Thema: Chemie und Pharmazie , Medizin
Notizen: Summary The effect of cimetidine treatment on the pharmacokinetics and pharmacodynamics of single doses of trimazosin was studied in 6 normotensive volunteers. Co-administration of cimetidine did not significantly affect the overall magnitude of the hypotensive effect of trimazosin. However, the time profile of the blood pressure response was significantly modified particularly with attenuation of the delayed component. Co-administration of cimetidine did not alter alpha1-adrenoceptor antagonism by trimazosin. There was no significant change in the clearnace and volume of distribution of trimazosin but there was a significant reduction in the area under the concentration-time curve for the metabolite, 1-hydroxytrimazosin. The reduction in the AUC of 1-hydroxy-trimazosin corresponds in time with the attenuation of the delayed hypotensive response. This is consistent with the suggestion that the delayed hypotensive response is related to an active metabolite, probably 1-hydroxytrimazosin.

Thema: Chemie und Pharmazie , Medizin
Notizen: Summary The inhibitory effect of omeprazole on gastric acid secretion was tested in a group of patients on haemodialysis for chronic renal failure. Single 30 mg doses almost totally inhibited basal acid output on both dialysis and non-dialysis days. Plateau acid output was reduced by a mean of 77% and 90% on non-dialysis and dialysis days respectively. The absorption and pharmacokinetic profile of omeprazole were not affected by dialysis. Omeprazole was not recoverable from dialysis fluid. It is concluded that omeprazole is a potent inhibitor of gastric acid secretion in patients with chronic renal failure, and its effect is not influenced by haemodialysis.

Notizen: Summary The pharmacokinetics of dexamethasone, given at low dose, were studied in 13 patients with congenital adrenal hyperplasia (CAH) to ascertain whether kinetics differed in this inherited disorder of cortisol metabolism from those seen in healthy individuals. Changes in plasma dexamethasone concentration after intravenous bolus, measured using a simple novel radioimmunoassay, were well described by a two-compartment open model with first-order kinetics. Values for λ2: 0.206 h−1, t1/2: 3.53 h, Vc: 24.41 and f: 0.64 were similar to those previously reported for normal subjects. There were considerable interindividual differences in parameter values and Cmaxp.o. (range 22–67 nmol/l). As suppression of the hypothalamo-pituitary-adrenal axis correlates with plasma dexamethasone levels, this variability may partly explain the differing dose and dose schedule requirements necessary to achieve adequate therapeutic control in the clinical management of CAH.

    Thema: Biologie
Notizen: Summary We have determined the copy number of 21 genes in an allotetraploid and several diploid species of cotton by gel and dot blot hybridization with cloned cDNAs. The legumin A, legumin B, and all 18 unique Lea (late embryogenesis-abundant) cDNA sequences isolated from the AD allotetraploid Gossypium hirsutum are present in one copy in A, D, E, and F diploid species and in two copies in G. hirsutum. Gel blot analysis of DNAs digested with EcoRI or BamHI usually detects different sized fragments in A and D diploids. Conservation of these restriction fragment length polymorphisms in G. hirsutum allows most of these fragments to be assigned to their respective subgenomes. Furthermore, both subgenomes in G. hirsutum can be distinguished from those in the interfertile allotetraploid G. barbadense. These results show that physical mapping of both sets of chromosomes in an allotetraploid should be possible by segregation analysis.

    Thema: Biologie
Notizen: Abstract In earlier studies, only two major patterns of transcript accumulation were seen for 18 late embryogenesis abundant (Lea) gene families in cotton (Gossypium hirsutum L.) during embryogenesis and early germination. Each of these gene families probably comprises two active alloalleles. The two polypeptides encoded by seven of the Lea families can be distinguished, and analysis of their translation in vitro indicated that regulation of the homeologous transcript abundance was similar in each. In the present study, two-dimensional gel electrophoresis of polypeptides synthesized in excised embryos was employed to determine if LEA polypeptide synthesis is regulated at the translational level. The relative in vivo synthesis rate of each of the two polypeptides of 7 Lea families was compared with the relative concentration of their transcripts measured earlier by in vitro translation. For 4 families, the relative translational efficiencies of the homeologous mRNAs do not change during embryogenesis. However, there are changes of 1.5–3-fold in the other 3 families. The translation efficiencies of all transcripts of 9 of the Lea family mRNAs in vivo can be calculated from the fraction of total protein radioactivity incorporated in each LEA polypeptide family and the fractional abundance of Lea family transcripts measured by RNA dot hybridization. Lea mRNAs are found to be translated at near average rates throughout embryogenesis and early germination. These observations suggest that regulation of Lea gene expression at the translational level is minor.

    Thema: Biologie
Notizen: Summary Earlier studies found that cotton (Gossypium hirsutum L.) cotyledons contain several mRNAs which are more abundant during late embryogenesis than in mid-embryogenesis or early germination. They are here termed ‘Late embryogenesis-abundant’ mRNAs, encoded by Lea loci. Complementary DNA clones for 18 such mRNA sequences, defined at a hybridization criterion of Tm-15°C, were identified in a mature embryo cDNA library by differential cDNA hybridization. At a lower hybridization criterion, some sequence homology was found within several of these cloned Lea mRNA sequences. Each Lea mRNA sequence comprises 0.04–1.3% of mature embryo poly(A)+ mRNA, a level ten-fold to several hundred-fold higher than in young embryo or 24 h seedling poly(A)+ mRNA. Of 18 Lea mRNA sequences examined in cultured young embryos, the level of at least 13 are specifically increased by exogenous abscisic acid (ABA), several to a level near that in normal mature embryos. However, the abundance of several of the sequences does not appear to be significantly modulated by ABA. The LEA polypeptides encoded by 10 Lea mRNA sequences were identified by hybrid-arrested translation. They include most of the late embryogenesis-abundant, ABA-inducible, polypeptides previously identified. Preliminary results suggest that many of the individual Lea mRNA sequences are transcribed from 1–3 genes in each of cotton's two subgenomes.

Beschreibung: This paper presents a representative example of an enhancement in energetic ion flux associated with the International Sun-Earth Explorer 3 (ISEE 3) spacecraft's encounter with a traveling compression region (TCR). Data from the energetic particle anisotropy spectrometer (EPAS) instrument on ISEE 3 are studied, along with magnetic field data from the vector helium magnetometer. It is concluded that the ion enhancements seen are spatial in nature, thus supporting the idea that TCRs are the lobe signatures of plasmoids moving along the magnetotail, away from earth.

Beschreibung: Magnetotail observations from the ISEE 3 distant (1983) tail mission taken during the Coordinated Data Analysis Workshop 8 (CDAW 8) A and G events are investigated. The ISEE 3 magnetic field, plasma, and energetic particle measurements taken in these two plasmoids have been analyzed and compared with various equilibrium structures and propagating waves/tail oscillation modes. Results indicate general agreement with either the closed-loop (Hones, 1977) or very small pitch angle flux rope (Hughes and Sibeck, 1987; Birn et al., 1989) models of plasmoid structure and poorer agreement with other hypotheses. Calculations based upon typical plasmoid and tail parameters are presented, indicating that the J and B force associated with the disconnected lobe field lines may be sufficient to accelerate plasmoids up to the speeds observed by ISEE 3. Overall, the energy expended in accelerating the plasmoids down the tail appears comparable to that dissipated in the inner magnetosphere and ionosphere. The study produces strong evidence in favor of the plasmoid model of substorm tail dynamics.

Beschreibung: The hypothesis that the IMF penetrates plasmoids causing them to be three- rather than two-dimensional is tested by comparing observations of By within plasmoids and related tail structures to upstream IMF By data. The magnetic topologies that result from the mergings of closed plasma sheet flux tubes and open tail lobe flux tubes at a near-earth neutral line, and merging near the tail flanks are described and studied. The particle signals and isotropic electron distributions are examined. It is observed that the IMF By penetrates plasmoids and that their structure is three-dimensional. In the three-dimensional model of plasmoids the reconnected plasma sheet field lines form a magnetic flux-ropelike structure. The three-dimensional model is utilized to analyze stagnant, slowly moving and earthward moving structures.

Beschreibung: It is pointed out that magnetospheric substorms have been referred to as the 'fundamental instability' of the magnetosphere which results from coupling between the solar wind and the earth's magnetic and plasma environment. The present paper is mainly concerned with magnetic observations made by the midlatitude Air Force Geophysics Laboratory (AFGL) Magnetometer Network, the GOES 2 and 3 synchronous satellites, and the near-geosynchronous P78-2 Spacecraft Charging at High Altitudes (SCATHA) satellite. Ground-based Pi 2 and magnetic bay observations are used to detect a clear substorm onset. An idealized model of the substorm current system is utilized to relate the observations to the spatial location and temporal development of the substorm disturbance near synchronous orbit.
