Notes: Ordinary corn kernel endosperm is composed of horny and floury portions. Corn varieties having different endosperm were investigated. Their chemical composition was almost identical, but in tissue structure the floury endosperm is soft and the horny endosperm is hard. The starch isolated from floury endosperm is easier to gelatinize, and higher in viscosity, swelling value, and α-amylase digestibility than the starch from horny endosperm. The floury endosperm is superior to the horny endosperm in such baking products as bread and cookies.

Description / Table of Contents: Summary Poly-p-xylylene crystallizes from some solvents into two single crystal forms, rectangular and hexagonal. These two single crystals show different X-ray and electron diffraction patterns, respectively. By X-ray measurements it was found that these two single crystals correspond toα- undβ-crystal forms, which were observed with bulk materials byBrown andFarthing. It was also found that these morphological forms depend on the crystallization-conditions, especially on crystallization-solvents. The annealing effects and the transformation fromα- toβ-form were studied by X-ray diffraction and differential thermal analysis.

Notes: Abstract The nonlinear optical coefficientd ij(kl) (2ω, 0,ω, ω) for electric field-induced optical second harmonic generation in semiconductors is calculated by means of a perturbation treatment. The result is given directly as a Brillouin zone integral over a resonant energy denominator. A simplified energy band structure model is used to carry out the Brillouin zone integral. The analytic closed-form expression ford ij(kl) (2ω, 0,ω, ω) thus obtained permits the calculation of the absolute value of its spectrum from available energy band parameters. The dispersion ofd 11(11) (2ω, 0,ω, ω) of Ge is numerically calculated, second harmonic photon energies being close to theE 0 andE 1 gaps. The results show pronounced resonant behaviours.

Notes: Summary A prenatal diagnosis of GM1-gangliosidosis was made in a pregnancy at risk, on the basis of a deficiency of β-galactosidase activity demonstrated in cultured aminiotic fluid cells. Biochemical analyses were performed in the aborted fetus. GM1-ganglioside β-galactosidase activity was reduced to 1% of the control value in both the brain and liver of the affected fetus. Lamellar bodies suggestive of membranous cytoplasmic bodies were found in cells of basal ganglions, while the accumulation of GM1-ganglioside in the brain was not remarkable.

Notes: Summary Oxidized glutathione (GSSG) markedly stimulated specific insulin binding, labelled glucose oxidation, and 2-deoxyglucose transport in rat adipocytes in a concentration-dependent manner. Reduced glutathione was inhibitory. Insulin stimulated in the presence of reduced and oxidized glutathione, but the effect was markedly diminished or absent at the highest concentrations of reduced or oxidized tripeptide. Oxidized glutathione did not activate % glycogen synthaseI activity. The stimulation of % glycogen synthaseI activity by insulin was decreased by oxidized glutathione in a concentration-dependent manner. In the presence of glucose, oxidized glutathione minimally activated glycogen synthase, and the stimulation by insulin again was reduced with increased oxidized glutathione concentration. Preincubation of rat adipocytes with N-ethyl-maleimide (NEM) for one minute abolished basal labelled glucose oxidation, basal 2-deoxyglucose transport, and also the insulin-stimulated activities. The concentration of NEM required to inhibit the insulin-stimulated activities (0.2 mM) was considerably lower than that required to inhibit basal activities (0.5 mM and greater), suggesting an involvement of at least two different populations of SH groups. Very short (5 s) preincubation with NEM abolished the stimulation of % glycogen synthaseI observed with insulin. Preincubation of adipocytes with insulin prevented this action of NEM on glycogen synthase indicating a cell membrane site of inhibition by NEM. Increased labelled glucose oxidation and 2-deoxyglycose transport observed with increased oxidized glutathione concentrations, as well as the insulin stimulation, were abolished by preincubation of adipocytes with trypsin or cytochalasin b, suggesting that trypsin-sensitive and cytochalasin B-binding protein(s) presumably in the cell membrane are involved in the activation by oxidized tripeptide. Oxidized glutathione thus acts at the cell membrane as a perturbant to influence cell membrane binding of insulin and hexose transport, but does not act to control intracellular glycogen synthase activation, similarly to the increase in pH, detailed in paper I and distinctly different from trypsin to be discussed in paper III.

Notes: Summary The effects of pH, oxidation reduction compounds and trypsin on insulin binding, hexose transport, and activation of glycogen synthase were studied utilizing rat adipocytes. In this paper the effect of pH is examined; while in the subsequent two papers the effects of glutathione and trypsin are examined. Increase in pH from 6 to 8.5 increased labelled glucose oxidation, 2-deoxyglucose transport as well as labelled insulin binding to the receptor. Enhanced insulin binding was due to an increased rate of association k+1 with no effect the rate of dissociation k−1 resulting in a decreased equilibrium dissociation constant KD. Glycogen synthase activity was unaffected by increase in pH when adipocytes were incubated with or without glucose. Insulin in contrast to pH was effective in increasing the activity of glycogen synthase. With 2-deoxyglucose, % glycogen synthaseI activity was increased by an increase in pH. Glycogen synthase activity was thus stimulated by insulin by the direct mechanism, previously termed mechanism 1, involving the formation of a chemical mediator, and clearly distinguishable from the activation of hexose transport, previously termed mechanism 2(1). Increase in labelled glucose oxidation and in 2-deoxyglucose transport with increased pH, as well as insulin stimulation, was abolished by preincubation with trypsin, or cytochalasin B; suggesting that trypsin-sensitive and cytochalasin B-binding protein(s) presumably in the plasma membrane are involved in these effects of pH. Since increase in pH alone activates cell membrane-mediated hexose transport and insulin receptor binding under conditions where glycogen synthase is not activated, increase in pH acts presumably by a non-mediator mechanism. Insulin acts at the membrane to enhance further the effects of increased pH, via a mediator mechanism.

Notes: Summary Brief treatment of rat adipocytes with low concentration of trypsin activated both cell membrane and intracellular insulin-sensitive functions in marked contrast to H2O2 (1), increase in pH, and oxidized glutathione (Papers I and II). Glucose oxidation was activated maximally by trypsin in 30 s and preceded maximal activation of glycogen synthase, which occurred in 60 s. Trypsin action to activate glycogen synthase was further enhanced by insulin. Mitochondrial pyruvate dehydrogenase was also rapidly activated by trypsin. With both insulin and trypsin action, mediator generation was directly demonstrated by glycogen synthase phosphoprotein phosphatase activation. Trypsin is thus the most insulin-like of these four agents studied since it acts by the formation of chemical mediator peptide(s) which are similar but not identical to those produced by insulin.
