    Notes: The product distribution of compounds 2, 3 and 4 formed in the electrohydrodimerization (EHD) of 1 was determined as function of the water content of the solvent (CH3CN). The presence of water favours the formation of the glycols 4 and reduces the relative amounts of 1,6-diketones 2 and hydroxyketones 3, as determined by GC./MS.-analysis.

    Notes: The parameters which influence the inverted wave appearing in the reverse (oxidation) sweep in the cyclic voltammogramme of the title compound 1 are discussed in detail. The inverted wave is due to the interaction of the reduction products of 1 with the electrode and the electrolyte (TBAFB). The key condition for the presence of the inverted peak is a slow scan rate on the reverse sweep, at least for the last 50 mV before the inversion of the current-potential curve.

    Notes: The reduction of cis-4 a-methyl-1,2,4a,7,8,8 a-hexahydronaphthalene-2,7-dione (1) was studied by cyclic voltammetry and product analysis. On a preparative scale, 6-hydroxy-3-methyltetracyclo[4.4.0.02,4.03,8]decan-10-one (2) was obtained in 22% yield via an intramolecular hydrodimerization/aldol reaction sequence. The CV. results (Hg, DMF) suggest that the cyclopropane ring is formed after the first electron transfer by coupling of the anion radical with the second C. C-double bond.

    Notes: Temperature control of subcooled reflux in a distillation column was studied experimentally using coolant flow rate as the manipulative variable. Methanol vapor was condensed and subcooled at one atmosphere in a vertical heat exchanger with 210 copper tubes (0.77 cm I.D., 91.5 cm long). Cooling water flowed countercurrently through the shell side (20.6 cm I.D.).A proportional-integral feedback controller provided good temperature control at normal design throughputs. But dynamic control performance deteriorated at low throughputs (or large areas). Thus the normal conservative design practice of providing excess area in this type of condenser leads to poorer temperature control.

    Notes: A monoclonal antibody of the IgG class, EGFR1, has been isolated using cells of the epidermoid carcinoma line A431 as immunogen. The A431 antigen recognized by EGFR1 has an apparent molecular weight of approximately 175,000, is a cell-surface molecule which can be specifically cross-linked to EGF, exhibits an EGF-stimulated protein kinase activity, binds to EGFR1 in a number of human cell lines to a degree which parallels EGF binding, and shows EGF-dependent internalization in A431 cells and human fibroblasts. We therefore conclude that EGFR1 is directed against an antigenic site on the human EGF receptor. EGFR1 is not mitogenic for human fibroblasts and does not inhibit EGF binding under a variety of assay conditions. The characterization of EGFR1 has allowed the unambiguous assignment of the structural gene for the human EGF receptor to chromosome 7. Preliminary results suggest that a convenient method for isolating a range of anti-EGF receptor monoclonal antibodies can be developed, based on a hybridoma supernatant screening assay in which positive supernatants bind selectively to a human-mouse cell hybrid containing human chromosome 7.

    Notes: PMR investigations on the diastereomeric phosphate methyl protons of the dinucleoside ethyl phosphotriesters Tp(C2H5)T, dA, and dIp(C2H5)dI have been used to study the conformational changes of these dimersin solution. In D2O (273°K), the diastereomeric phosphate-methly groups of Tp(C2H5)T appear as a triplet. The methyl resonances of dIp(C2H5)dI and dAp(C2H5)dA appear as two sets of triplets and their chemical shift differences (δ1 - δ2), decrease with increasing temperature, finally becoming zero at 292°K and 333°K, respectively. The same phenomenon is observed for dAp(C2H5)dA in CD3OD; in this detacking solvent, the difference (δ1 - δ2) diminishes to zero at a lower temperature (261°K). At room temperature in D2O, the chemical shift of the phosphate methyl of Tp(C2H5)T appears at lower field than those of dIp(C2H5)dI or dAp(C2H5)dA. The differences between the chemical shifts of these groups (δI - δT or δA - δT) increase with increasing temperature, and reach maximal values at 301°K and 333°K, respectively. The results suggest that at low temperature the largest fraction of the dimer population exists in a stacked state, with the phosphate-ethyl groups outside the stack. Increasing temperature causes an oscillation of the bases and a shift in the dimer population away from the stacked state. Finally at high temperature, the planar bases rorate with respect to one another and in the case of dIp(C2H5)dI and dAp(C2H5)dA, the ethyl groups experience shielding by the anisotropic ring current of the five-membered ring of the bases. Thus, the current pmr studies and those reported earlier from our laboratory support an “oscillation-rotation model” for the unstacking process of the dimers. The relationship of this model and the “two-state model” is discussed.

    Notes: The proton nmr spectra of the oligoribonucleotides in the series CpXpG, ApXpG, CpApXpUpG, and ApGpXpC (X = A, G, C, and U), together with the reference compounds CpG, ApG, CpApUpG, and ApGpC, have been measured. A complete analysis of all the nonexchangeable base protons and the ribose H-1′ protons was made. The insertion of a nucleotide X into a oligoribonucleotide led to shift changes at both nearest-neighbor and next-nearest-neighbor positions, which were rationalized in terms of the shielding abilities of the various bases. The derived shielding trends in the ApGpXpC series of compounds were successfully used to predict the chemical shifts of resonances in the related ApGpXpCpU series.

    Notes: Metallurgical observations of phenomena which influence the clinically successful performance of permanent implants and implant materials are reported. The effect of casting voids in cast cobalt chromium femoral stems of total hip replacements is discussed first. Pitting corrosion occurred in a retrieved stainless-steel implant type AISI 316 containing 2.7% Mo. The fractographic pattern of this device retrieved for fatigue failure shows typical characteristics of corrosion-enhanced fatigue.

    Notes: Platelet-derived growth factor (PDGF) is a potent mitogen for cultured cells of mesenchymal origin. Known sources of PDGF or PDGF-like protein are blood platelets, several transformed cell lines, and cultured endothelial cells (EC). We have examined the regulation of production of a PDGF-like protein in cultures of bovine aortic EC using a specific radioreceptor assay for PDGF. EC constitutively secreted PDGF-like protein into serum-containing or serum-free medium. The rate of production of PDGF-like protein was constant for at least 3 weeks and was not due to release of an internal store, since cell lysis by repeated freeze/thaw cycles did not relase significant amounts of the protein. Synthesis of PDGF-like protein was sensitive to changes in the pH of the media and was maximal at pH 8.5. Production of PDGF-like protein was independent of EC growth rate: rapidly dividing cells and confluent, quiescent cells produced equal amounts per cell. However, sparse, quiescent EC produced more PDGF-like protein per cell than did confluent, quiescent cells. Several phorbol esters stimulated production of PDGF-like protein. At a concentration of 10-6 M, a twofold stimulation was observed upon addition of the tumor promoter 12-O-tetradecanoylphorbol 13-acetate (TPA) and nearly a fourfold stimulation upon addition of the nonpromoting analog, methyl TPA. Incubation of EC with endotoxin (10 μ/ml) resulted in a twofold stimulation of PDGF-like protein production. In all experiments with endotoxin and phorbol esters, an increase in the production of PDGF-like protein was accompanied by morphological changes in the EC cultures. The cells appeared elongated and fibroblastic and exhibited low viability. A mathematical model was developed in which PDGF-like protein production was shown to consist of two separate components - production at a constant rate by healthy cells and a large burst of synthesis and secretion by dying cells. These results suggest that injurious agents may be capable of stimulating production of a growth factor by the endothelium.
