    Notes: SYNOPSIS. Novel serotypes appeared among the F2 progeny of a single cross involving alleles at the H locus. Two of the clones manifesting aberrant types have been subjected to breeding analysis. The aberrant phenotypes disappear at conjugation and have not been recovered in either F1 or F2. Comparable types have not been found again in large scale crosses involving a variety of different H alleles. The basis for the unusual types remains obscure.

    Notes: SYNOPSIS. Strains of 3 unnamed mating groups of the Tetrahymena pyriformis complex have been subjected to starch gel electrophoresis followed by staining the gels for the enzymes isocitrate dehydrogenase (NADP), tyrosine aminotransferase, and tetrazolium oxidase (superoxide dismutase). With respect to the electrophoretic mobilities of these enzyme systems, the mating groups referred to here as 5, 13 and 14 are very similar to Tetrahymena americanis (syngen 2), the most common North American species of the complex.Cultures in our collection labeled Tetrahymena cosmopolitans (formerly syngen 4) are either amicronucleate, with unique isozyme patterns, or micronucleate cells which mate with and have isozyme patterns similar to Tetrahymena canadensis (syngen 7). Immature progeny have been derived from crosses between the latter strains and T. canadensis recently collected in Colorado. The amicronucleate strains are now placed in the Tetrahymena sp. category, and we conclude that strains identifiable as T. cosmopolitanis are no longer available.The reliability of isozymes as characters in ciliate taxonomy was evaluated by comparing the present results for 3 enzymes in 15 groups of strains (syngens and phenosets) that had been compared in an earlier study. These enzyme systems gave correlation coefficients (r) of 0.75 or higher in the separate studies, and can be considered useful diagnostic traits. Other enzymes that were present at threshold levels of detectability or varied highly in concentration from species to species are too unreliable to be of diagnostic value. Some of the strains in the complex are so evolutionarily divergent at the molecular level that we have difficulty finding growth and electrophoretic conditions under which orthologous enzyme activities can be detected simultaneously for all the strains being compared.

    Notes: An experiment extending over a 2½-year period from June 1976 to October 1978 assessed the effect on herbage yield of method of establishment, species and level of N application.Perennial ryegrass cv. Gremie and Italian ryegrass cv. RvP were established by broadcasting or drilling into cultivated ground or by direct-drilling, using a Howard Rotaseeder, into paraquat-sprayed but uncultivated ground. These treatments were combined with annual rates of N application of 0, 120, 240 or 360 kg ha-1.Over the experiment drilling into cultivated ground gave the highest yield—3·6 and 4·8% higher than the broadcast and direct-drilled treatments, respectively. Italian ryegrass consistently outyielded perennial ryegrass, with an overall mean dry matter yield advantage of 23·7%. There was no significant interaction between sowing method and species; direct-drilled Italian ryegrass produced a mean yield 17·3% higher than that of drilled perennial ryegrass.

    Notes: The most likely reasons for seeding failures of sod-seeded lucerne (Medicago sativa L.) were determined from eight field experiments, where the effects of a range of pesticides on the establishment of and damage to lucerne were compared. The experiments were conducted during 1979–80 at four sites near Ithaca, N.Y., USA. Pesticides evaluated were: methyl bromide (MeBr), carbofuran, acylalanine (1979, 1980); fonofos, diazinon, methiocarb, metaldehyde and combinations of carbofuran with methiocarb or metaldehyde (1980). Seeded areas were pretreated with glyphosate for vegetation control.In 1979 seedling losses were decreased by carbofuran and methyl bromide whereas in 1980 seedling losses were decreased by methyl bromide at two sites and by methiocarb at all sites. Methiocarb was the only pesticide to reduce defoliation damage. The effect of combining methiocarb and metaldehyde each with carbofuran tended to be additive in terms of reducing seedling numbers and damage, and increasing yield. Treatment effects on lucerne yield were still apparent up to 12 months after seeding. Based on the consistent results of methiocarb, it was concluded that molluscs were probably the main factor limiting the establishment of sod-seeded lucerne but an arthropod component also appeared to be involved.

    Notes: Experiments were carried out over three seasons in which swedes were sown by precision drilling after ploughing and conventional cultivation or by direct drilling with two types of drill at two row widths with and without shallow cultivation. The swedes were sown in June or early July following a first silage cut and sward desiccation with paraquat. Precision drilling with inter-row cultivation and singling gave a uniform weed-free stand and had the highest mean yield of over 8 t DM ha−1 Where hoeing was omitted yields were reduced due to weed competition. Yields following direct drilling were lower than with precision drilling due to uneven plant establishment, weed competition and grass regrowth. A shallow rotavation prior to direct drilling improved uniformity and yields and reduced grass regrowth. Direct drilling produced high numbers of small roots. The highest root yields were associated with sowing in early June and harvesting in December or January. Overall yields from the silage cut plus direct drilled swedes and silage cut plus precision drilled swedes were 12 and 13 t DM ha−1respectively compared with about 14 t DM ha−1expected from a grass sward.

    Notes: The strains B 10.S(7R), B 10.S(23 R) and B 10.S(24R), all thought to be genetically identical, differ in levels of susceptibility to infection with Trichinella spiralis. In a series of nine independent experiments, B 10.S(7R) was shown to be more susceptible than the other two strains. In another series of seven experiments, the strain B 10.A(18R) was shown to be more susceptible to infection with T. spiralis than the strains B 10.S(21R) or B 10.BAR-5, all of which were thought to share common H-2 alleles. These results indicate that a gene mapping between the S and D loci influences susceptibility to infection with T. spiralis. Typing of these strains for Qa and Tl loci rule out the possibility of a double crossover accounting for the differences observed. The new gene is designated Ts-2. Previously published data have also been reinterpreted and another gene Ts-1 is shown to be associated with the Aβ locus. When the d allele is expressed at the Ts-2 locus, strains of mice expressing s, q, f or b alleles at Ts-1 are rendered more susceptible to infection.

    Notes: SUMMARY. Surveys of the Wye catchment in March and September 1975, and July and September 1976 and 1977, yielded 227 macro-invertebrate taxa, 77 of which were Chironomidae. Total macro-invertebrate density ranged from 520 to 22020 m−2. Total density estimates in July 1976. during a period of severe drought, were significantly higher than in July 1977 but no differences were established between estimates on other comparable dates in 1975, 1976 and 1977. The total number of taxa and total macro-invertebrates collected from sites with total dissolved solids concentrations less than 50 mg I−1 were significantly lower than at sites with higher concentrations. In general, upstream sites were dominated numerically by Plecoptera in March; Ephemeroptera, Trichoptera and Diptera were relatively more abundant during the summer months. Sites lower in the catchment were more variable in composition with Oligochaeta, Ephemeroptera, Trichoptera and Diptera all abundant at certain sites and times. The densities of Ecdyonuridae (Ephemeroptera). Hydropsychidae (Trichoptera) and Elminthidae (Coleoptera) in the impounded R. Elan, a tributary of the R. Wye, were significantly lower than at nearby sites on the Wye although no differences in total macro-invertebrate density were established. Average linkage clustering of Spearman rank correlation coefficients between selected taxa allowed the comparison of taxa groups with contemporary site groups by nodal analysis. Six major taxa groups were established and these characterized five site groupings.

A total of 155 taxa was collected from the Ystwyth (111) and Rheidol (134) catchments and total macro-invertebrate densities ranged from 435 to 9105 m−2 and 155 to 4290 m−2 in the two catchments, respectively. The fauna in each catchment was dominated by Insecta, principally Plecoptera, Ephemeroptera and Diptera. Trichoptera and Oligochaeta were proportionally more abundant in the Rheidol catchment. There was no evidence that the distribution or relative abundance of the faunas of the catchments were related simply to heavy metal contamination (mean zinc concentration up to 2.0 mg I−1 and 0.34 mg I−1 in the Ystwyth and Rheidol, respectively). Average linkage clusters of Spearman rank correlation coefficients suggested that community affinities were probably related principally to location within the catchment. There was no evidence in the Rheidol catchment that irregular discharges (up to c. 15 m3 s−1) of water for hydroelectric purposes had any substantial effect on the invertebrate fauna although total invertebrate density below the discharge was significantly higher than at an upstream site.
