Notes: Abstract The occurrence of endogenous plasmid DNA in Rhizobium meliloti is reported. R. meliloti strains carrying RP 4 were used to work out a suitable plasmid isolation method. By sarcosyl lysis RP 4 plasmid DNA as well as cryptic plasmid DNA was isolated. Cryptic plasmid DNA possesses a buoyant density of 1.717 g cm-3 and shows a heterogeneity in contour length. Large plasmid molecules of 130 μm length were detected in the electron microscope. We also found that RP4 is able to restore the nitrogen fixation capacity of an ineffective R. meliloti strain.

Notes: The main ionization methods in a mass spectrometer for isotope ratio determinations of the elements are discussed in this review. These methods are thermal ionization, spark source, electron impact, inductively coupled plasma and field desorption. As concerns thermal ionization, electron impact and field desorption, a survey of the possibilities of isotope analyses in the periodic table of the elements is given. Besides kinetic studies, trace element determination by isotope dilution technique is the main application for isotope ratio measurements of the elements. The definitive method, isotope dilution mass spectrometry, is discussed as a potential tool for achieving accurate and precise trace analyses. Using field desorption mass spectrometry, one example of calcium kinetics in man and one example of thallium trace determination in an animal tissue are given. Other metal trace analyses with the isotope dilution technique are presented for biological and medical samples using positive thermal ionization mass spectrometry. Negative thermal ions are formed for the mass spectrometric analysis of non-metals and non-metal compounds in food samples, e.g. for iodine and nitrate in milk powder. Preliminary results with the isotope dilution technique are presented for a new quadrupole thermal ionization mass spectrometer which is a low-cost instrument and can be easily handled.

Description / Table of Contents: Chlorine Isotope Separation in a Heterogeneous Electrolyte System Dependence on the Concentration and the Counter Ion of a Fluoride SolutionIn a heterogeneous electrolyte system of a strongly basic anion exchanger and a concentrated fluoride solution the dependence of the chlorine isotope separation on the concentration and the counter ion of the fluoride solution is determined by chromatographic experiments. The results show the greatest separation effect of the chlorine isotopes so far achieved, using ion exchange reactions. 37Cl- is enriched in the solution phase, whereas 35Cl- is enriched in the ion exchanger. With increasing fluoride concentration the isotope separation effect decreases. The strong dependence of the isotope separation on the counter ion of the fluoride solution is shown using CsF and KF respectively.

Notes: Im heterogenen Elektrolytsystem stark basischer Anionenaustauscher/konzentrierte Fluoridlösung wird die Abhängigkeit der Chlorisotopenseparation von Konzentration und Gegenion des Fremdelektrolyten in chromatographischen Versuchen bestimmt. Die Ergebnisse zeigen den bisher größten bekannten Elementartrenneffekt der Chlorisotope bei Ionenaustauschreaktionen, wobei sich das 37Cl- - gegenüber dem 35Cl--Ion in der Lösung des Systems anreichert. Mit steigender Fluoridkonzentration nimmt der Isotopentrenneffekt ab. Am Beispiel einer CsF - gegenüber einer KF - Lösung wird die starke Abhängigkeit der Isotopenseparation vom Gegenion des Fremdelektrolyten aufgezeigt.

As minor components 7,8,7′,8′-tetrahydro-ψ, ψ-carotene (ζ-carotene), β, β-carotene (β-carotene), and tentatively, a 2,3,2′(or 3′)-trihydroxy-β, β-caroten-4-one and a 2,3,2′(or 3′)-trihydroxy-β, β-carotene were identified.
