Notizen: The calculation of the uncertainty in an estimated rotation requires a parametrization of the rotation group; that is, a unique mapping of the rotation group to a point in 3-D Euclidean space, R3. Numerous parametrizations of a rotation exist, including: (1) the latitude and longitude of the axis of rotation and the angle of rotation; (2) a representation as a Cartesian vector with length equal to the rotation angle and direction parallel to the rotation axis; (3) Euler angles; or (4) unit length quaternions (or, equivalently, Cayley-Klein parameters). The uncertainty in a rotation is determined by the effect of nearby rotations on the rotated data. The uncertainty in a rotation is small, if rotations close to the best fitting rotation degrade the fit of the data by a large amount, and it is large, if only rotations differing by a large amount cause such a degradation. Ideally, we would like to parametrize the rotations in such a way so that their representation as points in R3 would have the property that the distance between two points in R3 reflects the effects of the corresponding rotations on the fit of the data. It can be shown mathematically that this is impossible, but for rotations of small angle, it can be done to close approximation by using vectors in Cartesian coordinates. Thus, we are led to parametrizing the uncertainty separately from the parametrization of the best fitting rotation. This approach results in simpler, more efficient calculations than if uncertainties are described in terms of rotation parameters (i.e., latitude, longitude, and the angle). We illustrate this with the example of equations for determining the uncertainty in a composite rotation from the uncertainties of its constituents.

Notizen: The structure, genomic organization and transcription of the gene encoding histone H2B in the protozoan parasite Trypanosoma cruzi have been studied. This gene consists of a 746-nucleotlde unit, tandemly repeated at least 18 times in each of two clusters. DNA probes corresponding to histones H2B and H3 hybridized to different chromosomes revealing that the genes coding for these two histones are not physically linked in the genome of T. cruzi. The primary transcription product of the H2B gene is processed by trans-splicing and polyadenylation. Inhibition of DNA synthesis with aphidicolin resulted in the reduction of histone H2B mRNA to undetectable levels in about two hours, suggesting that its abundance is regulated throughout the cell cycle as it occurs in other eukaryotes. in addition, a concomitant inhibition of translation by cycloheximide reverted this effect indicating that de novo protein synthesis is required for RNA instability. Histone mRNA abundance was dependent on the life-cycle stage of T. cruzi: abundant in amastigotes and epimastigotes, the dividing forms in the host cell and the insect vector, respectively, white undetected in trypomastigotes, the parasite's non-dividing life stage.

Notizen: Although the phylogenetic relationships of the major groups of fishes have been extensively studied with morphological characters, not all have been convincingly resolved. Analyses of molecular sequences from these groups may provide additional insights into problematical relationships, but are only just beginning to appear. We compare our own results from analyses of 18s ribosomal RNA sequences with those of other studies using globins, parvalbumins, insulin, 28s ribosomal RNA, and portions of two mitochondria1 genes (12S ribosomal RNA and cytochrome b). Our evaluation of these studies reveals some of the difficulties encountered in reconstructing ancient divergences within the fishes, including unequal rates of evolution (among regions of a molecule as well as among lineages), gene duplication, extinction of lineages, and a possible rapid radiation of gnathostome higher taxa. The importance of evaluating the robustness of particular phylogenetic hypotheses is stressed. Some molecules appear to be inappropriate for investigating higher level divergences within the fishes; others are more promising, but must be examined in more taxa to allow an adequate evaluation of their utility. Convincing support for particular hypotheses of relationship will ultimately require congruence of trees generated from independent molecular data sets.

Notizen: In combination with observations in spring staging and wintering grounds in western Europe, a detailed etho-ecological study of nesting dark-bellied brent geese Branta b. bernicla in western Taimyr, Krasnoyarsk, Russia, was made in 1990. Most brent geese arrived on the breeding grounds from 14–19 June and started nesting within a few days. In the study area 264 nests of breeding brent geese were found, mainly on islands but also along small rivers on the mainland. The mean clutch size was 3.0 and 80% of the eggs hatched. Time budget studies showed that incubating females spent on average 138 minutes per 24 hours on feeding. Despite favourable weather conditions and a low density of arctic foxes, only about one-third of the mature birds in the study area bred. In the autumn an intermediate breeding success of 20% juveniles was recorded in the wintering areas. This was probably due to the relatively poor condition in which the brent geese left their spring staging areas.

Notizen: Abstract Saccharomyces cerevisiae genes encoding functions necessary for inhibition by the Pseudomonas syringae pv. syringae cyclic lipodepsipeptide, syringomycin-E, were identified by mutant analyses. Syringomycin-E-resistant mutants were isolated, shown to contain single recessive mutations, and divided into eight gene complementation groups. Representative strains from five groups were resistant to nystatin, and deficient in the plasma membrane lipid, ergosterol. All of the mutant strains were resistant to the related cyclic lipodepsipeptides, syringotoxin and syringostatin. The findings show that: 1) at least eight gene-encoded functions participate in the inhibitory response to syringomycin; 2) ergosterol is important for this response; 3) the three related lipodepsipeptides have similar modes of action.

Notizen: Abstract Within the framework of the hydrodynamic model the exact coupling constants of the coupling of the giant multipole resonances to the surface quadrupole oscillations of nuclei are derived. The differences to former variational calculations are discussed. It is found that the coupling constants for the terms linear in the surface variables are almost the same whereas the coupling constants for the quadratic terms differ appreciably. The influence on the photon absorption and scattering cross sections of spherical nuclei is discussed.
