    Notes: The need for alternatives to autogenous bone grafts is widely recognized. This study compared the torsional strength of canine femora 1 year after grafting with one of three forms of a collagen/hydroxyapatite/tricalcium phosphate bone grafting material (COLLAGRAFT15), autogenous bone, or no graft. The groups were compared to each other and to the unoperated contralateral femora. Results of torsional testing were evaluated for torsional strength, torsional displacement, total energy to fracture and White fracture mode. Data analysis showed lower torsional strength of the operated vs. unoperated femora with the exception of morsellized COLLAGRAFT15 material, which had higher strength. However, the only difference in the operated groups was that the morsellized COLLAGRAFT15 had greater strength than several groups including the autogenous bone group. There was no difference found in angular displacement between any of the groups. However, there was a difference in the energy to fracture in both strip forms of the COLLAGRAFT15. The final conclusion is that in this model, grafting with COLLAGRAFT15 provided torsional properties at one year postoperatively at least equivalent to autogenous bone. © 1994 John Wiley & Sons, Inc.

    Notes: Hydrogles, sensitive to pH, were prepared by copolymerizing 2-hydroxyethyl methacrylate (HEMA) with 2-dimethylaminoethyl methacrylate (DMA). The mole fraction of the DMA monomer present during synthesis was fixed at 0.3, while the volume fraction of total monomer varied from 0.29 (I) to 1.0 (II). Swelling equilibria were measured in citrate of phosphate buffer for pH from 3.0 to 7.6 Swelling of the hydrogels depends on the volume fraction monomer present during synthesis, solution, pH, and identity of the buffer, Hydrogel I reached a four-fold larger swelling ratio (g swollen hydrogel/g dry hydrogel) in pH 3.0 citrate buffer, as compared to hydrogel II. Experimental results are interpreted on the basis of ideal Donnan equilibria. © 1994 John Wiley & Sons, Inc.

    Notes: Fluorocarbon elastomer seals were subjected to single stress radiation and temperature tests and combined stress tests with the seals immersed in silicone oil, to obtain a service life estimate. Their sealing capability was mechanically tested during the tests. In single environment tests the seals were resistant to temperature-induced stresses and very hard to radiation, showing no failures up to 108 rads. The combined tests, using three matched pairs of temperature and radiation dose rate, indicated a reduction in service life from 40 years at 145°C with no radiation to 21 years at 50°C with radiation, pointing to a significant synergism between radiation and temperature.

    Notes: Tricalcium phosphate (TCP) is an osteoconductive bioceramic which, when applied to a porous-coated prosthesis, may enhance osseous ingrowth and mechanical stability. TCP plasma-sprayed and unsprayed porous-coated tibial intramedullary rods were bilaterally implanted in seven adult rabbits. All rabbits were killed at 12 weeks. Pull-out tests were performed on 4 rabbits while all were evaluated histologically for osseous response and adverse tissue reaction. TCP-sprayed implants showed significantly greater osseous ingrowth in comparison to unsprayed implants. Neither implant type exhibited adverse tissue reactions. Average pull-out strengths were 69 lb for treated rods and 72 lb for controls (p 〉 0.05); quality of fit for all pull-out specimens except one was deemed poor. We conclude that plasmasprayed TCP enhances osseous ingrowth into porous-coated devices. However, our data further suggest that enhanced ingrowth may not always lead to enhanced fixation.

    Notes: The canine is frequently used as a model for human hip arthroplasty research. In order to better understand the appropriateness of the canine as a model for human total hip replacement studies, the external morphology of canine and human femurs were examined and compared. Several differences were found between canine and human femora, including angular measurements, anterior bow, and femoral head position relative to the femoral diaphysis. In addition, the human femur was noted to undergo age-related changes in several of the measured parameters. The canine femur did not exhibit any age-related changes in the measured parameters. This study suggests that there are limitations to the use of the canine model in human hip arthroplasty research, and that discretion must be exercised when attempting to extrapolate results from a canine study to the human clinical condition. © 1993 John Wiley & Sons, Inc.

    Notes: CD and nmr studies have been carried out on aqueous trifluoroethanol (TFE) solutions of bradykinin (BK) and a bradykinin antagonist. The CD results exhibit a striking effect of TFE on the spectra of BK, with sequence Arg-Pro-Pro-Gly-Phe-Ser-Pro-Phe-Arg, and the BK antagonist, with sequence D-Arg-Arg-Pro-Hyp-Gly-Thi-D-Ser-D-Cpg-Cpg-Arg [where Hyp is 4-hydroxy-L-proline; Thi refers to β-(2-thienyl)-L-alanine and Cpg refers to α-cyclopentylglycine]. The effect of increasing concentration of TFE in water on the difference ellipticity at 222 nm was examined and showed that BK may be a mixture of at least two different conformers, one of which largely forms when the TFE concentration is increased beyond 80%. The linear extrapolation of 100% of the difference ellipticity of BK at low TFE concentrations yields a value in agreement with that shown by the BK antagonist, indicating that the conformation of BK at the lower TFE concentrations is similar to that of the BK antagonist. The conformational analysis was carried out using both one-dimensional and two-dimensional 1H-nmr techniques. The total correlation spectroscopy (TOCSY) spectrum of BK in a 60/40% (v/v) TFE/H2O solution at 10°C and a nuclear Overhauser effect spectroscopy (NOESY) spectrum that shows only sequential Hα(i) - NH(i + 1) or the Hα(i) - Hδδ′(i + 1) NOEs indicate that the majority of the molecules adopt an all-trans extended conformation. The TOCSY for BK in the 95/5% (v/v) TFE/H2O solution shows that there are two major conformations in the solution with about equal population. The NOESY experiment shows two new important cross peaks for one conformation, namely Pro2(α)-Pro3 (α) and the Pro2(α)-Gly4(NH), indicating a cis Pro2-Pro3 bond and a type VI β-turn between residues Arg1 and Gly4 involving cis proline at position 3, respectively. The low temperature coefficient of Gly4 for this conformation suggests the presence of an intramolecular hydrogen bond, therefore a type VIa β-turn is present. The other conformation is all trans and extended.The BK antafonist shows difference CD spectra in TFE solutions referred to H2O that are superficially indicative of a β-bend. However, nmr speaks against this possibility, as only one set of peaks were observed in the TOCSY and NOESY experiments, indicating an all-trans extended confirmation over the range of TFE concentrations. The BK-antagonist CD data suggest that solvent perturbation of the CD of an extended confirmation perturbation of the optical activity of the thienyl moiety of the peptide since the CD spectrum of N-acetyl-β-thienyl-L-alanine N-methylamide is strongly perturbed by TFE. The present results again demonstrate the complementary relationship between CD and nmr. © 1994 John Wiley & Sons, Inc.
