    Notizen: A prominent feature of tip growth in filamentous plant cells is that the nucleus often migrates in step with the tip as it extends. We have studied this long-recognized but unexplained relationship in root hairs of the legume Vicia hirsuta by a variety of microscopic techniques. Using rhodaminyl lysine phallotoxin, and antitubulin antibodies, root hairs are shown to contain axial bundles of F-actin and a complex microtubular system. To the basal side of the nucleus the microtubules are cortical and net axial but in the region between nucleus and tip the arrangement is more complicated. Electron microscopic thin sections demonstrate that internal bundles of microtubles exist in addition to the plasma membrane-associated kind. Computerized deblurring of through-focal series of antitubulin stained hairs clarifies the three-dimensional organization: bundles of endoplasmic microtubules progress from the nuclear region toward the apical dome where they can be seen to fountain out upon the cortex.The relationship between nucleus and tip can be uncoupled with antimicrotubule herbicides. Time lapse video microscopy shows that these agents cause the nucleus to migrate toward the base. This contrary migration can be inhibited by adding cytochalasin D, which fragments the F-actin bundles.It is concluded that microtubules connect the nucleus to the tip but that F-actin is involved in basipetal migration as is known to occur when symbiotic bacteria uncouple the nucleus from the tip.

    Notizen: β-D(+)-Glucose pentaacetate was hydrolyzed both chemically and enzymatically. In contrast to the alkaline hydrolysis, esterase-catalyzed deacetylations afforded significant accumulation of intermediate glucose esters at different degrees of substrate conversion. Aspergillus niger lipase, the most suitable of the four enzymes tested, was used for preparative hydrolysis of glucose pentaacetate. As a result, gram quantities of pure glucose-2,3,4,6-tetraacetate, glucose triacetate (a mixture of two positional isomers, 2,4,6- and 3,4,6-), and glucose-4,6-diacetate were prepared.

    Notizen: The relationship between total glutathione (GSH) content and cell growth was examined in 3T3 fibroblasts. The intracellular GSH level of actively growing cultures gradually decreases as these cells become quiescent by either serum deprivation or high cell density. Upon mitogenic stimulation of sparse, quiescent (G0/G1) cultures with serum, there is a rapid 2.3-fold elevation in intracellular GSH levels which, is maximal by 1 h and returns to baseline by 2 h. This is followed by a more gradual increase in GSH content as cells enter the S phase. In addition, the elevation in GSH content is required for maximum induction of DNA synthesis. Treatments that prevent the early increase in intracellular GSH levels do not affect protein synthesis but result in a reversible dose-dependent decrease in the percent of cells capable of entering S phase. These results indicate that GSH may be important in the regulation of cellular proliferation.

    Notizen: Cumulus-intact and -denuded unfertilized oocytes from two mouse strains were exposed to 1.5 m ethanol (EtOH) or two cryoproteclant solutions, 1.5 M propanediol (PROH) or 1.5 M dimethylsulfoxide (DMSO), for 4.5 min at 27°C, and the proportion of activating or degenerating oocytes studied. Exposure to DMSO did not significantly increase activation above that of oocytes not exposed to DMSO. Treatment of oocytes in PROH resulted in the activation of up to 87% of viable oocytes. This was significantly higher (P 〈01) than in control oocytes and comparable to the rate of activation after treatment with EtOH (59-96% activation). In solutions at 1°C, 47% of control oocytes were activated, which was not significantly different from the rate of activation in EtOH (36%) or PROH (50%) at 1°C. Following treatment with PROH, up to 87% of oocytes degenerated within a period of 6 h in vitro. The age of the oocytes (h post hCG) and the time of cumulus removal with the enzyme hyaluronidase, relative to the time of exposure to the chemicals, influenced the level of degeneration in most groups. Significantly fewer oocytes degenerated when cumulus cells were removed before treatment (0-31%) than when the cumulus was left intact throughout the treatment and 6 h culture period (10-87%). Exposure to PROH at 1°C reduced oocyte degeneration to 5%. We conclude that PROH causes significantly greater losses of oocytes as a result of parthenogenetic activation and degeneration than of exposure to DMSO.

    Notizen: High-resolution transmission electron microscopes operating at 300 and 400 kV were used to investigate the crystallography and microstructure of the perovskitelike YBa2Cu3O7-x. In this paper, we evaluate the performance attainable with these microscopes both empirically and by computer modelling. Based upon the assumption that oxygen may be a key to superconductivity properties, we have also investigated the visibility of the oxygen sites as well as the heavier yttrium and barium ion positions and the lighter Cu atom positions. We propose a scheme for observing different twin orientations in these structures and hence the oxygen atom positions seen in projection for the [100] and [010].Our observations of both thick and thin regions of Y-Ba-Cu-O materials are reported as well as the problems of adjusting microscope parameters and specimen alignment to obtain interpretable images. We also give a preliminary report on the effects of heat treatment as seen in high-resolution micrographs to assess disorder of the heavy atoms and oxygen vacancies.
