    Description: The ability of a regulatory protein to sense the integrity of the bacterial flagellar structure was investigated. In response to a defective hook-basal body complex, the anti-sigma 28 FlgM protein inhibits flagellin transcription. In cells with a functional hook-basal body complex, the flagellin genes are transcribed normally and the FlgM protein is expelled into the growth medium. In strains with a defective hook-basal body structure, FlgM is absent from the media. The presence of flagellin protein in the media is substantially reduced in strains carrying a FlgM-LacZ protein fusion, suggesting that the fusion is blocking the flagellar export apparatus.     Notes: A collective Thomson scattering diagnostic is being implemented at JET to diagnose the velocity distribution and density of alpha particles and energetic ions. This diagnostic consists of a number of major subsystems including: a high-power 140-GHz gyrotron, a 32-channel 12-GHz bandwidth heterodyne receiver, efficient corrugated waveguide systems, variable polarizers, and in vacuum beam steering mirrors. Proper calibration of these subsystems for signal level losses, positioning, and polarization is imperative for the successful operation of the diagnostic and to facilitate the interpretation of the scattered data. A low-power Gunn oscillator was used to verify the polarization rotation induced by the seven miter bends in the receiver transmission line. The full bandwidth receiver front end losses, including the sapphire tokamak vacuum window, were initially calibrated by using the hot tokamak vessel wall as a blackbody source at approximately 310 °C. Background ECE measurements will be presented.

    Notes: It is noted that the hypothesis of independent random complex elements for the off diagonal blocks (say, M2 and M3) of the transfer matrix describing conductance in a mesoscopic wire allows the eigenvalue distribution of the matrix product M3°M3 (or M2°M2) to be computed exactly. Exact expressions, in terms of double Wronskian and Toeplitz determinants, are derived for the distribution of the smallest and second smallest eigenvalue of this and similar random matrix products. © 1994 American Institute of Physics.

    Notes: : A model is developed for real-time operation of an irrigation reservoir with the objective of maximizing the value of multiple crop yields during a growing season. The model employs monthly additive and product forms of crop yield functions for dry matter and grain crops, respectively. The resulting nonlinear optimization model uses a log transform to reduce nonlinearities in the model. An application of the proposed model is compared to a common operating rule used in simulation models. The proposed model results were better in terms of net benefits from crop yields. The model uses GAMS (General Algebraic Modeling System) language. It requires an IBM-compatible microcomputer and is suitable for use by a reservoir manager.

    Notes: There has been a recent revival of interest in one of the most abundant Escherichia coli proteins, H1 (also called H-NS). This protein was first identified many years ago as a major component of the bacterial nucleoid, and has been characterized biochemically by several groups. However, no clear function for the protein emerged from these studies. Our thinking has been transformed by recent findings which complement the biochemistry with genetic data. Several mutations, selected over many years by virtue of their diverse effects on gene expression, have turned out to be allelic and to fall within the structural gene for H1. Bringing together the genetics and the biochemistry has demonstrated that the whole is worth more than the sum of the parts! These findings have far-reaching implications for the mechanisms by which gene expression is regulated and also, perhaps, for the control of bacterial virulence.

    Notes: Summary Studies in Mytilus edulis have indicated that immunoregulatory activities comoparable to those in vertebrates also exist in invertebrates. Mytilus immunocytes resemble cells of the vertebrate monocyte/macrophage lineage and are activated by similar substances. We searched for differential effects of opioids on these cells in comparison with those of lipopolysaccharide (LPS), in order to determine if different subpopulations of immunoactive hemocytes are involved. We showed that Mytilus immunocytes respond to LPS in a fashion similar to that in vertebrate granulocytes by flattening, and increasing in cellular perimeter and mobility, that LPS administered in vivo results in a lowering of the number of free hemocytes that can be obtained from the animal, and that distinct immunoactive cell populations seem to exist since apparently different subsets of cells react when exposed to LPS or opioids and the opioid antagonist naloxone.

    Notes: Abstract A 2.1 kb Bam H1 DNA fragment encoding a pectate lyase (PL) enzyme was isolated from an Erwinia carotovora subsp. atroseptica (Eca) cosmid library. The fragment was labeled with 32P-CTP and hybridized to total DNA digests from selected bacteria which included plant-invasive as well as plant associative organisms. The pel gene probe hybridized to E. carotovora subsp. carotovora (Ecc) DNA under all conditions tested. Hybridization to DNAs from Agrobacterium tumefaciens and Pseudomonas marginalis was observed at low stringency conditions (45°C). No hybridization was observed between the pel gene probe and six other DNA samples.

HIV gp120 selectively reduces the glutamate-induced inward current and the acetylcholine-induced outward current in specific and identifiedAplysia neurons without affecting dopamine (DA)- and serotonin (5-HT)-induced responses. 2. gp120 specifically decreases DA levels without significantly altering norepinephrine and 5-HT levels inMytilus pedal ganglia. 3. The gp120-associated decrease in DA levels inMytilus is dose dependent and exhibits a threshold level. 4. The alteration ofin vitro DA levels is specific for gp120 since anti-gp120 blocks the effect. 5. gp120 and its effects appear to be stable due to the duration of treatment and the failure of secondary effects to materialize following antibody treatment.

    Notes: Abstract When tritium is used as a fusion fuel in the JET tokamak it will be necessary to use double, interpumped windows on the vacuum vessel. Methods are discussed for reducing the frequency dependence of the transmission of double windows to be used for various millimetre wave diagnostics, over the appropriate frequency ranges.
