    Notes: Fibronectin and other cell attachment proteins provide molecular models for beginning to unravel the complex interactions of the cell surface with the extracellular matrix. This area has been reviewed in considerable detail previously [1-10]. Our brief review will therefore be selective rather than comprehensive, and it will focus on some recent generalizations about this class of proteins, as well as on recent advances in the molecular analysis of the functions of these proteins and their receptors. we shall also present various popular or provocative hypotheses and speculations about future work in the field.

    Notes: We have characterized a 140-kDa glycoprotein complex purified by a monoclonal antibody and implicated in cell adhesion to the extracellular molecule fibronectin. Three major polypeptide components were purified by monoclonal antibody JG22E, which had apparent molecular weights of 155,000 (band 1), 135,000 (band 2), and 120,000 (band 3). In two-dimensional gel electrophoresis, each subunit migrated as either a broad band or a series of spots at acidic isoelectric points. After treatment with neuraminidase, the spots became focused around pH 6.2 (band 1), pH 5.6 (band 2), and pH 5.3 (band 3). These three major bands were compared by two-dimensional peptide mapping in a series of pairwise combinations and were found to be distinct proteins. In sucrose gradients, these proteins co-migrated as a complex sedimenting at approximately 8.4 S either before or after affinity purification, whereas separated subunits migrated at 4.7 to 5.8 S. Amino acid analysis revealed no detectable hydroxyproline and a composition characterized by a substantial number of cysteine residues compared to the average protein. Our results suggest that a noncovalent complex of structurally distinct glycoproteins is involved in adhesive interactions of fibronectin with cells.

    Notes: When demembranted axonemes of Chlamydomonas were reactivated with Mg ATP, the proportion of motile axonemes was significantly increased by the preence of either phosphodiesterase (PDE) or protein inhibitor of cAMP-dependent kinase (PKI). The effect of PDE was cancelled by the addition of cAMP. These findings strongly suggest that the axoneme samples have endogenous cAMP, which can reduce the proportion of motile axonemes via phosphorylation. This inhibitory effect of cAMP on Chlamydomonas axonemes is opposite to its stimulatory effect on the axonemal motility in other organisms so far reported. PKI or PDE activated the motility motility either in the absence of Ca2+, when the axonemes beat with an asymmetric waveform, or in 10-5M Ca2+, when the axonemes beat with a symmetric waveform. This cAMP-dependent regulation of motility was observed with the axonemes from which detergent-soluble material had been removed, indicating that the proteins responsible for the regulation still remained in the axonemes. Preliminary in vitro phosphorylation stdies have implicated two polypetides as candidates for the target protein of cAMP-dependent protein kinase: one with a molecular weight of 270 kD and the other with a much larger molecular weight.

    Notes: Several steps in the synthesis in vitro of infectious bacteriophage RNA can now be described. The reaction catalyzed by the Qβ RNA polymerase is known to involve several components, including the enzyme, host cell factors, Qβ RNA template, and the strand complementary to the Qβ RNA. The interaction of these components and the mechanims of the reaction appears to be considerably more complex than was proposed in earlier models.

    Notes: We characterized human umbilical vein (HUV) endothelial cells as to cell growth and prostacyclin production to get a better understanding of the properties of endothelial cells. Endothelial cell growth supplement (ECGS) and basic fibroblast growth factor (FGF) stimulated HUV endothelial cell growth. Heparin further enhanced the cell growth stimulated by ECGS, but not the cell growth stimulated by FGF or in the absence of these growth factors. In the presence of ECGS, the prostacyclin-producing capacity of the cells was inhibited by heparin. However, in the presence of FGF of in the absence of growth factors, heparin did not inhibit prostacyclin production. Therefore, it is likely that there is a specific correlation between heparin and growth factors for endothelial cells in the blood vessel to maintain nonthrombogenicity properly. Heparin-treated cultures may not be suitable for some examinations of prostacyclin production by vascular endothelial cells.

    Notes: Crystalline 2,5-distyrylpyrazine (DSP) was found to polymerize into a linear high polymer with extremely high crystallinity by the action of sunlight or ultraviolet light. Based on this discovery, photopolymerizability of the related diolefins having the general formula, trans,trans-Ar′—CH=CH—Ar—CH=CH—Ar′ was surveyed. As a result, 1,4-bis[β-pyridyl-(2)-vinyl]benzene (P2VB) was found to polymerize in the same way as DSP. The polymers as polymerized, showed a high melting temperature (〉330°C) and were soluble only in strong acids. The structure of the polymers was investigated by means of several customary methods involving infrared and NMR spectrum studies. It was concluded that both poly-DSP and poly-P2VB consist of a linear structure having a recurring cyclobutane ring in the main chain. General properties of this novel polymer were studied.

    Notes: Linear polyhydrazides have been obtained successfully by heating diphenyl dicarboxylate with 1,6-dihydrazino hexane. The structure of the polymers was established by the N-deuteration method. The resulting polymers have many remarkable characteristics, such as high water-absorption ratio (20-100%), basic character (pKa = 3.15), the ability to form a metal complex, and the power to initiate vinyl polymerization as a cocatalyst with ferric ion.

    Notes: Four possible structural models with novel steric character are proposed for poly-2,5-distyrylpyrazine: two of them are extended chains and the others are kinked coils. In order to prove the molecular characteristic of the new polymer, a procedure to solubilize the polymer into some organic solvents was developed. It was recognized by viscometry that no significant degradation of the polymer does occur during the preparation. The lack of the shear rate dependence of the dilute solution viscosities would seem to indicate that the polymer molecule in solution can be regarded as a chain, not extended linearly, but kinked randomly. The crystallization of the polymer proceeds only in the presence of a sufficient amount of the solvent which enables the polymer chains to move and crystallize. These imply that the possible structure of the polymer is constructed of 1,2- or 1,3-cis type cyclobutane linkages with restricted rotation.

    Notes: The solid-state photopolymerization of phenylene diacrylic acid (PDA) and its derivatives was studied as an application of solid-state photodimerization of cinnamic acid to photopolymerization of corresponding bifunctional molecule which has two cinnamic units in a single molecule. p- and m-PDA, and their esters and amides were prepared and investigated with respect to their photopolymerizability. Many of them have been found to polymerize into linear high polymers with the cyclobutane rings in the main chain on irradiation by ultraviolet or visible light. The polymerization process, the structure of the polymers, and their general properties were investigated in several ways. All the polymers are very similar to poly-2,5-distyrylpyrazine and poly-1,4-bis(β-pyridyl-2-vinyl) benzene with respect to their polymerization behavior, polymer structure, and some polymer properties: these polymers are soluble in a limited number of solvents, they have a high melting point and an extremely high crystallinity. On the basis of chemical behavior of poly-PDA and its phenyl ester the possible steric configurations of these polymers are discussed. It is demonstrated for the PDA series that solid-state dimerization can be generally extended to solid-state photopolymerization of the compound having two dimerizable units in a single molecule, although the crystal structure renders polymerization impossible in certain cases.
