ΔHdvan't Hoff close to that from microcalorimetry. We conclude that these proteins exhibit large-scale side-chain motions that precede the macroscopic cooperative transition.

    Notizen: In many commercial finite element method systems, the formulation of constraint equations from rigid elements is based entirely on rigid body mechanics. However, when initial (thermal) strains exist, a more general approach is required for rigidizing the element. This new approach also opens up a more versatile generation scheme of constraint equations for rigid or partially rigid elements.

    Notizen: Wild-collected adults of Bombina orientalis are bright green dorsally and red to red-orange ventrally. As a prelude to an analysis of the differentiation of pigment cells in developing B. orientalis, we describe structural and chemical aspects of the fully differentiated pigment pattern of the “normal” adult.Structurally, differences between dorsal green and ventral red skin are summarized as follows: (1) Dorsal green skin contains a “typical” dermal chromatophore unit comprised of melanophores, iridophores, and xanthophores. Red skin contains predominantly carotenoid-containing xanthophores (erythrophores), and skin from black spot areas contains only melanophores. (2) In ventral red skin, there is also a thin layer of deep-lying iridophores that presumably are not involved in the observed color pattern. (3) Xanthophores of red and green skin are morphologically distinguishable from each other. Dorsal skin xanthophores contain both pterinosomes and carotenoid vesicles; ventral skin xanthophores contain only carotenoid vesicles. Carotenoid vesicles in dorsal xanthophores are much larger but less electron dense than comparable structures in ventral xanthophores.The presence of carotenes in ventral skin accounts for the bright red-orange color of the belly of this frog. Similar pigments are also present in green skin, but in smaller quantities and in conjunction with both colored (yellow) and colorless pteridines. From spectral data obtained for xanthophore pigments and structural data obtained from the size and arrangement of reflecting platelets in the iridophore layer, we attempt to explain the phenomenon of observed green color in B. orientalis.

    Notizen: A new method has been developed for the measurement of overall volumetric mass transfer coefficient (KLa) in gas-liquid-solid systems. This method is based on the examination of gas phase dynamics in a three-phase contactor and consists of measuring continuously the response of the outlet gas composition to a step input change of CO2 in the inlet gas stream. The advantages and limitations of the new method are presented and its sensitivity is discussed on the basis of model predictions. Preliminary results on the implementation of the CO2 method are also reported. Experimental data obtained in a nonviscous electrolyte solution show that the proposed method compares favorably with the conventional dissolved oxygen technique, provided that a correction is made to take account of the difference in diffusivity of oxygen and carbon dioxide.

    Notizen: The disruption of Candida utilis cells in suspensions subjected to different types of stress was investigated. Stresses caused by impingement of a high velocity jet of suspended cells against a stationary surface were found to be significantly more effective for disruption than either shear or normal stresses. The fraction of cells disrupted by impingement is a first order function of the number of passes through the disruptor and, over a prescribed range of operating pressures, is a power function of pressure. These results indicate that impingement is the predominant mechanism causing cells disruption in high pressure flow devices such as Manton-Gaulin homogenizers. The impingement results suggest that cells grown in cyclic batch culture are easier to disrupt than cells grown at a lower specific growth rate in continuous culture. In addition to determining the fraction of cells disrupted, the release of invertase activity was determined for the impingement experiments. The fraction of total invertase activity released was found to be somewhat greater than the fraction of cells disrupted.

    Notizen: The kinetics of cellular reproduction and the rate and extent of synthesis of extracellular polymeric substances (EPS) were investigated for P. aeruginosa growing in glucose-limited chemostats. μmax and Ks estimates of 0.4 h-1 and 2 mg glucose C/L, respectively, at 25°C were obtained for this bacterium. The extent of EPS formation was inversely related to the growth rate of P. aeruginosa. The rate of EPS formation had both growth- and non-growth-associated components. The growth-associated polymer formation rate coefficient (k) was 0.3 mg polymer C/mg cellular C and the non-growth-associated polymer formation rate coefficient (k′) was 0.04 mg polymer C/mg cellular C/h. The values for k and k′ must be regarded as provisional since the product formation data were quite variable at low dilution rates. Estimates of the cellular (Yx/s) and polymer (Yp/s) yield coefficients were 0.3 mg cellular C/mg glucose C and 0.6 mg polymer C/mg glucose C, respectively. Most of the non-growth-associated consumption of glucose detected was due to exopolymer formation.

    Notizen: Aerobic glucose metabolism by Pseudomonas aeruginosa in steady-state biofilms at various substrate loading rates and reactor dilution rates was investigated. Variables monitored were substrate (glucose), biofilm cellular density, biofilm extracellular polymeric substance (EPS) density, and suspended cellular and EPS concentrations. A mathematical model developed to describe the system was compared to experimental data. Intrinsic yield and rate coefficients included in the model were obtained from suspended continuous culture studies of glucose metabolism by P. aeruginosa. Experimental data compared well with the mathematical model, suggesting that P. aeruginosa does not behave differently in steady-state biofilm cultures, where diffusional resistance is negligible, than in suspended cultures. This implies that kinetic and stoichiometric coefficients for P. aeruginosa derived in suspended continuous culture can be used to describe steady-state biofilm processes.
