    Notes: Novel 1,3,4-thiadiazole-containing polyazomethines and copolyazomethines were synthesized by the solution polycondensation, in m-cresol at 25°C, of aromatic dialdehydes, isophthalaldehyde and terephthalaldehyde, with 2,5-bis (m-aminophenyl)-1,3,4-thiadiazole (BMAT) and with BMAT and aromatic diamines, bis (4-aminophenyl) ether and 1,5-diaminonaphthalene, respectively. These polymers were tan yellow to yellow in color and had reduced viscosities up to 0.32 dL/g in concentrated sulfuric acid and electric conductivity as high as 10-9-10-11 S cm-1 at 25°C. All the polymers were insoluble in common organic solvents but dissolved completely in concentrated sulfuric acid and formic acid. However, they were readily hydrolyzed in concentrated sulfuric acid. X-ray diffraction diagrams showed that the crystallinity of polyazomethines were low, but copolyazomethines were highly crystalline. These azomethine polymers are highly thermally and thermooxidativelly stable and exhibited no appreciable decomposition up to 400°C in both air and nitrogen atmospheres. Doping with iodine dramatically raised the conductivity and produced the dark brown- to completely black-colored semiconductive polymers with a maximum conductivity of the order of 10-5 S cm-1. Electronic spectra of the undoped polymers indicated a large bathochromic shift of the π-π* absorption band (310 nm) due to C=N bonds of BMAT. This result suggests that π-electrons of the polymers are extensively delocalized along the main chain.

    Notes: Anodic polarization measurements of several orthopaedic implant alloys - AISI 316L stainless steel, Co—Cr—Mo alloy (ASTM F-75), nickel-titanium (NiTi) alloy, commercial pure titanium, and Ti—6Al—4V - were carried out in Ringer's solution containing 1% bovine serum albumin (BSA) and in simple Ringer's solution. Constituent metals in the medium were analyzed by inductively coupled plasma emission spectrophotometry after measurements. Pitting potentials and total anodic current of former two alloys were constant in the two mediums and not affected by the addition of bovine serum albumin. However, the amount of dissolved metals from pure nickel and AISI 316L stainless steel were increased by the addition of BSA; dissolved metals from Co—Cr—Mo alloy were decreased by it.

    Notes: A simple and effective technique of electrofusion of the mouse myeloma cells and lymphocytes induced by AC pulses was investigated. Instead of pearl chain formation, we used a mild centrifugation of high-density cell suspensions to enhance the cell-cell contacts. The optimal frequency of the AC field used for cell fusion was 10 kHz. We confirmed the production of monoclonal antibodies against human serum albumin (HSA) by the hybridoma obtained by this method. Under the conditions used in the present work, the efficiency of hybridoma formation is several times higher than those obtained by the chemical method using polyethylene glycol (PEG). Hybridoma colonies were detected in as high as 95% of the wells and 33% of them proved positive against HSA.

    Notes: In animal models, grafts derived from several different tissues, principally fetal substantia nigra and adrenal medulla from young adults, have been found to be effective in alleviating some of the manifestations of lesions of the substantia nigra. It has been suggested that these grafts function by diffusely secreting dopamine, by exerting trophic effects on the host brain, or by producing a new innervation of the host corpus striatum. Evidence for each of these modes of action is briefly reviewed. Several brain tissue transplantation techniques have been described. Each of these techniques has significant limitations in animal models. The significance of these limitations for human application is described, and possibilities for improving the efficacy of brain tissue transplantation in animal models and for human application are discussed.

    Notes: The transformation of certain cells reduces the requirement of extracellular Ca2+ for growth. The SV-40 transformed human lung fibroblasts, WI-38 VA13, require less Ca2+ than normal WI-38 cells. Spreading area of normal cells decreases when cultured in 10 μM Ca2+ medium. Intracellular calcium concentration ([Ca2+]i), of the normal and transformed cells cultured in 10μM and 2 mM Ca2+ media was measured by the fluorescence microscope technique using fura-2 as a probe. The [Ca2+], is measured in the resting state and during mobilization by serum or bradykinin stimulation. The lowering of extracellular calcium concentration results in a decrease in the resting state [Ca2+],i of both normal and transformed cells. Although the total decrease in [Ca2+]i is the same for both cell, the rate of decrease is much faster in normal cells than in transformed cells. Low extracellular Ca2+ reduces the number of cells responsive to the serum or bradykinin stimulation and decreases the peak [Ca2+]i value in both cells. In addition, we investigated, using BCECF as a fluorecent probe, the intracellular pH (pHi) of normal and transformed cells maintained at low and normal Ca2+. The low Ca2+ condition makes pHi acidic in normal cells but not in transformed cells. The acidification of the normal cell is accompanied by a decrease in the spreading area of the cells. The decrease of the cell attacment, followed by the reduced spreading area, induced the acidic pHi. These results suggest that the reduced Ca2+ requirement of transformed cells for growth is related to the mechanism of pHi regulation rather than Ca2+ homeostasis and, possibly, to the anchorage-independent growth, which is a unique feature of transformed cells. © 1993 Wiley-Liss, Inc.

    Notes: The dipole moments of small protein molecules were determined experimentally in order to validate the calculated dipole moments by previous investigators. We found that the agreements are satisfactory for some proteins. There are, however, many proteins for which the agreement is less than satisfactory. In order to find the cause of the disagreement, the dipole moments of these proteins were recalculated using the Brookhaven Protein Data Bank. We calculated the dipole moment due to fixed surface charges and the bond moments of all the carbonyl groups in main chain and side chains. The calculation consists of the mean moments and their mean square fluctuations. In addition, we investigated the effect of electrostatic interactions between charged sites for several proteins. These results show that incorporation of the interactions does not affect substantially the calculated dipole moments. The rms fluctuation of the dipole moment is found to be small but not negligible. In conclusion, recalculated dipole moments are in good agreement with the observed values. © 1993 John Wiley & Sons, Inc.

    Notes: Neurotoxic peptides from venoms of scorpions and honey bees exhibit a consensus pattern in the two disulfide bridgings related to the sequence portions Cys-X-Cys and Cys-X-X-X-Cys. A revised three-dimensional structure of charybdotoxin, as determined by two-dimensional nmr spectroscopy, confirms that the consensus cystine dislocation generates in all these toxins a common structural element, i.e., the cystine-stabilized α-helical (CSH) motif, which may be correlated with their common ion channel blocking activity.

    Notes: Alkaline phosphodiesterase I from cultured fibroblasts from patients with neurofibromatosis was partially purified and characterized following extraction with Triton X-100, and fractionation with high-performance liquid chromatography. Some properties were compared with the enzyme extracted from normal-appearing fibroblasts. The isoelectric points of both the tumour and normal-appearing cell enzymes were 6·0. The enzyme required Zn2+ for its activity, was heat labile, and nicked superhelical covalently closed circular φX174 DNA. The activity was inhibited by GTP, DTT and EDTA. The native molecular weight of alkaline phosphodiesterase I was determined to be 430 000. No differences were found in properties of the tumour-derived and normal cell enzymes. On purification it was observed that the peak pattern of enzyme activity corresponded to that of 125 kDa protein, which was more abundant upon SDS-PAGE analysis in tumour cells than in normal cells. The most active fraction of isoelectric focusing, which was performed using disulfide cross-linked polyacrylamide gel, was used to produce an antibody. The bands of 125, 60 and 40 kDa were immuno-stained in tumour cell preparation. These results indicate that alkaline phosphodiesterase I, of which the molecular weight is probably 125 kDa, is over-expressed in tumour-derived fibroblasts from neurofibromatosis patients.
