    Notes: An unknown, ninhydrin-positive substance detected on paper chromatograms of the endogenous metabolites of mixed rumen ciliate protozoa was isolated and purified by column chromatography with ‘Dowex’ 50-X8 resin and identified as 2-aminobutanoic acid (α-amino-n-butyric acid) on the basis of elementary analysis, mass spectrometry, paper chromatography, infrared spectrometry, and melting point.

    Notes: Hard hydrogenated amorphous carbon (a-C:H) films were coated on amorphous silicon photoreceptors by the plasma decomposition of acetylene (C2H2). This overlayer proved to be a much better protective layer for the adsorption of external species compared to an a-SiNx or a-Si1−xCx overcoating layer, and the image degradation induced by such adsorption was considerably reduced. The highly protective properties were attributed to the hydrophobic nature of C–Hn (n=1–3) and C–C bonds contained in the a-C:H film. The optical properties of the a-C:H films are also described.

    Notes: Abstract In many shrimp farms in the Kyushu and Chugoku areas of Japan, the so-called mid-gut gland cloudy disease of kuruma shrimp larvae(Penaeus japonicus) has occurred since 1971. The pathological changes associated with this baculoviral mid-gut gland necrosis (BMN) are extensive cellular necrosis, collapse of mid-gut gland cells, nuclear hypertrophy and finally karyorrhexis. Electron microscopic examination revealed the presence of virions and virogenic stages in the affected nuclei. Average length and diameter of the virions detected was 310 and 72 nm, respectively; nucleocapsids were 250 nm in size. Virions enclosing 2 nucleocapsids within a single envelope were rarely found. The spirally arranged capsomeres were at an angle of 37 to 38° to a horizontal line meeting at right angles with the long axis of the virion. Infectivity trials resulted in high mortality of healthy mysis and juveniles (2nd post-larval stage). Juveniles at the 9th post-larval stage showed no mortality, although they could be infected easily by the agent. Hypertrophied nuclei in squashed and stained preparations of the affected gland cells can be considered to be of reliable presumptive diagnostic character, and fluorescent antibody staining can be employed to confirm the diagnosis of BMN.

    Notes: Summary Histamine N-methyltransferase (S-adenosylmethionine: histamine N-methyltransferase, E.C. 2.1.1.8) was purified to homogeneity from rat kidney, and antibody was raised against it in guinea pigs. The antibody immunoprecipitated histamine N-methyltransferase. Immunofluorescent histochemical studies with anti-histamine N-methyltransferase antibody as the first antibody and goat antiguinea pig IgG conjugated with fluorescein isothiocyanate as the second, showed the presence of immunoreactive structures in the proximal tubules of rat kidney. The brain showed no immunoreaction with the antibody.

    Notes: Summary Changes in the spatial relationship between actin filaments and microtubules during the differentiation of tracheary elements (TEs) was investigated by a double staining technique in isolatedZinnia mesophyll cells. Before thickening of the secondary wall began to occur, the actin filaments and microtubules were oriented parallel to the long axis of the cell. Reticulate bundles of microtubules and aggregates of actin filaments emerged beneath the plasma membrane almost simultaneously, immediately before the start of the deposition of the secondary wall. The aggregates of actin filaments were observed exclusively between the microtubule bundles. Subsequently, the aggregates of actin filaments extended preferentially in the direction transverse to the long axis of the cell, and the arrays of bundles of microtubules which were still present between the aggregates of actin filaments became transversely aligned. The deposition of the secondary walls then took place along the transversely aligned bundles of microtubules. Disruption of actin filaments by cytochalasin B produced TEs with longitudinal bands of secondary wall, along which bundles of microtubules were seen, while TEs produced in the absence of cytochalasin B had transverse bands of secondary wall. These results indicate that actin filaments play an important role in the change in the orientation of arrays of microtubules from longitudinal to transverse. Disruption of microtubules by colchicine resulted in dispersal of the regularly arranged aggregates of actin filaments, but did not inhibit the formation of the aggregates itself, suggesting that microtubules are involved in maintaining the arrangement of actin filaments but are not involved in inducing the formation of the regularly arranged aggregates of actin filaments. These findings demonstrate that actin filaments cooperate with microtubules in controlling the site of deposition of the secondary wall in developing TEs.

    Notes: Summary The level of tubulin in cultured matureZinnia mesophyll cells increases to between 4 and 5 times its initial value when DNA synthesis occurs. The rapid increase in the level of tubulin requires the presence of auxin and cytokinin, as does the induction of DNA synthesis. Inhibitors of DNA synthesis suppress the rapid increase in the level of tubulin. These results imply that the rapid increase in the level of tubulin is dependent on the occurrence of DNA synthesis,i.e., on the reinitiation of the cell cycle. The presence of microtubules is not important for either the increase in the level of tubulin or for the induction of DNA synthesis, because both the increase in the level of tubulin and DNA synthesis occur even when microtubules are depolymerized by colchicine.

    Notes: Continuous cellulase production by Trichoderma viride QM 9123, immobilized in 6 mm diameter, spherical, stainless steel biomass support particles, has been achieved using a medium containing glucose as the main carbon source. Experiments were carried out in a 10-L spouted bed fermentor. In this type of reactor-recycled broth is used to create a jet at the base of a bed of particles, causing the particles to spout and circulate. During the circulation, particles pass through a region of high shear near the jet inlet. This effectively prevents a buildup of excess biomass and thus enables steady-state conditions to be achieved during continuous operation. Continuous production of cellulase was achieved at significantly higher yield and productivity than in conventional systems. At a dilution rate of 0.15 h-1 (nominal washout rate for freely suspended cells is 0.012 h-1), the yield of cellulase on glucose was 31% higher than that measured during batch operation, while the volumetric productivity (31.5 FPA U/L· h) was 53% greater than in the batch system. The specific cellulase productivity of the immobilized cells was more than 3 times that of freely suspended cells, showing that diffusional limitations can be beneficial. This offers significant opportunity for the further development of biomass support particles and associated bioreactors.
