    Notes: A subcellular fraction containing fragments of endogenous microtubules stabilized in 50% glycerol was separated by diferential centrifugation of rat brain homogenates. The pellets were suspended in glycerol-deficient media, and microtubule depolymerization was monitored by measuring the decrease of sedimentable tubulin. Concomitantly, the number and size of microlubules in the suspensions were followed via electron microscopy. Depolymerization was accompanied by a proportional decrease in the number of microtubules, whereas the average size did not change significantly. After approximately 20 min, a subpopulation of microtubules became stable and did not suffer further depolymerization. These results indicate that upon dilution some microtubules completely depolymerize, whereas others remain stable in the glycerol-deficient medium. The degree of depolymerization depended on both the volume of the resuspension media and on the final glycerol concentration. The results suggest that the depolymerization of the remaining microtubules is prevented by stabilizing factors released from depolymerizing microtubules. Tubulin dimers are not one of these factors, since depolymerization was not altered by the addition ofcolchicine or by changing the concentration of free tubulin in the medium.

    Notes: The theory of optimal control is used to analyze the performance of a batch PMMA (poly(methylmethacrylate)) polymerization reactor. Optimal dynamic profiles of temperature, initiator feed rate, and initiator concentration in the reactor are generated in order for specified conversion of monomer, number molecular weight, and polydispersity of the final product to be obtained. Based on tae physical limitations of the system, some restrictions are imposed to the control variable variations. The application of Pontryagin's Minimum Principle leads to a mixed initial/final value problem, which is solved by discretizing the control law in the state-space, with the usage of a first order holder. A gradient method is used to minimize the Hamiltonian. The simulation results show that the gel-effect equation and restrictions imposed on the system are of primary importance to the analysis of the reactor performance.

    Notes: Development of neurons in the area triangularis of Gallotia galloti was investigated in Golgi-impregnated brain tissue. Four major neuronal types present in adults were found to originate from two migratory neuroblast types, which were followed from embryonic stage S.32. One type has a thick main medial process, whereas the second type has a long main lateral process. As they migrate toward the periphery of the nucleus, morphological characteristics of maturation appear, including growth cones, filopodia, and outgrowth of axons. Neuroblasts with a main lateral process differentiate into two immature neuronal types, bipolars and pyramidals, observed at S.33 and thereafter. The neuroblasts with a main medial process undergo some somatic translocation through a transitory tangential shaft. Then they develop into monopolar immature forms with a long varicose medial, process, appearing from S.36. onward. Immature bipolar neurons do not experience great changes in their dendritic arborization during development to the adult stage, but pyramidals and monopolars undergo a rapid development of the dendritic tree after S.36. By S.38 archetypes of adult neuronal forms are established. Hairlike appendages first appear on neurons at S.36 They decrease suddenly in S.38 and then proliferate in S.39 when spines first appear. Around the time of hatching, the hairlike appendages begin to disappear and spines become established. Reduction of spines occurs after hatching and continues to the adult stage. Possible influences of several external factors on neuronal maturation are discussed.

    Notes: In a Golgi study of the area triangularis (AT), a rostral nucleus of the ventral thalamus of Gallotia galloti, we have identified four major neuronal types on the basis of their morphological characteristics: medium-sized fusiforms with two processes, medium-sized fusiforms with three or four processes, small bipolars, and small and medium-sized multipolars. These neurons are characterized by a simple morphology and radial arrangement. Cell size varies from small to medium, and all axons project laterally. These characteristics distinguish AT neurons from those of neighboring nuclei. In addition, we found some evidence of differential topographic distribution of each neuronal type within the nucleus. Medium-sized fusiform neurons with two processes are located in the most ventral part, where they constitute the ventral nuclear limit. Small multipolar neurons prevail in the dorsal and ventromedial parts, and in the rest of the nucleus medium-sized neurons, including both fusiform with three or four processes and multipolar types, are normally found. Finally, we discuss a putative homology of the reptilian AT with a part of the mammalian zona incerta.

    Notes: Using several in vitro motility assays, we found that motility driven by the microtubule (MT) motors, kinesin and cytoplasmic dynein, could be inhibited by MAP2 but not by tau protein or the MT-binding proteolytic fragment of MAP2.In MT gliding assays, even the presence of one MAP2 molecule per sixty-nine tubulin dimers caused an inhibition of about 75% of MT motility at low concentrations of both motors. The percent inhibition of motility decreased with increasing concentration of either motor, suggesting that the inhibition was the result of competition for access to the MT surface. The decrease in the number of moving MTs with MAP2 was correlated with an increase in the frequency of release of moving MTs from the motor-coated glass. In assays of in vitro vesicular organelle motility and formation of ER networks, the presence of MAP2 inhibited small vesicle movements and to a lesser extent ER network formation.To determine if competition for specific sites on the MT or coating of the MT surface inhibited motility, we used tau protein and the chymotryptic MT-binding fragments of MAP2 to coat MTs. No inhibition was observed and there was even an increase in the number of attached and moving MTs in the gliding assay with tau-coated MTs. Because MAP2, tau and the chymotryptic MT-binding fragments of MAP2 bind to the same domain on tubulin, masking of the MT surface sites does not appear responsible for the inhibition of motility by MAP2. Rather, we suggest that the sidearm of MAP2 interfered with the interaction of motors with MTs and caused a dramatic increase in the rate of MT release. In vivo, MAP2 could play a major role in the generation of cellular polarity even at substoichiometric levels by inhibiting transport on microtubules in specific domains of the cytoplasm. © 1993 Wiley-Liss, Inc.

    Notes: The content of vinculin, a cytoplasmic protein found in focal contacts and cell-cell junctions, was increased in BALB/c 3T3 cells by gene transfection. The vinculin expressed from the full length chicken cDNA, incorporated into focal contacts and its pattern was identical to that of the endogenous protein. Cells stably expressing vinculin by 20% over the endogenous level had altered locomotory properties. In these cells, the ability to migrate into a wound formed in a confluent monolayer and the locomotion of individual cells were drastically reduced. The results provide direct evidence that cell locomotion can be regulated by modulating vinculin expression. © 1992 Wiley-Liss, Inc.

    Notes: The modified van der Waals theory of interfaces (gradient theory) is used with the equations of state of Peng-Robinson and Mohanty-Davis to predict surface tensions of various nonpolar, quadrupolar, slightly polar and polar substances as a function of temperature. The influence parameter of the gradient theory is estimated as 0.785ab2/3 in terms of the attractive and repulsive van der Waals parameters using the Lennard-Jones potential. The predictions made with the Mohanty-Davis equation of state agree better with experimental data than those made with Peng-Robinson, even for nonpolar fluids.

    Notes: A membrane-assisted solvent extraction (MASX) system coupled to a membrane-assisted distillation stripping (MADS) system for use in decontaminating ground-water is evaluated. Volatile organic compounds (VOCs) in the groundwater are extracted in the MASX unit using a sunflower oil solvent. In the MADS unit, VOCs are stripped from the sunflower oil, which is recycled to the MASX unit. Thermodynamic data for the sunflower oil-water-VOCs system were measured. The results were used along with published membrane mass-transfer data to design MASX and MADS modules.
