    Notes: In order to correctly interpret the chemical images obtained using ion microscopy (IM), it is useful to correlate them with the information provided by conventional light microscopy (LM), secondary electron imaging (SEI), backscattered electron imaging (BEI), and electron probe microanalysis (EPMA). Accordingly, we have devised a technique of specimen preparation which allows for the application of several different microanalytical techniques to a single histologic section mounted on the same substrate. Sections are cut onto polyester plastic coverslips (devoid of peaks for any element with atomic number 〉 9 using EPMA) and studied by LM. After a light rotary coating with carbon (to prevent charging), the section can then be examined by SEI, BEI, and EPMA. Specific areas can be marked for IM study either with an objective-mounted pin tissue microlocater, or by placing small pieces of metal foil, cut in specific geometric shapes, over features of interest. After sputter-coating the sample with platinum, metal-free shadows are visible using a low-power reflected light microscope available on a typical IM sample chamber as a guide for ion beam placement. The conductive coatings also minimize specimen charging during IM. Post-IM light microscopy, SEI, and BEI are used to confirm the location of specific areas probed in the IM experiments and to provide information on differential ion-sputtering artifacts and tissue contaminants. This new correlative technique should permit better understanding of the images obtained with these diverse instruments.

    Notes: We have found that exogenous leukotriene B4 modifies calcium homeostasis in rabbit neutrophils in a manner essentially analogous to that of the chemotactic peptide f-Met-Leu-Phe. Leukotriene B4 causes a rapid and dose-dependent increase in membrane permeability to calcium and a release of calcium from previously unexchangeable intracellular pool(s). The net result of these changes is to transiently elevate the intracellular level of exchangeable calcium. A stereoisomer of leukotriene B4 with greatly reduced secretory activity toward neutrophils (5S, 12S-di HETE) is essentially without effect on the rate of 45Ca uptake at concentrations equal to those that produce near maximal enhancement by leukotriene B4. Leukotriene B4, in addition to its effects on calcium metablolism, also increases the rate of 22Na influx into rabbit neutrophils. The relationships between the action of leukotriene B4 on calcium homeostasis and the neutrophil-directed activities of arachidonic acid and its lipoxygenase metabolites are discussed.

    Notes: The possible involvement of membrane-bound calcium in the mechanism of action of leukotriene B4 was examined using the fluorescent chelate probe, chlortetracycline. Leukotriene B4 was found to cause a rapid release of membrane-bound calcium at physiologically relevant concentrations. This effect of leukotriene B4 is stereospecific and its magnitude is decreased upon the transformation of leukotriene B4 into its ω-hydroxy and ω-carboxy metabolites. The pool of calcium affected by leukotriene B4 appears to be the same as that released by other chemotactic factors such as formyl-methionyl-leucyl-phenylalanine (f-Met-Leu-Phe). Similarly, preincubation with f-Met-Leu-Phe results in a decreased responsiveness of the cells to the addition of leukotriene B4. These results extend further the analogy between the mechanism of action of peptidic and lipid chemotactic factors, and emphasize the central role of the intracellular redistribution of calcium, as inferred and monitored by chlortetracycline fluorescence and steady-state isotopic flux studies, in neutrophil activation.

    Notes: Five to six percent (by mass) of AKR-2B mouse embryo cell polysomal RNA consists of messenger RNA sequences which may exist in polyadenylated form. In the steady state, however, only 30-40% of these molecules are retained by extensive passage over oligo(dT)-cellulose, the remainder being present in the form of poly(A)-deficient analogues. Within experimental limits, these poly(A)-deficient analogues contain representatives of all poly(A)-containing mRNA sequences in these cells. An analysis of the kinetics of hybridization of cDNA probes enriched for either abundant or rare poly(A)-containing mRNA sequences suggests that the frequency distributions of poly(A)-containing and poly(A)-deficient analogues are dissimilar, and that a relationship exists between the intracellular frequency of a given mRNA sequence and the number of poly(A)-deficient analogues of that sequence. High frequency sequences appear to be enriched in the poly(A)-containing fraction, while low frequency sequences are predominately associated with the poly(A)-deficient fraction, thus, poly(A) may play a role in the regulation of mRNA frequency in the cytoplasm.

    Notes: We have begun to characterize the development of the excitation - response coupling sequence in the human promyelocytic leukemia cell line HL60. Using the recently developed fluorescent calcium probe quin-2, it was found that DMSO induced myeloid differentiation of the HL60 cells is accompanied by the development of a calcium response to the addition of the chemotactic factors fMet-Leu-Phe and leukotriene B4. The characteristics (time course, concentration dependence, stereospecificity, and metabolic dependence) of the calcium response are extremely similar to those previously described in human neutrophils. These results imply that functional receptors for leukotriene B4 appear in HL60 cells upon the induction of differentiation and also lend strong support to the use of these HL60 cells as a model of human myeloid differentiation. We have also characterized the emergence of a secretory response to fMet-Leu-Phe and leukotriene B4 in cytochalasin B treated HL60 cells. In addition, it is found that differentiation was required for the calcium ionophore A23187 to express its secretory activity toward the HL60 cells. This last set of results implies that differentiation is accompanied by the coordinated appearance of surface receptors and cytoplasmic factors required for the expression of cellular responsiveness.

    Notes: Ovaries from pubertal mice were dissected into fragments containing 1-4 antral follicles, which were cultured on the surface of a chemistry defined medium. After 17-20 hrs oocytes were released from the cultured follicles and their meiotic status recorded. In the presence or absence of exogenous gonadotropins oocytes in these cultures behaved meiotically as expected if they had been studied in the intact animal. Media containing 100 μM of progesterone for the duration of the culture period caused atresia of the oocytes, and although the addition of gonadotropins reduced this necrosis they did not induce the resumption of meiosis. The meiotic stimulating effect of gonadotropins was also inhibited if progesterone was present in the culture media for the first 3 hours only. Estradiol (100 μM) similarly inhibited the meiotic inducing action of gonadotropins when left in the medium for the duration of culture, but had no effect when administrated for a shorter period. The results from this model system, would indicate that the meiotic-initiating action of gonadotropins can be modulated by exposure to progesterone and estradiol.

    Description: The planetary nebulae LMC P40, SMC N2, and SMC N5 and their central stars were observed with IUE. The C abundances in the nebulae, compared with those in galactic planetaries, indicate that convective dredgeup of locally nucleosynthesized C has occurred. The progenitors of the nebulae were C stars at the theoretical upper luminosity threshold, thus such stars do occur as predicted, although none so bright have been found in the Clouds. The central stars of the nebulae have masses approximately 1 solar mass, luminosities approximately 40,000 solar luminosity, and radii approximately 0.7 solar radius; they have probably not yet reached their maximum luminosities. With M(subv) 19.1-19.8, they may be the visually faintest stars yet observed by UV spectroscopy. Clearly, it is not true that planetary nebulae nuclei all have masses M = (0.6 + or - 0.1) solar mass.

    Description: Lightcurves of asteroid 44 Nysa obtained during 20 nights in 1979 as part of a global compaign are presented. The synodic period was 6 hours and 25.3 minutes. The phase coefficient of the primary maximum was 0.026 mag/deg and the absolute V magnitude 7.05. The phase function is linear from 2 to 25 deg, no opposition effect is present.

    Description: Five pairs of isolated pairs of galaxies with radio sources from the sample of Stocke et al. (1978) were observed with a three-element VLBI array operating at 4996 MHz. Parsec-scale components were detected in NGC 3894, Arp 102, and NGC 6500. Arp 102B was found to be a strong X-ray source with a luminosity in the 0.5-4.5 keV band of about 10 to the 43rd ergs/sec. Arp 102B was also found to be an elliptical galaxy for which no emission lines have been reported, and was the only source in the sample which appeared unresolved on the milli-arcsec scale, indicating that it may be related to BL Lac type objects.

    Description: The amount of telluric water vapor along the line of sight of the Kuiper Airborne Observatory telescope as obtained concommitantly on 23 flights is compared with the NASA-Ames Michelson interferometer and with the NOAA-Boulder radiometer. A strong correlation between the two determinations exists, and a method for computing the atmospheric transmission for a given radiometer reading is established.
