    Notizen: Abstract The mouse B-cell lymphoma WEHI 279.1 is a tumor which synthesizes both membrane and secreted immunoglobulin M (IgM). We have immunoselected variants which fail to express the membrane form (mIgM−); the most frequently isolated phenotype is a complete loss of both membrane expression and synthesis of the μ heavy chain within the cells. We have chosen four of these mIgM − mutants for detailed molecular investigation. One of these has suffered a large deletion which covers the region of chromosome 12 containing the expressed μ gene, but three have no detectable changes in the DNA arrangement of the μ gene. All of the mutants, including the deletion mutant, synthesize 10–30% of the wild-type level of cytoplasmic μ RNA; however, none is the appropriate size for membrane μ (μm) or secreted μ (μs) message. Based on our studies of the deletion mutant, which retains its nonproductively arranged allele, at least some of these RNAs may be ‘sterile’ transcripts from the nonproductively arranged allele. However, if all of these mRNAs derive from the other allele, they represent a substantial elevation of these sterile messages relative to the wild-type level. Furthermore, the three nondeletion mutants transcribe μ RNA at a level indistinguishable from the wild type. It is likely that their defects lie in the stability, processing, or transport of the μ RNA within the nucleus. Somatic cell hybrids between P3X and the IgM− variants produced mostly mIgM− hybrids. However, a few mIgM+ hybrids were produced, suggesting that the μ− defects may be partly complemented by the P3X fusion partner.

The asymmetric unit consists of two crystallographically independent molecules of tetraphenylphthalic anhydride (TPPA). Crowding from four adjacent phenyl substituents produces large average displacements of the substituents from the central ring (av. ~0.08 Å, max. 0.17 Å) and distortions in the central aromatic ring. The torsion angles relating the plane of the substituent phenyl rings to the central ring plane (av. ∼62°) is consistent with other polyphenylated aromatic structures.

    Notizen: Abstract Postcollapse lavas of the Infiernito caldera grade stratigraphically upward from nearly aphyric, high-silica rhyolite (76% SiO2) to highly prophyritic trachyte (62% SiO2). Plagioclase, clinopyroxene, orthopyroxene, magnetite, ilmenite, and apatite occur as phenocrysts throughout the sequence. Sanidine, biotite, and zircon are present in rocks with more than about 67% SiO2. Major and trace elements show continuous variations from 62 to 76% SiO2. Modeling supports fractional crystallization of the observed phenocrysts as the dominant process in generating the chemical variation. Temperatures calculated from coexisting feldspars, pyroxenes, and Fe-Ti oxides agree and indicate crystallization from slightly more than 1100° C in the most mafic trachyte to 800° C in high-silica rhyolite. The compositional zonation probably arose through crystallization against the chilled margin of the magma chamber and consequent rise of more evolved and therefore less dense liquid. Mineral compositions vary regularly with rock composition, but also suggest minor mixing and assimilation of wall rock or fluids derived from wall rock. Mixing between liquids of slightly different compositions is indicated by different compositions of individual pyroxene phenocrysts in single samples. Liquid-solid mixing is indicated by mineral compositions of glomerocrysts and some phenocrysts that apparently crystallized in generally more evolved liquids at lower temperature and higher oxygen fugacity than represented by the rocks in which they now reside. Glomerocrysts probably crystallized against the chilled margin of the magma chamber and were torn from the wall as the liquid rose during progressive stages of eruption. Assimilation is indicated by rise of oxygen fugacity relative to a buffer from more mafic to more silicic rocks. Calculation of density and viscosity from the compositional and mineralogical data indicates that the magma chamber was stably stratified; lower temperature but more evolved, thus less dense, rhyolite overlay higher temperature, less evolved, and therefore more dense, progressively more mafic liquids. The continuity in rock and mineral compositions and calculated temperature, viscosity, and density indicate that compositional gradation in the magma chamber was smoothly continuous; any compositional gaps must have been no greater than about 2% SiO2.

    Notizen: Summary Fluorescent dyes are commonly used to study permeable (gap) junctions, but only rarely have quantitative values for junctional dye permeability been determined. In the present study, junctional permeance (PA, i.e., the product of the junctional permeability coefficient,P, times the junctional area,A) to Lucifer Yellow CH (LY) has been obtained for pairs of Novikoff hepatoma cells. Dye was microinjected into one cell and the subsequent transfer monitored by a SIT camera and recorded on video tape. The intensities of fluorescence in the injected and “recipient” cell were measured using a Digisector (Microworks) digitizing board and an Apple II Plus computer to analyze the video records. These changes in intensity, along with an estimate of volume of the spherical cells, were used to calculate the junctional permeance (PA) of cell pairs according to Fick's diffusion equation. Junctional permeances show considerable variation ranging from 0.08×10−11 to 27.0×10−11 cm3/sec. Using the meanPA and a previous estimate of the mean number of junctional channels per interface in the Novikoff cultures, a value for diffusion coefficient of LY through gap junctions is calculated to be about 1.4×10−6 cm2/sec. There is a general proportionality between meanPA and cell volume for hepatoma cell pairs of a certain size range. Such a relationship between cell volume and junctional capacity suggests one source of variation ofPA. Other possible sources, e.g., related to position in the cell cycle, are discussed.

    Notizen: Abstract The γ-irradiation of malic acid in aqueous solutions was studied under initially oxygenated and oxygen-free conditions in an attempt to determine the possible interconversion of malic acid into other carboxylic acids, specifically those associated with Krebs cycle. The effect of dose on product formation of the system was investigated. Gas-liquid chromatography combined with mass spectrometry was used as the principal means of identification of the non-volatile products. Thin layer chromotography and direct probe mass spectroscopy were also employed. The findings show that a variety of carboxylic acids are formed, with malonic and succinic acids in greatest abundance. These products have all been identified as being formed in the γ-irradiation of acetic acid, suggesting a common intermediary. Since these molecules fit into a metabolic cycle, it is strongly suggestive that prebiotic pathways provided the basis for biological systems.
