    Notes: Summary Monospecific antibody directed to cysteine protease of 2-day-old rat epidermis recently characterized as being different from the proteases previously reported was produced in rabbits. By immunofluorescence microscopy and immunoperoxidase staining with an avidin-biotin-peroxidase method the protease was found to be present in the epidermis of rodents of different ages as well as that of humans, but not in the dermis. The staining in germinative cells was more intense than in cells in the superficial layers. It appeared as irregular patches in the nuclei and stained more diffusely in the cytoplasm where small granular components, strongly stained, were identified. The staining patterns in granular cells showed accumulation of the antigen in a granular form. The morphology and distribution of granules resembled those of keratohyalin-like granules in the nucleus and dense homogenous deposits in the cytoplasm. In cornified cells the reaction product was localized by the plasma membrane where concentration of the dense homogenous deposits occurred, suggesting that the cysteine protease is one component of the unique and characteristic structure of differentiated keratinocytes. In addition, the cysteine protease antigen having the same molecular weight as the epidermal enzyme was detected in liver, kidney and lung indicating a wider tissue distribution of the protease. The significance of the protease in regulation of cellular functions remains to be investigated.

    Notes: Summary The chemical nature of the thickened plasma membrane of cornified cells in stratified squamous epithelium was investigated in comparison with that in noncornified epithelium. Localizations of transglutaminase, molecular weight 92000 daltons, and detection of epidermal cysteine proteinase inhibitor were effected with a monoclonal antibody and a monospecific rabbit anti-inhibitor immunoglobulin, respectively, directed to the antigens. N-(7-dimethylamino-4-methylcoumarinyl) maleimide was used to demonstrate S-S cross-linking. In all keratinizing epithelia, the enzyme and inhibitor were deposited on membranes of granular cells. S-S bonds were formed in cornification with the appearance of electron-dense material by the inner leaflet. Both enzyme and inhibitors occurred on the corneal epithelium, but S-S linkage and the thickened plasma membrane did not form even at the last stage of maturation. On the other hand, the internal vaginal epithelium in the proestrous stage without keratinization contained the enzyme, but neither inhibitor nor S-S linkage. Both antigens and S-S bonds were detected when keratinization proceeded during estrus. The staining patterns in the epithelium near the vaginal introitus were identical to those in the skin. Cuboidal and simple epithelia exhibited none of those constituents. The findings indicated that heterogenous components contribute to modification of the plasma membrane of cornified cells, but S-S cross-linkages are associated exclusively with formation of the ultrastructurally unique membrane structure. In addition, findings suggested hormonal regulation in the chemical modification of the membrane in estrogen-sensitive internal vaginal epithelium.

    Notes: Rapid technological advancements have contributed to the increasing application of adhesives. Because of these accomplishments, because of continuing materials research and process development activities, and because of their inherent advantages, adhesives have a great potential for the future. Requirements for the future impose a severe challenge on developmental capabilities. More severe structural requirements and environmental conditions may be expected. Because of increasing complexity and costs, there are requirements for significantly higher orders of reliability and lower fabrication costs. Improvements in bonding processes and development of comparatively new techniques are desirable. Test methods require further development and standardization - especially nondestructive methods. Continuing work in collecting and reporting pertinent information, especially on properties and design parameters for adhesive bonding is helping to advance the utilization of adhesives. Current trends indicate that adhesives are progressing from the realm of “art” to science, a factor which should substantially aid in advancing the future potential for adhesives.

    Notes: We report the biocompatibility in the rat brain of a controlled-release, biodegradable polymer, the polyanhydride poly-[bis(p-carboxyphenoxy)propane-sebacic acid] copolymer (PCPP-SA) in a 20:80 formulation. The biodegradable polyanhydride can be used for drug delivery directly into the brain, circumventing the difficulties posed by the blood - brain barrier and avoiding the consequences of having to administer toxic doses systemically to reach therapeutic doses in the central nervous system. The tissue reaction in the presence of PCPP-SA was compared to that seen with other standard neurosurgical implants. Fifty-six adult Sprague-Dawley rats were assigned to one of seven groups and underwent bilateral frontal lobe implantation of PCPP-SA (42 hemispheres), Surgicel (oxidized regenerated cellulose) (35 hemispheres), or Gelfoam (absorbable gelatin sponge) (35 hemispheres). None of the animals showed any behavioral changes or neurological deficits suggestive of either systemic or localized toxicity from the biodegradable polyanhydride, all surviving to the scheduled data of sacrifice. PCPP-SA evoked a well localized inflammatory reaction, comparable to that of Surgicel, which resolved as the PCPP-SA polymer degraded over five weeks. The biodegradable polyanhydride has been shown in this study to be nontoxic and biocompatible in the rat brain, when compared to standard neurosurgical implants.

    Notes: The rate of consumption of oxygen by cotton and cottom cellulose from solutions of hypochlorite in the pH range 5-10 has been studied. Kinetic plots were obtained either by periodically titrating aliquots or by automatically recording the amount of alkali required to keep the pH constant. In the case of pure cotton cellulose the plots are linear, whereas in the case of cotton the plots consist of two parts, an initial, rapid oxidation of the impurities followed by a slower, linear oxidation of the cellulose.     Notes: Functional groups and degradation of cotton oxidized by hypochlorite at several pH values in the range 5-10 were studied. The carboxyl group content of the oxidized cottons increases, while the aldehyde and ketone group contents decrease with increase in pH. Approximately 40% of the oxidant consumed is accounted for by the functional groups at all pH values. The ratio of carboxyl, aldehyde, and ketone groups formed per scission is 1-1.5:3:3.5 at the acidic pH range and 5:0.8:0.0 at pH 10. Oxygen consumption per scission is 26 atoms, out of which 10.4 atoms are accounted for by the functional groups. The rest of the oxygen is explained by oxidation of the soluble products of the short chain material in the fiber phase and by the functional groups of the short chains with D.P. up to 37. The degradation was found to be random at the whole pH range studied, as proved by the ratio of the number-average and weight-average molecular weights and by the linearity of the plot of log [η] vs. log (Oc + Oc0) where Oc is the oxygen consumed and Oco a constant. It is suggested that the degradation and oxidation reactions occur simultaneously and, at a given pH, with similar rates, so that all carbon atoms of an anhydroglucose unit in all chains of the amorphous phase will have a similar probability of being attacked.
