    Notes: A new interferometer has been designed in order to perform measurements of refractive indices and refractive index increments on liquid systems under elevated pressure at different temperatures. The method of measurement, the calibration of the instrument and the treatment of data are described. With a series of test measurements on trans-decalin it is shown that with this instrument reliable results are obtained.

    Notes: Light scattering, viscosity and GPC measurements were performed on a homologous series of linear polyethylenes with viscosity average molar masses in decahydronaphthalene at 135°C between 1,0 · 105 and 2,7 · 106 g · mol-1 and relatively broad molar mass distributions. 1,2,4-Trichlorobenzene, α-chloronaphthalene, decahydronaphthalene (decalin) and decane were used as solvents. In the case of linear polyethylene in decahydronaphthalene and decane relationships between intrinsic viscosity and weight average molar mass were established. The values of the weight average molar masses obtained by light scattering are considerably higher than those obtained by viscosity and GPC measurements. These discrepancies are explained by molecular aggregation of the polyethylene macromolecules.

    Notes: Refractive indices and refractive index increments of the system polystyrene/trans-decalin were measured under pressures up to 500 bar and at temperatures between 20 and 60°C. The measurements were performed with a new, recently designed interferometer. The results demonstrate a sufficient agreement of the refractive index values measured at different temperatures and pressures with the equations of Eykman and Gladstone-Dale whereas the Lorenz-Lorentz equation indicates larger deviations. The experimental data of the refractive index increment follow none of the refractive index mixture rules. It is therefore recommended to determine refractive index increments at different pressures experimentally.

    Notes: Scattering functions for linear polyethylenes were calculated, using different assumptions, and compared with experimentally determined scattering functions of the system polyethylene/decane. It is pointed out, that the polyethylene macromolecules in a good solvent can be treated as statistical coils with a non-uniform expansion and a constant average expansion exponent ε and not as statistical coils with uniform expansion. Comparison of the experimentally determined and the calculated scattering functions yields a molar mass distribution of polyethylene which is in agreement with the results from GPC-measurements within the limits of experimental error.

    Notes: Films of poly(methyl methacrylate) (PMMA), polystyrene, and a styrene/acrylic terpolymer have been cast from solutions of varying thermodynamic quality and the film properties studied by inverse gas chromatography and by critical surface tension measurements. Surface properties of the non-polar polystyrene were independent of solvent medium, but significant variations in these properties were observed in the case of PMMA and the terpolymer. Solvent balance also appeared to affect the bulk properties of the latter films, as judged by the penetration rates of interacting liquids. The observations indicate the feasibility of controlling film properties of the solid by the appropriate selection of solution media; a time-dependent variation in solid properties is to be expected, however, as the film structure attains an equilibrium state.

    Notes: The progesterone 11α-hydroxylase of Rhizopus nigricans ATCC 6227b is an inducible enzyme system that is primarily induced by its substrate progesterone. Maximum induction was found at a progesterone concentration of 0.5 g/liter or above. Oxygen is the other substrate for the hydroxylation and this was found to have a major effect on the amounts of hydroxylase synthesized. Optimum induction of the hydroxylase in a fermentation with a 3.1 m/sec impeller tip speed was found to occur at a dissolved oxygen tension (DOT) of 10% of air saturation. The agitation rate also effects the amount of hydroxylase synthesized with an apparent maximum at 3.1 m/sec impeller tip speed. The DOT for a maximum hydroxylation rate was much higher than for enzyme synthesis so that it was preferable to increase the DOT after induction was completed.

    Notes: The filamentous fungus, Rhizopus nigricans, was immobilized in polyacrylamide, alginate, and agar gels and its ability to 11α-hydroxylate progesterone was examined. No activity was detected using polyacrylamide gel but both agar and alginate gels have proved capable of hydroxylation. Agar gels displayed faster rates and higher yields. It was possible to induce hydroxylase synthesis within agar and alginate gels, and microscopical examination provided evidence for hyphal growth within these gels. The concept of increased biomass was used to explain the observed increase in the rates of hydroxylase activity of the immobilized cells. Conversely, hyphal overcrowding was postulated for the rapid inactivation observed under some operating conditions.

    Notes: Nickel-NiO-BSA-chymotrypsin has been used to hydrolyze casein in both batch and continuous stirred tank reactors. The kinetics of hydrolysis in both types of reactor were considered. Some operational problems encountered using fine powder catalysts in batch stirred tank reactors are discussed. High gradient magnetic separation was found to be a powerful catalysts retention method for magnetic support particles, particularly when using a ferromagnetic collection matrix. Nickel particles of diameter 3-7 μm were easily separated from water at a processing velocity of 39 mm/sec. Nickel powder and precipitated Fe3O4 were also separated satisfactorily using a rotating drum magnetic separator.
