    Notes: Abstract Antimetabolite-resistant and auxotrophic mutants of the hyperthermophilic bacterium Thermotoga neapolitana were isolated to provide strains with genetic backgrounds amenable to genetic analyses. Norleucine, azaleucine, 4-nitropyridine-N-oxide, and 3-amino-1, 2, 4-triazole did not affect growth, while 5-fluorouracil (5 μg/ml), 5-methyltryptophan (250μg/ml), 6-azauracil (100 μg/ml), and 4-fluorophenylalanine (30 μg/ml) inhibited growth at the indicated minimum inhibitory concentrations. The effect of 5-fluorouracil was analyzed and found to be bacteriostatic. These inhibitors were used to select spontaneously arising resistant mutants. In addition, auxotrophic mutants requiring leucine, tryptophan, adenine, and histidine were isolated following mutagenesis with ethyl methanesulfonate. Six other auxotrophs with undefined growth requirements were also isolated. These strains will be useful for the development of genetic methods for T. neapolitana.

    Notes: Abstract The aim of this paper was the screening of the variability of growth patterns, biomass and tropane alkaloid production of 500 hairy root lines ofDatura stramonium. Data on the long term stability in alkaloid production of these lines for more than 5 years are also provided. In an effort to obtain high alkaloid-producing root clones, it is demonstrated that systematic selection is necessary. Comparisons are made, mainly concerning alkaloid production and its stability, with normal root cultures initiated from the same mother plants when necessary. Hairy root cultures were found to have a hyoscyamine and scopolamine bioproductivity of 2 orders of magnitude higher than mother plants.

    Notes: Abstract Two year old, transformed root cultures of Catharanthus roseus accumulate ajmalicine and catharanthine (0.57 and 0.36 mg g-1 DW, or 7.0 and 3.0 mg l-1, respectively). Changes in the concentration of the medium components, as well as the addition of hydrolytic enzymes and biotic elicitors, were used as strategies to increase these alkaloid yields. Regarding the components of the medium, the results obtained, when sucrose was raised from 3 to 4.5%, are noteworthy. The nitrogen source induced differential responses in the individual alkaloid yields. No net change in the alkaloid content was observed either with changes in the concentration of vitamins or macro-and micronutrients. Though the root culture only shows a limited response to elicitors, Aspergillus treatment and the use of macerozyme increased the accumulation of ajmalicine selectively, while the addition of methyl jasmonate increased the yield of both alkaloids.

    Notes: We have studied the effects of several different macronutrients on the kinetic behaviour of rat renal glucose 6-phosphate dehydrogenase (G6PDH) and 6-phosphogluconate dehydrogenase (6PGDH). Rats were meal-fed with high-carbohydrate/low-protein, high-protein/low-carbohydrate and high-fat diets. High-protein increased renal G6PDH and 6PDGH activities by 66 per cent and 70 per cent respectively, without significantly changing the Km values of either and each Hexose monophosphate dehydrogenase activity increased steadily, reaching a significant difference on day 4. A rise in carbohydrate or fat in the diets, produced no significant change in either the activity or the kinetic parameters, Vmax and Km of the two dehydrogenases. In addition, the administration of a high-protein diet for 8 days significantly increased both the pentose phosphate pathway flux (92·6 per cent) and the kidney weight (35 per cent), whereas no significant changes in these parameters were found when the animals were treated with the other diets. Our results suggest that an increase in the levels of dietary protein induces a rise in the intracellular levels of these enzymes. The possible role of this metabolic pathway in the kidneys under these nutritional conditions is also discussed.
