Notes: Hands can be used to demonstrate the three-point model of chiral recognition. The points of attachment are thumb, forefinger, and middle finger. This vivid model has the advantages of simplicity, perspicuity, and availability at any time, although two persons are necessary. It can be shown that two interactions are not sufficient for chiral recognition but that three attractive or two attractive and one repulsive attraction are needed. It can also be used to explain some possibilities of weakening or elusion of the three-point model.

Notes: Tetrachlorodiphenoquinones have the same exact mass and elemental composition as the toxic environmental contaminant 2,3,7,8-tetrachlorodibenzo-p-dioxin. However, analysis of 3,3′-5,5′-tetrachlorodiphenoquinone showed a pronounced tendency toward chemical reduction in the mass spectrometer to the quinol compound, producing a molecular ion two mass units higher than 2,3,7,8-tetrachlorodibenzo-p-dioxin. Distinct differences were also apparent between the mass spectral fragmentation patterns of 3,3′,5,5′-tetrachlorodiphenoquinone and 2,3,7,8-tetrachloridibenzo-p-dioxin. The 3,3′,5,5′-tetrachlorodiphenoquinone spectrum shows a successive loss of carbon monoxide, with the most prominent fragment corresponding to loss of two molecules of carbon monoxide plus chlorine. In the mass fragmentation of 2,3,7,8-tetrachlorodibenzo-p-dioxin carbon monoxide loss is suppressed, but loss of one molecule of carbon monoxide plus chlorine is a major fragment ion. During an alumina column clean-up procedure 3,3′,5,5′-tetrachlorodiphenoquinone did not coelute with the fraction containing 2,3,7,8-tetrachlorodibenzo-p-dioxin. This evidence indicates that tetrachlorodiphenoquinones are unlikely to interfere with mass spectrometric determination of 2,3,7,8-tetrachlorodibenzo-p-dioxin in environmental samples.

Notes: Because of their incompatibility and the different refractive indices of the homopolymer components, polyurethane/polystyrene interpenetrating polymer networks are turbid by nature. Different parameters likely to enhance their transparency are examined: the crosslink density of each network and the level of internetwork grafting. The results prove that the latter factor is the most effective, as in some cases, very clear and transparent samples are obtained. Correspondingly, preliminary investigations of the dynamic mechanical properties show an inward shift of the glass transition temperatures for such systems. It is concluded that parameters able to cause a higher degree of phase dispersion can yield transparent materials.

Notes: The structural organization of the compound eye of the largest known isopod, Bathynomus giganteus, is described from four specimens maintained in the laboratory for as long as two months. Living specimens have not previously been available for study. The two triangular compounds eyes measure about 18 mm on the dorsal edge and are separated by an interocular distance of 25 mm. They face forward and slightly downward and may have significant overlap in visual fields. Each eye contains about 3,500 ommatidia in animals of body lengths from 22.5 cm to 37.5 cm. The packing of ommatidia is not uniform across the retina, but is nearly hexagonal in the dorsal central region and nearly square in the ventral and lateral periphery. The dioptric elements in each ommatidium consist of a laminar cornea, which is flat externally and convex internally, and a bipartite crystalline cone. Sometimes seven and sometimes eight retinular cells closely appose the proximal tip of the cone and bear the microvilli of the rhabdom. Proximal to the rhabdom the retinular cells form thin pillars near the periphery of the ommatidium, and the central portion along the optic axis at this level is occupied by interstitial cells that contain massive arrays of clear vesicles thought to serve as reflective elements. The arhabdomeral segments of the retinular cells and the interstitial cells rest on a basement membrane. Within each ommatidium the basement membrane has two extensions with cylindrical cores and thin sheets of dense material and collagen-like filaments. These sheets occupy spaces between adjacent interstitial cells up to the level of the rhabdomeral segments of the retinular cells. Arrays of pigment cells with relatively weak light-screening properties separate adjacent ommatidia.Animals were fixed both in light within a week of being brought from depth into daylight, and after 2 months of maintenance in constant darkness following such daylight exposure. In both cases, microvilli of the rhabdom were severely disrupted and the retinular cytoplasm contained numerous multivesicular bodies. Exposure to natural daylight appears to cause irreversible structural damage to the photoreceptors of these animals.

Notes: Fermentation (stoichiometric) equations are derived for anaerobic fermentations of propionic-acid bacteria (of both the Propionibacterium and acrylate pathways) and for production of various oxychemicals (butanol, acetone, isopropanol, butanediol, butyrate, acetate, propionate, succinate, lactate, and acrylate) from pentoses, hexoses, and cellobiose. The derivations of the equations are based on the fermentation biochemistries of the various bacterial classes. The validity of the equations is tested using fermentation data from the literature. The equations are shown to be valuable, among other uses, for calculating maximal yields and selectivities of the various fermentation products, as “gateway sensors” for monitoring of the fermentations, and for calculating the extents of the various intracellular reactions of the fermentation biochemistry.

Notes: A fermentation system has been designed to demonstrate the use of gas chromatography (GC) for on-line monitoring of the butanol-acetone and other complex saccharolytic fermentations. Tangential flow ultrafiltration was used to sterilely and continuously obtain a cell-free filtrate from the fermentation broth for on-line GC analysis of butanol, butyrate, acetate, acetone, ethanol, and acetoin. The liquid injection system consists of a phosphoric acid contactor, a slider-type injection valve, and a heater to address the difficulties (ghosting) encountered in the analysis of carboxylic acids. The fermentation headspace gas was also analyzed by on-line GC for nitrogen and carbon dioxide, while hydrogen was measured by difference. Raw chromatographic data were analyzed by a chromatography data system. Both raw and processed data were transmitted to a VAX 11/750 computer for further processing (using the fermentation equation) and archiving. The fermentation equation, which has recently been derived and tested on completed fermentation data, was also found to be valid during transient fermentations and thus useful as a gateway sensor for calculating various fermentation parameters on-line. Such parameters include glucose concentration and gas composition, as well as a number of unobservable parameters (such as YATP, excess ATP, and NAD reduced by FdH2), which characterize the state of the fermentation.

Notes: An artisanal static process for protein enrichment of cassava by solid-state fermentation, developed in laboratory and tested on pilot units in Burundi (Central Africa), provides enriched cassava containing 10.7% of dry matter protein versus 1% before fermentation. Cassava chips, processed into granules of 2-4-mm diameter, are moistened (40% water content) and steamed. After cooling to 40°C, cassava is mixed with a nutritive solution containing the inoculum (Rhizopus oryzae, strain MUCL 28627) and providing the following per 100 g dry matter: 3.4 g urea, 1.5 g KH2PO4, 0.8 g MgSO4·7H2O, and 22.7 g citric acid. For the fermentation, cassava, with ca. 60% moisture content, is spread in a thin layer (2-3 cm thick) on perforated trays and slid into an aerated humidified enclosure. The incubation lasts ± 65 h. The production of protein enriched cassava is 3.26 kg dry matter/m2 tray. The effects of the variation of the nutritive solution composition and the inoculum conservation period on the protein production are equally discussed.

Notes: Using the available information of fermentation biochemistry, fermentation (stoichiometric) equations are derived for anaerobic saccharolytic fermentations of butanediol and mixed acids. The equations describe the interrelations among the fermentation products, biomass, and consumed substrate (glucose). The validity of the equations is tested using a variety of batch data from the literature. The validity of the equations is expected to extend to steady-state and transient fermentations, as well. Uses, improvements, and extensions of the equations are also discussed in detail. Among others, it is shown that the equations are useful for checking the consistency of experimental data, for calculating maximal yields and selectivities for the fermentation products, and calculating the extent of utilization of the Embden-Meyerhof-Parnas pathway versus the Hexose Monophosphate pathway of glucose utilization.
