    Description: Sediment whole-round cores from a dedicated hole (798B) were obtained for detailed microbiological analysis, down to 518 m below the seafloor (mbsf). These sediments have characteristic bacterial profiles in the top 6 mbsf, with high but rapidly decreasing bacterial populations (total and dividing bacteria, and concentrations of different types of viable heterotrophic bacteria) and potential bacterial activities. Rates of thymidine incorporation into bacterial DNA and anaerobic sulfate reduction are high in the surface sediments and decrease rapidly down to 3 mbsf. Methanogenesis from CO2/H2 peaks below the maximum in sulfate reduction and although it decreases markedly down the core, is present at low rates at all but one depth. Consistent with these activities is the removal of pore-water sulfate, methane gas production, and accumulation of reduced sulfide species. Rates of decrease in bacterial populations slow down below 6 mbsf, and there are some distinct increases in bacterial populations and activities that continue over considerable depth intervals. These include a large and significant increase in total heterotrophic bacteria below 375 mbsf, which corresponds to an increase in the total bacterial population, bacterial viability, a small increase in potential rates of sulfate reduction, and the presence of thermogenic methane and other gases. Bacterial distributions seem to be controlled by the availability of terminal electron acceptors (e.g., sulfate), the bioavailability of organic carbon (which may be related to the dark/light bands within the sediment), and biological and geothermal methane production. Significant bacterial populations are present even in the deepest samples (518 mbsf) and hence it seems likely that bacteria may continue to be present and active much deeper than the sediments studied here. These results confirm and extend our previous results of bacterial activity within deep sediments of the Peru Margin from Leg 112, and to our knowledge this is the first comprehensive report of the presence of active bacterial populations from the sediment surface to in excess of 500 mbsf and sediments 〉 4 m.y. old.

    Notes: Abstract— The plastic deformation of thin-walled cylinders has been experimentally examined for the loading conditions of ±1% axial strain with hoop stresses of approximately 0, 1/4, 1/2 and 3/4 of the initial uniaxial yield stress.Two materials similar to those used in the pipework of PWR nuclear plant in the U.K. have been tested, namely 304S11 stainless steel and En6 low-carbon steel. The results of the tests were to be compared with the allowable stresses and deformations specified in the ASME Boiler and Pressure Vessel Code, Section III. The code specifies that a prescribed combination of primary stresses must not exceed 1.5Sm, where Sm is a stress value defined for each material.The results indicate that the limit of 1.5Sm is excessively low for both materials and that in particular, the stainless steel could tolerate 5Sm. Although the En6 steel is more prone to ratchetting than the stainless steel, the results suggest that it too could tolerate a higher primary stress than the code allows. Both materials are shown to satisfy the proposed ASME ratchet strain limit of 5% hoop strain after 10 cycles of ±1% axial strain range, for any value of internal pressure.

    Notes: Summary A study was conducted to evaluate differences in rangeland grasshopper communities over environmental gradients in Gallatin Valley, Montana, USA. The concept of habitat type (Daubenmire 1966) was used as a basis for discriminating between groupings of patches based on vegetation. A total of 39 patches were selected that represented five recognized grassland habitat types (Mueggler and Stewart 1980), as well as two disturbed types (replanting within a known habitat type). Repeated sampling in 1988 of both the insect and plant communities yielded a total of 40 grasshopper (19 664 individuals) and 97 plant species. Detrended Correspondence Analysis (DCA) indicated that patch classifications based on presence and percent cover of plants were appropriate and showed good between-group (habitat type) separation for patches along gradients of precipitation/elevation and plant community complexity. Results from undisturbed habitats showed that plant and grasshopper species composition changed over observed environmental gradients and suggested that habitat type influenced not only species presence, but also relative abundance. Discussion is presented that relates results with patch-use and core and satellite species paradigms.

    Notes: Summary Two terpenic constituents, E,E,-α-farnesene and E-β-farnesene, were found to be elevated in dominant male urine when compared to subordinate or control males. These two urinary compounds were absent in the bladder urine of males; however, they were the most prominent constituents of the preputial gland's aliquots. The results of a two-choice preference test, conducted on ICR/Alb subordinate males, gave a strong indication that these two terpenic constituents introduced into the previously attractive stimulus significantly discouraged prolonged investigations by male mice. The compounds, whether present in the urine matrix or water, rendered the stimulus with a quality behaviorally similar to the urine of dominant males. It appears that they may be synonymous with the previously described aversion signal produced by dominant males. We suggest that these compounds may play a wide-ranging role in the territorial marking behavior of male mice.

    Notes: Abstract Experimental data of the radial incorporation of labeled cholesterol [14C-4] into the artery wall is regressed against a mathematical model that predicts macromolecular transport in this biological system. Data is obtained using excised canine carotid arteries that are perfusedin vitro under pulsatile hemodynamic conditions for 2 hr. Vessels are exposed to either normotensive hemodynamics, hypertensive hemodynamics, or simulations in which the rate of flow or vessel compliance is deliberately altered. Several arteries are studied under normotensive conditions following balloon catheter deendothelialization. Transmural concentration profiles of [14C-4] activity are determined by microcryotomy of longitudinal sections of perfused vessels. Nonlinear Marquardt regression on 12 experimental cases yields parameter estimates of effective diffusivity,D and solute filtration velocity,V. Results of this experimental investigation support our hypothesis that hemodynamics and the endothelial lining influence wall flux in intact vessels. Exposure to altered (vs normotensive) hemodynamics is associated with increased incorporation of labeled cholesterol. A similar observation is made for deendothelialized vessels (e.g. a greater accumulation of label and a rise in convective flux). Based upon our companion measurements of vessel wall forces and endothelial cellular morphology accompanying hemodynamic simulations, we suggest that hemodynamically induced alterations to endothelial structures lead to the increased permeability, convection and incorporation that we observe in this work.

    Notes: A neonatal incubator has been custom modified to enable measurement of initial platelet retention on biomaterials in vitro under clinically relevant hemodynamic conditions. To calibrate this device, platelet retention on several materials having microconduit geometry (0.7-1.0 mm i.d.) has been measured after perfusion with citrated whole blood (containing 111 Indium-labelled platelets) at a shear rate of 312 s-1, 37°C, and 80 cm H2O transmural pressure. The relative reactivity of these materials toward platelets was: glass 〈 Fibrinogen(Fg)-coatedglass 〈 Fg-coated polyethylene 〈 polyethylene ∼ = Expanded polytetrafluoroethylene. Interindividual variation is relatively large (coefficient of variation = 35.5 ± 9.3%), but comparison to intraindividual controls reduces the variability to 14.8 ± 10.3%, a level which is suitable for economical testing of platelet retention to biomaterials in the presence or absence of drugs. This approach may have particular value in the study of the mechanism of platelet interactions with artificial microvascular grafts under perfusion conditions which are relevant to the first moments of flow, when initial platelet deposition occurs.

    Notes: Abstract Previous studies of the rate constants for the elementary steps of ATP hydrolysis by the soluble and membrane-bound forms of beef heart mitochondrial F1 supported the proposal that ATP is formed in high-affinity catalytic sites of the enzyme with little or no change in free energy and that the major requirement for energy in oxidative phosphorylation is for the release of product ATP. The affinity of the membrane-bound enzyme for ATP during NADH oxidation was calculated from the ratio of the rate constants for the forward binding step (k +1) and the reverse dissociation step (k −1).k −1 was accelerated several orders of magnitude by NADH oxidation. In the presence of NADH and ADP an additional enhancement ofk −1 was observed. These energy-dependent dissociations of ATP were sensitive to the uncoupler FCCP.k +1 was affected little by NADH oxidation. The dissociation constant (K d ATP) increased many orders of magnitude during the transition from nonenergized to energized states.
