    Notes: Summary Heat shock and ethanol stress of brewing yeast strains resulted in the induction of a set of proteins referred to as heat shock proteins (HSPs). At least six strongly induced HSPs were identified in a lager brewing strain and four HSPs in an ale brewing strain. Four of these HSPs with molecular masses of approximately 70, 38, 26 and 23 kDa were also identified in two laboratory strains ofSaccharomyces cerevisiae. The appearance of HSPs correlated with increased survival of strains at elevated temperatures and high concentrations of ethanol. These results suggest that HSPs may play a role in the ethanol and thermotolerance of yeasts. The properties of these proteins and membrane fatty acids in relation to heat and ethanol shock are being investigated.

    Notes: Summary Growth of yeast cells on glucose resulted in complete inactivation of maltose transport and repression of the high affinity glucose transport system. When the cells were grown on maltose or subjected to substrate starvation, an increase in glucose and maltose transport was observed in both brewing and non-brewing yeast strains. The concentration of glucose employed in the growth medium was also observed to affect sugar transport activity. The higher the glucose concentration, the more pronounced the repressive effect. In addition, the time of growth of yeast on glucose or maltose also intermining the rate of sugar transport. These results are consistent with the repressive effect of glucose on the high affinity glucose and maltose transport systems.

    Notes: Abstract A Cayley tree model of idiotypic networks that includes both B cell and antibody dynamics is formulated and analysed. As in models with B cells only, localized states exist in the network with limited numbers of activated clones surrounded by virgin or near-virgin clones. The existence and stability of these localized network states are explored as a function of model parameters. As in previous models that have included antibody, the stability of immune and tolerant localized states are shown to depend on the ratio of antibody to B cell lifetimes as well as the rate of antibody complex removal. As model parameters are varied, localized steady-states can break down via two routes: dynamically, into chaotic attractors, or structurally into percolation attractors. For a given set of parameters percolation and chaotic attractors can coexist with localized attractors, and thus there do not exist clear cut boundaries in parameter space that separate regions of localized attractors from regions of percolation and chaotic attractors. Stable limit cycles, which are frequent in the two-clone antibody B cell (AB) model, are only observed in highly connected networks. Also found in highly connected networks are localized chaotic attractors. As in experiments by Lundkvistet al. (1989.Proc. natn. Acad. Sci. U.S.A. 86, 5074–5078), injection ofAb 1 antibodies into a system operating in the chaotic regime can cause a cessation of fluctuations ofAb 1 andAb 2 antibodies, a phenomenon already observed in the two-clone AB model. Interestingly, chaotic fluctuations continue at higher levels of the tree, a phenomenon observed by Lundkvistet al. but not accounted for previously.

    Notes: Summary Human IL-4 (hIL-4) has been cloned from a human T cell line based on its homology to the murine IL-4 cDNA sequence [36]. We have compared cytoplasmic and extra-cytoplasmic expression of this basic protein inEscherichia coli using various combinations of promoters, replicons and host strains. Strains producing a cytoplasmic product were most successful at heterologous protein expression, producing up to 500 mg/l of an inactive aggregated form of the protein. The biological activity of the protein could be restored by refolding the protein with guanidine hydrochloride and glutathione giving a specific activity identical to that of IL-4 derived from CHO cell lines stably transformed with an hIL-4 expression plasmid. Strains designed to secrete human IL-4 into the periplasmic space produced far less protein (approximately 5 mg/l). However, a significant fraction of this protein was detected in the culture medium. This fraction appeared to be soluble after ultracentrifugation, and demonstrated high specific activity without refolding. Leakage of heterologous protein into the culture medium may be a viable way to recover biologically active products without relying on the denaturation and refolding in vitro that can, at times, yield incorrectly folded gene product.

    Notes: Summary Copolymers of methyl acrylate (MA) with an electron-donor comonomer (N-vinylcarbazole) (NVC) are mixed with copolymers of methyl methacrylate (MMA) with a electron-acceptor comonomer (2-(3,5-dinitrobenzoyl) oxyethyl methacrylate) (DNBM) such that 1:1 mol ratios of NVC:DNBM are present at all times. The blends obtained are phase separated up to an average of 27 mol% donor/acceptor structural unit in the copolymer. Above 27 mol% “perturbation”, poly(MA) and poly(MMA) become compatible.

    Notes: Abstract This investigation examined the effectiveness of three training procedures (elaborative rehearsal, rehearsal-plus, and behavioral rehearsal) in producing children's acquisition and maintenance of fire safety skills, as well as knowledge of fire safety skills. In addition, reduction of fire-related fears was targeted. Fifty-two randomly assigned second- and third-grade children served as subjects. The children were assessed before, immediately following, and three months after their training. The three training groups demonstrated significantly greater behavioral performance gains than an untrained control group at post-test. However, treatment gains did not persist at follow-up. Regarding knowledge, the elaborative rehearsal group reported significantly more knowledge of skills than rehearsal-plus, behavioral rehearsal, and untrained control groups at post-test. At follow-up, elaborative rehearsal and rehearsal-plus groups reported significantly more knowledge of skills than behavioral rehearsal and untrained control groups. Finally, no differences were found among groups concerning fear at post-test and follow-up.
