    Notes: 3611 MSV, a raf-oncogene-transducing murine retrovirus, induces fibrosarcomas and erythroid hyperplasia in newborn mice after a latency of 4-8 wk. In contrast, new recombinant murine retroviruses carrying the myc oncogene (J-3, J-5 construct viruses) do not induce tumors before 〉 9 wk. A combination of both oncogenes in an infectious murine retrovirus (J-2) induces hematopoietic neoplasms in addition to less prominent fibrosarcomas and pancreatic adenocarcinoma 1-3 wk after inoculation. The hematologic neoplasms consist of immunoblastic lymphomas of T and B cell lineage and erythroblastosis. If animals were inoculated with a variant of the J-3 virus, which induces altered foci in cultures of NIH 3T3 cells, carcinoma developed in the pancreas with a 2-6 mo latency. In parallel to the synergistic action of both oncogenes on hematopoietic cells in vivo, we find that raf-oncogene-induced transformation of bone marrow cells in culture is enhanced by the addition of myc, which by itself does not transform these cells when grown in standard media. We conclude that concomitant expression of raf and myc oncogenes in hematopoietic and epithelial cells alters their respective transforming activities. The contribution of v-myc in this synergism was examined by use of a series of recombinant murine retroviruses capable of expressing the avian v-myc to study the effect of altered myc expression on hematopoietic/lymphoid cells. With either interleukin 3- or interleukin 2-dependent cell lines, introduction of the recombinant viruses abrogated the requirement for IL 3 or IL 2 for growth, and associated with this was the suppression of c-myc expression. The findings suggest that myc is a component in the signal transduction pathway for IL 3 and IL 2 and support an autoregulatory mechanism of c-myc expression. In contrast to v-myc, expression of v-raf primary lymphoid hematopoietic cells has an immortilizing function without abrogating the requirement for IL 3 for growth. This suggests that v-raf and v-myc affect different components of growth regulation, as, for example, commitment (v-myc ) and cell cycle progression (v-raf).

    Notes: The process of vitellogenesis in oviparous vertebrates embraces a number of integrated physiological and developmental phenomena. Since this process is readily subjected to experimental control in Xenopus laevis, we have been able to undertake a preliminary survey of the pertinent mechanisms operating in this animal.The information at hand is discussed as it relates to (a) the hormonal (estrogen) induction of yolk protein synthesis by the liver, (b) the characterization of the yolk protein produced and its relationship to the proteins of the mature egg, (c) the transport of the yolk protein to the ovary and its specific uptake by the developing oocyte, and (d) the transformation of the accumulated protein into crystalline yolk platelets.

    Notes: An effort has been made to ascertain the site of calcium uptake in rabbit psoas muscle by electron microscopy. Following a brief glycerol extraction, small groups of fibers were exposed in a suitable electrolyte environment to calcium and ATP. Conditions were such that calcium was accumulated as an insoluble oxalate salt. Conventional preparatory procedures for electron microscopy were modified so that the calcium oxalate deposits were retained in situ in ultrathin sections. These were found localized in the longitudinal component of the sarcoplasmic reticulum including the terminal dilatations which form the outer elements of the triads. The possible participation of the intermediate element of the triad in the uptake of calcium could not be determined. The results are regarded as direct evidence that the sarcoplasmic reticulum is intimately concerned with the uptake and release of calcium associated with the contraction-relaxation cycle.

    Notes: Rabbit psoas muscle fibers, extracted in 50% (w/v) glycerol for 20 hours, concentrate up to 300 μmol calcium per gm protein when placed in a suitable electrolyte environment at pH 6.5 containing ATP and 47Ca in the form of a Ca/EGTA metal buffer system. This property is retained at [Ca++] down to 2 × 10-8 M, the lowest studied. The kinetics of Ca pickup are consistent with a mathematical model based on diffusion of Ca as the rate-limiting factor. The pickup is dependent upon ATP and is increased by inclusion of CP or PEP. It is promoted by oxalate, fluoride, phosphate and pyrophosphate, which share the property of forming relatively insoluble calcium salts. Ability to concentrate calcium disappears on prolonged extraction in glycerol and after preincubation with desoxycholate.These properties are similar to those previously described for a granular fraction of muscle homogenates, and it is concluded that both are probably attributable to functional remnants of sarcoplasmic reticulum.

    Notes: Optimal imaging of complex structures requires proper alignment relative to the optic axis of the electron microscope. This is especially important for high-voltage and intermediatevoltage microscopes, which form an in-focus image throughout the entire thickness of the object. As a result, structures at different specimen heights form overlapping and confused images that severely curtail the usefulness of these instruments.The work described here provides a generalized, flexible method for optimizing specimen orientation and eliminating or limiting image overlap by means of a commonly used double-tilt stage. Analysis of the motion about the two axes provides accurate tilting for any azimuthal direction whether or not it corresponds to a mechanical axis of the stage. An object can be positioned to minimize image overlap, to record stereopairs for any parallax axis, and to record three-dimensional data sets by the conical collection geometry.Images of muscle paracrystals are shown after tilting about an axis perpendicular to a symmetry direction. The tilted image displays higher-order symmetry, which is altered by changes of one degree. Precision double-tilting for optimizing stereopairs is shown for a desmosome recorded using different parallax axes and pretilts. A tomographic conical data-collection scheme is demonstrated by imaging a microtubule axoneme for a specific cone half-angle and arbitrary azimuthal angles.

    Notes: Application of electron microscopy in a wide variety of fields of investigation has placed ever-expanding demands on the various components of the instrument. In situ specimen manipulation is one such demand and can often be critical to the success of an experiment. Control of specimen orientation is the most common manipulation, but control of a variety of other physical and chemical parameters may also be important. Temperature, gaseous and/or liquid environment, and mechanical operations are examples. Control and variation of these parameters in a small device (occupying a few cm3) operated in a strong magnetic field inside a vacuum system is often a considerable challenge. This must also be done at extreme stability: at least as good as the resolution limit of the microscope. Optimization of stage performance is too often sacrificed for optical performance or vice versa. The next generation of objective lenses and specimen stages are being designed in concert: an approach which should lead to an improved in situ laboratory, observed with optimum optics.
