    Notes: Several structurally similar, biologically active macrocyclic trichothecenes were ionized under thermospray conditions followed by collisionally activated dissociation of the ammonium adducts. Most of the observed daughter ions were formed by bond cleavage at the exocyclic ester bridges. Compounds with similar ester bridges formed several common daughter ions and underwent similar neutral losses. Fragmentation pathways proposed for the dissociation of the adducts were confirmed from the corresponding neutral loss spectra. Simple experiments designed for the sequential monitoring of the characteristic daughter ions were used for the rapid, direct and accurate analysis of macrocyclic trichothecenes in real samples. The primary drawback of the method is its inability to distinguish between isomers.

    Notes: A number of cell-cycle-specific temperature-sensitive (ts) mutants have been isolated from animal cells, especially Syrian hamster cells. These ts mutants, like cell cycle ts mutants of yeast, can be complemented by specific genes, some of which have been molecularly cloned. We have isolated a cDNA clone that complements TK- ts13 cells, but only temporarily. This clone, called B1, differs from a previously isolated clone (Sekiguchi et al.: EMBO Journal 7: 1683-1687, 1988) that specifically complements ts13 cells. In addition, B1 also complemented temporarily three other ts mutants of the cell cycle, tsAF8, ts694, and ts550C cells. These mutants have different mutations since, in cell fusion experiments, they complement each other. Sequencing of the B1 cDNA clone revealed that it was a mutant of human ADP/ATP translocase in which some human sequences at the 5′ end have been replaced by SV40 sequences. The wild-type translocase was less effective but could still increase the survival time of cell cycle ts mutants at the restrictive temperature. Using the polymerase chain reaction, it was possible to demonstrate that the B1 plasmid is expressed in TK-ts13 cells undergoing temporary complementation.

    Notes: The tricyclic indole derivative 6-hydroxymethtryptoline (7-hydroxy-1-methyltetrahydro-β-carboline) was identified as a normal constituent of human and cat urine. The identification was based on the correct retention time on a chiral capillary gas chromatographic column and on selected ion monitoring of the molecular ion with the mass spectrometer set at high resolving power (RP 10 000). The quantitation utilized a 13C-labeled analog as internal standard and demonstrated levels of 135 ± 35 and 174 ± 42 (pmol ml-1, mean ±SEM) for the S(-) and R(+) enantiomers, respectively, in human urine. In cat urine, the levels were 203 ± 55 and 108 ± 44 (pmol ml-1, mean ± SEM) for the S(-) and R(+) enantiomers, respectively. In human urine, the compound occurred predominantly in a conjugated form while in cat urine the free compound predominated. It was concluded that 6-hydroxymethtryptoline is naturally occurring in mammalian urine and that it most likely constitutes a metabolite of endogenous methtryptoline (1-methyltetrahydro-β-carboline).

    Notes: An analytical procedure for the isomer-specific quantification of polychlorinated dibenzo-p-dioxins and dibenzofurans in samples of human and animal origin is described. It consists of a large-scale clean-up including various kinds of column chromatography and high-resolution gas chromatographic/mass spectrometric quantification referring to internal 13C-labelled standards. By additional application of gel-permeation chromatography and the use of a retention gap, detection limits in the low ppq range (10-15) were achieved.

    Notes: Cytokines are polypeptides released by activated vertebrate blood cells which have profound effects on other blood cells and which have hormone-like properties affecting other organ systems as well. In recent years a wide variety of these mediators has been isolated and characterized. Many of these molecules have subsequently been cloned and expressed in E. coli. The tremendous importance of these proteins to host immune and non-specific defense systems along with the striking similarities of their properties among different species suggested to us that cytokines may have been proteins that have been conserved through evolution. Investigations of the evolution of cytokines will help us decipher the complex cellular, humoral and molecular interactions that regulate host defenses. Studies of the invertebrates will shed light on the phylogenetic emergence of these molecules as well.

    Description: Estimates of global aerosol production suggest that mobilization of natural eolian material greatly exceeds anthropogenic-related emissions, and it appears that soil material transported from arid regions by wind might be mainly responsible for the distribution of certain clay materials in oceanic sediments. In connection with studies related to an investigation of these possibilities, the present paper provides a discussion of the aerosol spatial distribution and its water-soluble chemical composition in the tropical North Atlantic troposphere during the ABLE-Barbados mission. Particular attention is given to the composition of the water-soluble fraction, since its chemical reactivity is important with respect to various atmospheric and biogeochemical processes. On the basis of the obtained results, it is suggested that Saharan dust has also a significant impact on the aerosol chemistry over the tropical North Atlantic.

    Description: Airborne differential absorption lidar and in situ data obtained during the April 20, 1984 flight experiment conducted over Nevada and California are analyzed. The O3 and aerosols profiles and in situ measurements reveal a 2.0-km-deep layer (with high O3 concentrations and enhanced aerosol backscattering) and a correlation of 0.8 between O3 and aerosol backscatter (with both values decreasing about 25 percent along the central axis of the fold). It is observed that the cold boundary of the fold has weaker gradients, larger-scale undulations, and more irregularity than the warm boundary. The potential vorticity distribution along the flight path was derived from radiosonde data. A positive correlation between the O3 mixing ratio and the potential vorticity values in the fold is detected; the average ratio between O3 and potential vorticity is 50.2 ppbv/10 to the -5th sq cm deg per g s.

    Description: A set of 14 pairs of vertical profiles of ozone and carbon monoxide, obtained with fast-response instrumentation, is presented. Most of these profiles, which were measured in the remote troposphere, also have supporting fast-response dew-point temperature profiles. The data suggest that the continental boundary layer is a source of tropospheric ozone, even in October and November, when photochemical activity should be rather small. In general, the small-scale vertical variability between CO and O3 is in phase. At low latitudes this relationship defines levels in the atmosphere where midlatitude air is being transported to lower latitudes, since lower dew-point temperatures accompany these higher CO and O3 concentrations. A set of profiles which is suggestive of interhemispheric transport is also presented. Independent meteorological analyses support these interpretations.
