    Notes: In a study of A-B type block copolymers of γ benzyl-L-gultamate and β-benzyl-L-aspartate use has been made of the observations: (1) that for poly aspartate esters the chemical shifts of the α-CH and NH resonances are sensitive to the helix sense, (2) that in both helical and random coil conformations the same resonances of poly-γ-benzyl-L-glutamate are well separated from those of poly aspartates. Since the sense of poly-β-benzyl-L-aspartate is very sensitive to the inclusion of γ-benzyl-L-glutamate residues, the degree of overlap between the blocks can be studied by monitoring the helix sense of the aspartate. The ability of the NMR method to make separate observation of the two blocks removes the necessity of relying on an overall ORD parameter such as b0. The copolymers studied include those having lefthanded, righthanded, and mixed-sense aspartate, corresponding to differing degrees of overlap.

    Notes: Denatured histones H1 and H5 can be readily refolded on salt addition. Their digestion by trypsin leads to limit peptides of about 80 residues having the same nmr and CD spectra as those of the intact parent histones. Scanning microcalorimetry shows that (1) the folded structures of H1 and H5 are located entirely in their limit peptides; (2) both have values of the specific denaturation enthalpy typical for small globular proteins; and that (3) both exhibit a classic “2-state” transition (ΔHdcal = ΔHdvan't Hoff). The heat-denaturation profiles of H5 measured using intrinsic and extrinsic Cotton effect and side-chain nmr peaks do not coincide at all. Only the intrinsic Cotton effects give a Tm and ΔHdvan't Hoff close to that from microcalorimetry. We conclude that these proteins exhibit large-scale side-chain motions that precede the macroscopic cooperative transition.

    Notes: Numerical solutions for two-dimensional or axisymmetric viscous fluid flow problems are usually based on the stream function/vorticity formulation. Frequently, however, the pressure distribution is of prime interest. Difficulties have been reported in the literature with the use of obvious pressure marching schemes. Consequently, several investigators have preferred to use an iterative method which involves solving a Poisson equation with Neumann boundary conditions. In this paper, the fundamental cause of failure of the marching schemes is investigated. The authors introduce the concept of compatible pressure and vorticity schemes and show that lack of compatibility has been the principal reason for the poor results obtained using marching schemes. Compatible pressure marching methods are developed and shown to give good results. Comparisons are made between the Poisson equation method and the compatible marching method. To make the comparisons meaningful, special test cases with analytical solutions have been used.

    Notes: High-resolution NMR is used to study two series of benzyl-L- or D-glutamate: benzyl-L-aspartate random copolymers. The helix sense of the L-aspartate residues determined from the αCH and NH chemical shifts agrees with that obtained from ORD. The stability of the helical copolymers to TFA addition shows a minimum at the composition of helix-sense inversion for the L-glutamate:L-aspartate copolymers but no minimum for the lefthanded D-glutamate: L-aspartate series. The helix-coil transition of the glutamate and aspartate residues in each polymer is compared. For the D-glutamate: L-aspartate series no differences are found, indicating random copolymerization. In the L-glutamate:L-aspartate series significant differences between the transition midpoints are interpreted as resulting from irregular distribution of component residues along the chain.

    Notes: The helix-coil transition has been studied by high-resolution NMR for three water-soluble polypeptides. Such systems are better models for protein behavior than those in TFA-CDCl3 solvent. An upfield shift of ∼7 cps is observed for the α-CH peak of poly(L-glutamic acid) and poly-L-lysine as the helix content increases over the transition. No such shift is found for copoly(L-glutamic acid42, L-lysine28, L-alanine30). The width of the α-CH peak for poly L-lysine increases rapidly as helix content rises but for poly L-glutamic acid and the copolymer, the width of this peak remains unchanged up to 60% helicity. This demonstrates a rapid rate of interconversion between helical and random conformations in partly helical polymer for the latter two polypeptides. All three polymers however, show no apparent α-CH peak at 100% helicity. Side-chain resonance lines also broaden as helix content increases and, to a greater extent, the closer the proton is to the main chain.

    Notes: Two mutant Chinese hamster lung fibroblast lines have been isolated that are resistant to the the toxic proline analog L-azetidine-2-carboxylic acid. The line designated AZCA-1 has 30-fold elevated activity of pyrroline-5-carboxylate synthase and a large increase in the rate of proline production and release compared to controls. Pyrroline-5-carboxylate synthase activity is not elevated in the resistant line designated AZCA-4, but the enzyme is less sensitive to inhibition by ornithine and proline than control enzyme. Intracellular proline is elevated in AZCA-4 cells, with no change in the rate of release of proline synthesized from glutamate. Resistance to azetidine carboxylic acid in both mutant lines is attributed to the expanded intracellular proline pool that results from alterations in pyrroline-5-carboxylate synthase. These results indicate that intracellular proline levels are determined at least in part by the regulated activity of pyrroline-5-carboxylate synthase.

    Notes: Singlet methylene was reacted with cyclopentadiene to give chemically activated bicyclo[3.1.0]hex-2-ene (BCH). The rate of isomerization of BCH to 1,4-cyclohexadiene, 1,3-cyclohexadiene, cis-1,3,5-hexatriene, and l-methylcyclopentadiene is compared with calculated rate constants using the RRKM theory and measured or estimated thermal Arrhenius parameters. Subsequent isomerizations of the C6H8 products are also measured and calculated. These include 1,4-cyclohexadiene to benzene and the reversible reactions between 1,3-cyclohexadiene, cis-1,3,5-hexatriene, and trans-1,3,5-hexatriene. The results provide new data for several of these reactions which have not been observed in thermal studies. Agreement between the observed and calculated rates using the strong collision assumption is satisfactory except for the trans-1,3,5-hexatriene to cis-1,3,5-hexatriene reaction.
