    Notes: Abstract Transcriptional and translational fusions were made between the reading frame coding for β-D-glucuronidase and sequences of either a constitutively expressed rice gene (GOS2) involved in initiation of translation or a light-inducible rice gene (GOS5). The transient expression of the fusions was studied via particle bombardment of seedling tissues of rice, perennial ryegrass and barley. Furthermore, the results of transient and stable expression were compared for cell suspensions of four rice varieties, one barley variety and one perennial ryegrass variety. TheGOS2-gusA fusions were active in all three monocots studied. Best results were obtained for a construct having both a transcriptional and a translational fusion as well as intron and exon sequences (PORCEHyg). The level of GUS activity was in the range of activities as obtained by the 35S CaMV promoter transcriptionally fused togusA. ThegusA fusion with the light-inducible gene (GOS5) was active in green seedling tissues of all monocots studied. Also a weak expression compared to theGOS2 constructs was found in stably transformed rice callus. ThegusA fusions with the mannopine synthase promoters 1′ and 2′ of the TR-DNA were transiently expressed at lower levels in cell suspensions than PORCEHyg. For stably transformed rice callus the expression of theGOS2-gusA fusion often decreased during prolonged subculture. This decrease in GUS activity and the various GUS-staining phenotypes of transgenic calli are explained by the presence of different cell types in the suspensions used and in the calli. It is presumed that the nature of the cells and their relative contribution in the calli change drastically upon further subculture.

    Notes: Abstract The cell-type-specific expression of three rice genes, GOS2, GOS5 and GOS9, was studied by mRNA in situ hybridization. Previous northern blot analysis revealed that these genes were constitutive, green tissue-specific and root-specific, respectively. In this study, GOS2 transcripts were observed in all leaf cell types. In roots, a temporal and spatial expression pattern was noticed. Higher mRNA levels were observed in lateral roots, especially in parenchymal cells of the vascular cylinder. Expression of GOS5 was mainly found in chloroplast-containing cells. For GOS9, significant levels of signal were observed in root and leaf sections.

    Notes: Summary Both naturally occuring and carcinogen-induced tumors display not only point mutations in cellular oncogenes but also more complex changes in cellular oncogenes and other cellular genes. For this and other reasons, it seems likely that DNA damage in mammalian cells can induce alterations in gene expression that may have both short and long term consequences in the target cell. The purpose of this review is to summarize current available information on inducible responses to UV-irradiation and other mediators of DNA damage in mammalian cells, and to provide some working hypotheses. We have divided these responses into three time frames, immediate (0–12 hours), early (12–48) and late (beyond 48 hours). Immediate responses include the action of DNA repair enzymes, some of which are induced as a consequence of DNA damage, and transient inhibition of DNA synthesis. Within the past few years considerable evidence has accumulated that during this immediate period there is increased expression of certain cellular oncogenes, proteases and proteins whose functions remain to be identified. It is of interest that the expression of some of these genes is also induced by certain growth factors, tumor promoters and heat shock. Alterations in gene expression during the subsequent “early” period (12–48 hrs.) have not been studied in detail, but it is during this period that one can detect increased replication of several types of viruses in cells that harbor these viruses. We have examined in detail the induction of asynchronous polyoma DNA replication (APR) in a rat fibroblast cell line carrying integrated copies of this DNA. We have obtained evidence that UV-irradiation of these cells leads to the synthesis of a 40 kd protein, within the first 1–24 hrs after irradiation, that binds to a specific sequence TGACAACA in the regulatory region of polyoma DNA. We suggest that this protein acts together with other proteins to induce APR and that this serves as a useful model for understanding the mechanisms responsible for amplification of cellular genes, a phenomenon often seen in malignant tumors. Finally, we discuss how the events occurring during the immediate and early periods following DNA damage might lead to late effects in the target cell that are stable and contribute to the genotype and phenotype of some of the progeny of these cells that are destined to become tumor cells.

    Description: The debate on the history of the Martian atmosphere continues without resolution. Much hinges on a satisfactory solution to the problem of what liquid was responsible for the extensive development of canyons, channels, fretted terrain, and similar features. The 1960s and 1970s saw much debate about the roles of various fluids, namely water or brine, liquid CO2, or H2O sources in CO2 clathrate (CC), or magma. The last was discounted long ago, CC is largely ignored, and the most recent look at CO2 was by ourselves, utilizing the phase diagrams for the CO2-H2O system. In this article, we will further investigate this dilemma.

    Description: An improved AOTF-based imaging spectrometer that offers several advantages over prior art AOTF imaging spectrometers is presented. The ability to electronically set the bandpass wavelength provides observational flexibility. Various improvements in optical architecture provide simplified magnification variability, improved image resolution and light throughput efficiency and reduced sensitivity to ambient light. Two embodiments of the invention are: (1) operation in the visible/near-infrared domain of wavelength range 0.48 to 0.76 microns; and (2) infrared configuration which operates in the wavelength range of 1.2 to 2.5 microns.

    Description: An acousto-optic tunable filter (AOTF) is employed to generate a display by driving the AOTF with a RF electrical signal comprising modulated red, green, and blue video scan line signals and scanning the AOTF with a linearly polarized, pulsed light beam, resulting in encoding of color video columns (scan lines) of an input video image into vertical columns of the AOTF output beam. The AOTF is illuminated periodically as each acoustically-encoded scan line fills the cell aperture of the AOTF. A polarizing beam splitter removes the unused first order beam component of the AOTF output and, if desired, overlays a real world scene on the output plane. Resolutions as high as 30,000 lines are possible, providing holographic display capability.

    Description: The addition of fine-grained magnetite to mafic silicate spectra can impart not only an overall blue slope, but also lower overall reflectance and band intensities. The reflectance spectra of the CO and CV magnetite-bearing carbonaceous chondrites are noted to exhibit many of these features; the low band depths of these meteorites suggest that an additional dark, neutral phase, such as ordered carbon, is present. Carbon + mafic silicate spectra possess a red overall slope at low amorphous carbon concentrations. The parent bodies of some of the darkest meteorites should exhibit spectral features attributable to mafic silicates.

    Description: In a search for diagnostic spectral parameters which can be used to distinguish different materials on the surface of asteroids and to provide information on the detection limits for mafic silicates, the 0.3- to 2.6-micron reflectance spectra of meteoritic enstatite (nearly pure MgSiO3), iron meteorite metal, magnetite, and amorphous carbon as well as various mixtures of these materials with mafic silicates were examined. Results are presented on the dependence of the spectral detectability of mafic silicates associated with metal, carbon, and magnetite on the particle sizes of the phases, their chemistries, crystal structures, and abundances. It is shown that the observational data for a representative M-class asteroid, (16) Psyche, are largely consistent with a fine-grained metal-rich surface assemblage, whereas data for the E-class asteroid (44) Nysa indicate that its surface is composed of fine-grained material similar to enstatite achondrites, with a small amount of material comparable to the chondritic inclusions found in the Cumberland Falls aubrite.
