    Notes: Summary The thiolase family is a widespread group of proteins present in prokaryotes and three cellular compartments of eukaryotes. This fact makes this family interesting in order to study the evolutionary process of eukaryotes. Using the sequence of peroxisomal thiolase from Saccharomyces cerevisiae recently obtained by us and the other known thiolase sequences, a phylogenetic analysis has been carried out. It shows that all these proteins derived from a primitive enzyme, present in the common ancestor of eubacteria and eukaryotes, which evolved into different specialized thiolases confined to various cell compartments. The evolutionary tree obtained is compatible with the endosymbiotic theory for the origin of peroxisomes.

    Notes: Summary We have previously suggested that two positioned nucleosomes are removed from the promoter of the Saccharomyces cerevisiae SUC2 gene upon derepression by glucose starvation. To gain further insight into the changes accompanying derepression at the chromatin level we have studied the chromatin structure of the SUC2 promoter in several mutants affecting SUC2 expression. The non-derepressible mutants snf1, snf2 and snf5 present a chromatin structure characteristic of the repressed state, irrespective of the presence or absence of glucose. The non-repressible mutants, mig1 and ssn6, as well as the double mutant snfs sn6 exhibit an opened chromatin structure even in the presence of glucose. These results suggest that the DNA-binding protein encoded by MIG1 is necessary to produce the characteristic pattern of repressed chromatin and that the SNF1 protein kinase is sufficient to produce the derepressed chromatin pattern. A model is presented for the transitions that result in opening up of the chromatin structure.

    Notes: We Propose a kinetic expression which accounts for the temperature dependence of ethanol yield losses in batch alcoholic fermentation. Moreover, the characteristic parameters of the microbial growth equation have been calculated for Saccharomyces cerevisiae under typical wine industry conditions. A substrate consumption equation is established which minimizes possible model deviations in the latter process stages. Experimental data were obtained in the laboratory and the proposed equations were then applied at an industrial level (2.5 × 104 L) where they described the data well.

    Notes: We have studied the chromatin structure of the Saccharomyces cerevisiae FBP1 gene, which codes for fructose-1,6-bisphosphatase. A strong, constitutive, DNase I, micrococcal nuclease and S1 nuclease hypersensitive site is present close to the 3′ end of the coding region. In the repressed state, positioned nucleosomes exist around this site, and subtle changes occur in this nucleosomal organization upon derepression. A DNase I hypersensitive region is located within the promoter between positions -540 and -400 and it extends towards the gene in the derepressed state, leading to an alteration of nucleosomal positioning. Psoralen crosslinking of chromatin, which is used for the first time to study the mobility of restriction fragments from an RNA polymerase II gene, revealed that part of the promoter is nucleosome-free, in accordance with the results of DNase I digestion. A model is presented that, based on the chromatin structure, puts forward the hypothesis that the promoter UAS is located between - 540 and - 340. Finally, psoralen crosslinking, as well as digestions with micrococcal nuclease or restriction endonucleases suggests that most if not all of the copies of the active FBP1 gene are covered by nucleosomes.

    Notes: We have previously shown that some changes occur in the chromatin structure of the 3′ flank of the yeast SUC2 gene in going from a repressed to an active state. In an attempt to find out the causes of these changes, we have carried out experiments in which mutant copies of SUC2 locus lacking either 5′ or 3′ flanks have been analysed for their transcriptional activity and chromatin structure. These experiments allowed us to discard any relationship between SUC2 transcription and chromatin changes within its 3′ flank. Sequencing of this flank and mRNA analysis, however, resulted in the location of a putative peroxisomal 3-oxoacyl-CoA thiolase gene (POT1), which is repressible by glucose. The disruption of the gene produced a yeast strain unable to use oleic acid as a carbon source. This is the first time that chromatin structure analysis has permitted the identification of new gene.

    Notes: The LUC gene coding for Photinus pyralis firefly luciferase was cloned in different yeast episomal plasmids in order to assess its possibilities as an in vivo reporter gene. Activity of the enzyme in transformed cells in vivo was measured by following light emission and assay conditions optimized in intact cells, with regard to oxygen concentration, temperature, cell concentration in assay mixtures and external ATP concentration. Among the factors tested, light emission was drastically influenced by the external pH in the assay (which resulted in a ten-fold amplification signal) and by substrate permeability. The growth phase of the cells was also important for the level of activity detected. Cloning of firefly luciferase gene under the control of different yeast-regulated promoters (ADH1, GAL1-10) enabled us to measure their strength which correlated well with previously described data. We conclude that firefly luciferase is an adequate gene reporter for the in vivo sensitive determination of gene expression and promoter strength in yeast.

    Description: The LBV characteristics of the new LBV candidate WRA 751 suggested by Hu et al. (1990) are studied in greater detail. A comparison is made with observational characteristics of the well-known LBVs Ag Car and HR Car using photometric and spectroscopic data of Hu et al. supplemented with an IUE spectrum and direct CCD images in H-alpha, blue, and red. It is found that the spectra of WRA 751 and HR Car discussed in this paper are similar to the minimum-phase spectrum of AG Car and therefore resemble the Of/WN 9 spectral type. The characteristics of the optical, near- and far-UV (IUE) spectra of the Fe II spectrum in these wavelength regions is due to continuum fluorescence. The H, He I, Fe II and forbidden Fe II emission lines are formed in different parts of the stars' envelopes, corresponding to their expanding velocities. It is concluded that WRA 751 has properties which fit well the LBV properties at minimum-brightness phase, and possesses circumstellar material with observational properties similar to those of the known LBVs AG Car and HR Car.

    Description: We present additional photometric observations (Stromgren y filter) of the Herbig Ae Star HR 5999. This new set of data, composed of 282 data points covering April 1983 to August 1989, was used in conjunction with the 362 data points compiled by Baade and Stahl (1989). Our aim was to detect single or multiple periods in the integrated set of data spanning nearly 20 yr. Through the use of series analysis techniques, and by removing any linear component present in the raw data, we were able to detect weak peaks in the power spectra (in the order of intensity the strongest peaks are at 301 and 113 d) which were not self-evident in the phase diagrams. The overall picture of the data shows a small linear component, which is more noticeable in the new set of data, indicating that the recent maxima appear brighter. By a detailed analysis of the full light curve we were able to detect the presence of well-defined 'pulses' or bursts. Further Gaussian fits of these bursts indicated that the pulses are relatively rapid, of the order of 10 d, with some of them being closely spaced. No periodicity was found for the pulses. A possible explanation of these aperiodic outbursts is that they detect a flow of matter accompanied by magnetic field disturbances originated from the interior of the star, unlike those originating externally such as binary perturbations, which tend to be periodic.

    Description: High-speed U-band photometry of the peculiar emission object MWC 560 obtained with the ground-based instrumentation, and V-band photometry obtained with the International Ultraviolet Explorer-Fine Error Sensor indicates irregular brightness variations are quasi-periodic. Multiple peaks of relative brightness power indicate statistically significant quasi periods existing in a range of 3-35 minutes, that are superposed on slower hourly varying components. We present a preliminary model that explains the minute and hourly time-scale variations in MWC 560 in terms of a velocity-shear instability that arises because a white dwarf magnetosphere impinges on an accretion disk. We also find evidence for Fe II multiplet pseudocontinuum absorption opacity in far-UV spectra of CH Cygni which is also present in MWC 560. Both CH Cyg and MWC 560 may be in an evolutionary stage that is characterized by strong UV continuum opacity which changes significantly during outburst, occurring before they permanently enter the symbiotic nebular emission phase.

    Description: The newly discovered hot-emission line star, HD 147196 in the Rho Oph dark cloud region was observed spectroscopically and photometrically and high and low resolution IUE spectra were obtained. The finding of Irvine (1990) that this relatively bright star show its H-alpha-line in emission is confirmed. Previous H-alpha-surveys of the Rho Oph star-forming region did not detect HD 147196 as an H-alpha-emission star, meaning that it must recently be very active and has perhaps transformed itself from a B-type star at shell phase to a Be-phase. The Mg II h + k resonance lines are in absorption and they appear to be interstellar in nature, which means that either the abundance of Mg in the extended atmosphere of the star is low or that the shell is not extended enough to produce emission lines of Mg II. Photometric observations of this B8 V type star do not show any variations during at least the years covered by our monitoring or any excess of NIR radiation in its spectral energy distribution up to the M-passband at 4.8 microns.
