    Notes: Fourteen tryptic peptides and nine intermediates were identified as products of trypsin digestion of reduced and S-3-(trimethylated amino) propylated lysozyme. Kinetics of the appearance and disappearance of these products were observed by monitoring the peak areas on the chromatogram. In spite of the complicated reaction pathways, kinetics of the digestion of proteins and several intermediate products show simple decay curves with a single rate constant. In this paper, the trypsin susceptibility of the individual cleavage site is defined as a hydrolytic rate constant of the susceptible peptide bond in the presence of 10 nM trypsin. The cleavage sites of unfolded lysozyme are classified into two groups in terms of the trypsin susceptibility: one has a high susceptibility (10-20 h-1) and the other a low susceptibility (1.0-2.0 h-1). In the unfolded state of lysozyme, in conclusion, the region from residues 15 to 61 has a strong resistance to trypsin digestion; on the other hand, the C-terminal half of the polypeptide chain is flexible enough to fit into the active site of trypsin.In addition, six kinds of pentapeptides were synthesized as analogues of lysozyme fragments including Arg 14, Arg 21, Lys 33, Arg 45, Arg 61, and Arg 73. Kinetics of typtic digestion of them were observed. Both kcat and KM were determined for these synthetic pentapeptides. The susceptibility of each cleavage site in pentapeptides is determined and compared with that corresponding in proteins. The susceptibility is usually higher when the susceptible peptide chain is flexible. However, susceptibilities of a few sites in proteins are lower than those in pentapeptides. This means that the peptapeptides, this means that the peptide chains tend to fold locally to prevent trypsin from binding to the sites. It was found that the sites of Arg 21 and Arg 45 are indeed resistant to trypsin, but the site of Lys 33 is not so much, although the hydrolytic rate at Lys 33 itself is extremely slow. © 1994 John Wiley & Sons, Inc.

    Notes: The crystal structure of (-)-corycavinium (+)-10-camphorsulfonate has been investigated by X-ray analysis. The structure of (-)-corycavinium ion ( = (-)-(7S,13S,14R)-5,6,13,13a-tetrahydro-13a-hydroxy-7-methyl-2,3;9,10-bis(methylenedioxy)-8H-dibenzo[a,g]quinolizinium), has been determined. The conformation with B/C-cis-conjunction, a twisted half-chair of ring B, and a half-chair of ring C, as well as α-oriented substituted groups N…Me, C…Me, and C…OH is revealed. Feeding experiments with cell suspension cultures of Corydalis incisa (Papaveraceae) defined the intermediacy of (-)-corycavinium in the route from protoberberine-type to hexahydrobenzo[c]phenanthridine-type of alkaloids. On the basis of the present crystal conformation, the stereospecificity of the relating enzyme is biogenetically considered.

    Notes: The ESR study of the radicals formed in mechanochemical polymerization of acrylamide (AAM) was undertaken. The nature of the radical formation was compared with the mechano-radical formation produced by mechanical fracture of polyacrylamide (PAAM). The structure of radicals formed were all identified to be an end-chain radical, equivalent to a polymer-chain propagating radical. The computer simulation disclosed that the observed spectra of the propagating radical can be approximated essentially by two kinds of component spectra, a large amount of triplet, and a small amount of quarter (a triplet of doublets). Thus, the existence of two major conformers in a single end-chain radical has been proposed. The ESR kinetics of the radical formation were further studied and its correlation with the nature of polymerization (the changes in molecular weight and polymer conversion) was discussed.

    Notes: Plasma-induced polystyrene radicals were first studied by electron spin resonance (ESR). The room temperature ESR spectrum was compared with those obtained by γ-irradiation, UV-irradiation, and mechanical fracture. It was found that even less than a few seconds of plasma-irradiation gave rise to a large amount of polystyrene radicals and the ESR spectrum consisted of two types of spectra, a triplet and a single broad line. The spectral feature of the triplet was nearly identical with that of γ-irradiated polystyrene. Thus, it was assigned to the structure of a cyclohexadienyl-type radical formed by a nearly random addition of a hydrogen atom to the aromatic ring. The single broad line, thought to be an outline of multicomponent spectrum, was assigned to an immobilized dangling-bond sites at the plasma-induced crosslinked portion of the polystyrene surface.

    Notes: We have examined the effects of in vitro aging on the growth capacity of human umbilical vein endothelial cells (HUVECs) under the influence of tumor necrosis factor (TNF) with or without interferon-γ (IFN-γ). The growth and colony-forming abilities of control cells were impaired with advancing age in vitro, especially at later stages (more than 70-80% of life span completed). It was found that treatment with TNF inhibited growth and colony-forming efficiency at any in vitro age. The effects of TNF were shown to increase with increasing in vitro age, as reflected by a more pronounced increase in doubling times, a decrease in saturation density, and a reduction in colony-forming efficiency. However, the characteristics of TNF receptors, including the dissociation constant, and the number of TNF-binding sites per cell-surface area remained rather constant. The effect of TNF was augmented by IFN-γ at a dose that alone affected growth and colony formation only slightly. The augmentation by IFN-γ was also found to depend on in vitro age; the synergy with TNF in the deterioration of colony-forming ability was observed only in “aged” cells. These results suggest that the intrinsic responsiveness of HUVECs to growth-inhibiting factors, as well as to growth-stimulating factors, changes during aging in vitro.

    Notes: Shear stress P21 and normal stress P11 -P22 functions of moderately concentrated aqueous solutions of sodium poly (acrylate) were measured under steady shear flow at various shear rates \documentclass{article}\pagestyle{empty}\begin{document}$\dot \gamma$\end{document} with a Weissenberg rheogoniometer. The compliance function Js = (P11 - P22)/2P212 is found to vary with shear rate. The double logarithmic plot of Js versus \documentclass{article}\pagestyle{empty}\begin{document}$\dot \gamma$\end{document} can be superposed with respect to molecular weight at constant concentrations of added neutral salt and polymer. These reduced plots differ markedly as the concentration of added salt is varied but converge to a single line at high shear rates.

    Notes: The potentiometric titration of random copolymers of L-lysine and L-alanine containing 0-35% alanine was carried out. The standard free-energy change for the transition of coil to helix was calculated from the titration curve, and was treated by taking account of first-neighbor interactions. For uncharged lysine ΔG° = -140 cal/mole, and for alanine ΔG° = -50 cal/mole in 0.06M NaBr at 25°C, indicating that the alanine helix is thermodynamically less stable than the lysine helix. The randomness in co-polymerization was confirmed by trypsin treatment.

    Notes: As model peptides of collagen, (Pro-Pro-Gly)n (n = 10, 12, 14, and 15) and (Pro-Pro-Gly)n(Ala-Pro-Gly)m(Pro-Pro-Gly)n (2n + m = 15; m = 1, 3, and 5) were synthesized by the solid-phase method.     Notes: Measurements of the molecular weight of (Pro-Pro-Gly)n and (Pro-Pro-Gly)n(Ala-Pro-Gly)m(Pro-Pro-Gly)n, which were synthesized by the solid-phase method, revealed that they formed a trimer in an aqueous solution, and dissociated into single-stranded chains on warming. Accompanying the transition, a large decrease of optical rotation was observed, like the collagen-gelatin transition. The shape of the trimeric molecule was rodlike, and the dimensions were 12 Å in diameter and 2.8 Å per residue in length, regardless of the length of Ala-Pro-Gly sequences in a peptide chain. The data indicate that both Pro-Pro-Gly sequences and Ala-Pro-Gly sequences from the triple-helical structure similar to that of collagen in aqueous solution. All optical rotational dispersion (ORD) curves of solutions of the peptides were represented by a single-term Drude equation, and the Drude constant λc was 200 nm for all peptides regardless of the length of Ala-Pro-Gly sequences. The resemblance between the helical structure formed by Pro-Pro-Gly sequences and that by Ala-Pro-Gly sequences was also suggested by the formation of the hybrid triple helix from two kinds of peptide chains with different lengths of Ala-Pro-Gly sequences.
