    Notes: . The application of an immunocytochemical method to identify precystic stages and to analyse the encystment kinetics, by using a polyclonal antiserum against isolated cyst walls from the ciliate Colpoda inflata, is reported for the first time. Three different precystic phases were chosen on the basis of morphological changes and degree of cyst wall formation. By using this procedure a better identification of mature resting cysts with regard to precystic cells or young cysts is provided. An average consensus encystment kinetics of C. inflata, by using an accumulated class frequency analysis, is reported.

    Notes: Nuclear and cortical phenomena during dividing and resting cyst formation of Colpoda inflata are described. Cell division forms a cyst and produces two or four tomites. In each tomite, the right oral field results from the proliferation of the anterior extreme of a single kinety, and the left oral field results from the proliferation of four, five, or six somatic kineties. After macronuclear division, each macronuclear mass undergoes a chromatinic extrusion process. During resting cyst formation, the oral infraciliature of the vegetative cell is resorbed. The somatic kineties dispose in a radial way and some pairs of kinetosomes disappear. As in cell division, there is an extrusion process. From these results we conclude that the resting cysts of Colpoda inflata cannot be included in any group of the previous classifications for hypotrich resting cysts. Thus, we propose a new additional group to Walker and Maugel's classification called PKR (partial-kinetosome-resorbing) cysts.

    Notes: Additive genetic variance in large-seeded common beans (Phaseolus vulgaris L.), mostly of Andean South American origin, was higher than nonadditive variance for all traits. Heritability was 0.43 ± 0.19 for yield, 0.49 ± 0.20 for pods/m2, 0.63 ± 0.21 for seeds/m2, 0.76 ± 0.23 for 100-seed weight, and 0.55 ± 0.22 for days to maturity. Expected gain from selection (expressed in percentage of the population mean) was 9.2 for yield, 12.8 for pods/m2, 21.0 for seeds/m2, 17.6 for 100-seed weight, and 4.6 for days to maturity.

    Notes: Two populations of common bean, Phaseolus vulgaris L., were used to make three independent location-specific selections at Palmira, Quilichao, and Popayán, Colombia. Also, three across-location selections were made by alternating generations between Palmira and Popayan, between Quilichao and Popayán, and among Palmira, Quilichao, and Popayán. The seven highest yielding lines selected from each environment and population along with 14 parents and two checks were evaluated for seed yield, seed weight, maturity, and reaction to an-thracnose, angular leaf spot, and common bacterial blight. Mean squares due to evaluation and selection environments, selected lines, and their interactions were significant for all traits. Mean yields of selected lines were higher than the mean of parents in both populations. Among location-specific selections, lines yielded higher at their respective selection site. Moreover, selections made at Popayán were higher yielding than those made at Quilichao or Palmira when tested across locations and years. Use of Popayán, a relatively wetter and cooler site, either alone or in combination with other locations resulted in higher yield, higher seed weight, and higher resistance to anthracnose. On the average, location-specific selections were lower yielding than the across-location selections. Among across-location selections, those selected between Palmira and Popayán were the highest yielding in both populations. Selections at Palmira and across all three locations resulted in greater stability.

    Notes: Stomatal regulation of transpiration was studied in hedgerow coffee (Coffea arabica L.) at different stages of canopy development encompassing a range of leaf area indices (L) from 0·7 to 6·7. Stomatal (gs) and crown (gc) conductance attained maximum values early during the day and then declined as both leaf-to-bulk air water vapour mole fraction difference (Va) and photosynthetically active photon flux density (I) continued to increase. Covariation of environmental variables during the day, particularly V, I, and wind speed (u), obscured stomatal responses to individual variables. This also caused diurnal hysteresis in the relationship between gc and individual variables. Normalization of gs and gc by I removed the hysteresis and revealed a strong stomatal response to humidity. At the crown scale, transpiration (E) increased linearly with net radiation (Rn) and seemed to increase with increasing wind speed. Increasing wind speed imposed higher leaf interior to leaf surface water vapour mole fraction differences (Vs) at given levels of Va. However, strong relationships between declining gc and E and increasing wind speed were obtained when gc and E were normalized by I and Rn, respectively, without invoking additional potential interactions involving temperature or CO2 concentration at the leaf surface. Apparent stomatal responses to wind were thus at least partially a reflection of the stomatal response to humidity.

    Notes: Melon yellows disease is a new disease of melon which has become very important in several areas of protected melon crops in the south-east of Spain. It is spreading to other regions of the Mediterranean Coast. On first studies a viral agent was suspected; however, there were no experimental data to support such a hypothesis. In 1988 it was demonstrated that the disease was transmitted by Trialeurodes vaporariorum.The disease was found to be associated with the presence of long flexuous viral particles typical of Closterovirus Group II. A preliminary method for purification of the virus has been developed and an antiserum has been prepared.

    Notes: The Escherichia coli treA gene encodes an osmotically inducible periplasmic trehalase. A strain carrying a treA–lacZ transcriptional fusion was constructed. The β-galactosidase activity produced in this strain growing exponentially in a medium of high osmotic pressure was 10-fold higher than that produced in a medium of low osmotic pressure, demonstrating that treA transcription is osmotically inducible. treA transcriptional induction depends neither on the presence of trehalase itself nor on the synthesis of cytoplasmic trehalose which occurs in response to osmotic stress in wild-type E. coli strains. The treA promoter was identified by S1 nuclease protection. Deletion analysis demonstrated that sequences sufficient for the osmotic induction lie downstream from nucleotide –40 with respect to the transcription start. Transcription initiation at treAp required the presence of a functional σ70 subunit of RNA polymerase. treA expression was increased in the presence of a mutation in osmZ, which was previously identified as leading to a partially constitutive expression of the osmotically inducible proU operon.

    Notes: A number of mutants lacking nitrate reductase (Nas−) or nitrite reductase (Nis−) activities have been isolated and characterized. An operon including two new genes (nasA and nasB) has been defined and cloned from an Azotobacter vinelandii gene bank. nasA encodes for nitrite reductase apoenzyme, whereas nasB is specific for nitrate reductase activity. Nitrate reductase exerts a regulatory effect on nasAB.
