    Description: Human growth hormone (hGH) elicits a diverse set of biological activities including lactation that derives from binding to the prolactin (PRL) receptor. The binding affinity of hGH for the extracellular binding domain of the hPRL receptor (hPRLbp) was increased about 8000-fold by addition of 50 micromolar ZnCl2. Zinc was not required for binding of hGH to the hGH binding protein (hGHbp) or for binding of hPRL to the hPRLbp. Other divalent metal ions (Ca2+, Mg2+, Cu2+, Mn2+, and Co2+) at physiological concentrations did not support such strong binding. Scatchard analysis indicated a stoichiometry of one Zn2+ per hGH.hPRLbp complex. Mutational analysis showed that a cluster of three residues (His18, His21, and Glu174) in hGH and His188 from the hPRLbp (conserved in all PRL receptors but not GH receptors) are probable Zn2+ ligands.     Description: A strategy of iterative site-directed mutagenesis and binding analysis was used to incorporate the receptor-binding determinants from human growth hormone (hGH) into the nonbinding homolog, human prolactin (hPRL). The complementary DNA for hPRL was cloned, expressed in Escherichia coli, and mutated to introduce sequentially those substitutions from hGH that were predicted by alanine-scanning mutagenesis and other studies to be most critical for binding to the hGH receptor from human liver. After seven rounds of site-specific mutagenesis, a variant of hPRL was obtained containing eight mutations with an association constant for the hGH receptor that was increased more than 10,000-fold. This hPRL variant binds one-sixth as strongly as wild-type hGH, but shares only 26 percent overall sequence identity with hGH.     Description: A method is described here for identifying good protease substrates among approximately 10(7) possible sequences. A library of fusion proteins was constructed containing an amino-terminal domain used to bind to an affinity support, followed by a randomized protease substrate sequence and the carboxyl-terminal domain of M13 gene III. Each fusion protein was displayed as a single copy on filamentous phagemid particles (substrate phage). Phage were then bound to an affinity support and treated with the protease of interest. Phage with good protease substrates were released, whereas phage with substrates that resisted proteolysis remained bound. After several rounds of binding, proteolysis, and phagemid propagation, sensitive and resistant substrate sequences were identified for two different proteases, a variant of subtilisin and factor Xa.     Description: We have obtained minimum age estimates for the sand units underlying the two largest meteorite deflation surfaces in Roosevelt County, New Mexico, USA, using thermoluminescence dating techniques. The dates obtained ranged from 53.5 (+/- 5.4) to 95.2 (+/- 9.5) ka, and must be considered lower limits for the terrestrial ages of the meteorites found within these specific deflation surfaces. These ages greatly exceed previous measurements from adjacent meteorite-producing deflation basins. We find that Roosevelt County meteorites are probably terrestrial contemporaries of the meteorites found at most accumulation zones in Antarctica. The apparent high meteorite accumulation rate reported for Roosevelt County by Zolensky et al. (1990) is incorrect, as it used an age of 16 ka for all Roosevelt County recovery surfaces. We conclude that the extreme variability of terrestrial ages of the Roosevelt County deflation surfaces effectively precludes their use for calculations of the meteorite accumulation rate at the Earth's surface.

    Description: The Martian southern hemisphere atmospheric water vapor column abundance measurements reported agree with Viking Orbiter atmospheric water detectors during early southern spring and southern autumnal equinox; profiles obtained in southern mid- and late summer, however, indicate the presence of twice as much water both in the southern hemisphere and planetwide. This discrepancy is accounted for by the high optical depths created by two global dust storms during the Viking year, while the present observations were obtained in the case of the relatively dust-free atmosphere of the 1988-1989 opposition.

    Description: The discovery of 154 meteorite fragments within an 11-sq km area of wind-excavated basins in Roosevelt County, New Mexico, permits a new calculation of the accumulation rate of meteorite falls at the earth's surface. Thermoluminescence dating of the coversand unit comprising the prime recovery surface suggests the maximum terrestrial age of the meteorites to be about 16.0 ka. The 68 meteorite fragments subjected to petrological analyses represent a minimum of 49 individual falls. Collection bias has largely excluded carbonaceous chondrites and achondrites, requiring the accumulation rate derived from the recovered samples to be increased by a factor of 1.25. Terrestrial weathering destroying ordinary chondrites can be modeled as a first-order decay process with an estimated half-life of 3.5 + or - 1.9 ka on the semiarid American High Plains. Having accounted for the age of the recovery surface, area of field searches, pairing of finds, collection bias and weathering half-life, an accumulation rate of 940 falls/a per 10 to the 6th sq km is calculated for falls greater than 10 g total mass. This figure exceeds the best-constrained previous estimate by more than an order of magnitude. One possible reason for this disparity may be the extraordinary length of the fall record preserved in the surficial geology of Roosevelt County. The high accumulation rate determined for the past 16 ka may point to the existence of periods when the meteorite fall rate was significantly greater than at present.

    Description: A spatial profile of the distribution of sodium in Io's corona has been constructed using measurements obtained during satellite mutual eclipses. The data reveal a fairly symmetric corona whose density falls steeply from the surface out to 6 r(Io) and more slowly outside. An upper limit of 700 km is placed on the exobase altitude, but the observations do not constrain the surface density. Several theoretical models adequately match some traits of the corona, but none satisfies all the observations. No strong upstream/downstream asymmetry of the corona is observed, so it is unlikely that the corona is primarily generated by the impact of corotating ions into the trailing hemisphere.

    Description: Results are presented from Apollo satellite observations (from May 1988 to July 1991) of sodium and potassium in the lunar atmosphere. The observations of Na and K show a wide range of scale heights, single-component temperatures, and surface number densities, while the column abundances remain within a factor of 3. The observed trends can be explained using a model of competing release mechanisms with different dependences on solar zenith angle and resulting two-component atmospheres. The theory is applied to the budget of atomic oxygen.
