    Notes: We have measured the specific viscosity of closed circular PM2 DNA in the presence of concentrations of ethidium bromide ranging up to 5 mg/ml. Both native viral PM2 DNA I and enzymatically prepared relaxed, closed circular PM2 DNA I0 exhibit a complex dependence of the specific viscosity upon the extent of supercoiling. As the number of superhelical turns is increased in the positive sense from zero, the viscosity first decreases to a minimum, then passes through a secondary maximum, and eventually again increases as the dye-induced duplex unwinding proceeds. In the case of DNA I, a corresponding behavior is mirrored in the negative sense as dye is removed from the principal viscometric maximum (complete relaxation of the DNA by dye). The shape of the curve relating specific viscosity to extent to supercoiling is similar for superhelical DNAs of either handedness, a result which we interpret to mean that the influence of any regions of special secondary structure (such as denatured loops) upon the viscosity is minimal. At very high dye concentrations the specific viscosity decreases dramatically. This effect might arise either from intermolecular aggregation or from a dye-induced collapse in the DNA secondary structure.

    Notes: Two black yeast polysaccharides show similar non-Newtonian behavior when the effects of polysaccharide concentration, shear rate, pH, salt, and temperature on the viscosity of dispersions are examined. Both polysaccharides readily form viscous aqueous dispersions and can be cast into flexible films, and both stabilize oil-water emulsions. Each of these two polysaccharides occurs extracellularly in the culture fluids of two different black yeast-like fungi - Rhinocladiella elatior Mangenot (strain NRRL YB-4163) and Rhinocladiella mansonii (Castellani) Schol-Schwarz (strain NRRL Y-6272). This report appears to be the first on the rheology of an extracellular polysaccharide which contains the unusual sugar N-acetyl-D-glucosaminuronic acid.

    Notes: Bone remodeling around three different endossesous dental implant designs placed in dog mandibles was studied using radiography during lengthy periods of function and by histology after animal sacrifice. The three designs investigated were (a) threaded (c.p. titanium), (b) fully porous-coated (titanium alloy), and (c) partially porous-coated (titanium alloy). The implants were kept in function for either 32 weeks (fully porous-coated) or 73 to 77 weeks (partially porous-coated and threaded). The studies indicated that some crestal bone loss occurred for both the threaded and partially porous-coated implants while no significant bone loss was seen with fully porous-coated implants in the absence of plaque-associated infection. It is suggested that these observed differences are a result of the different stress states that develop in bone surrounding the three designs underlying the importance of implant design on bone remodeling.

    Notes: Depth profiles calculated using simple peak-to-peak or peak height or area measurements are subject to inaccuracies caused by spectral noise, sloping backgrounds and possibly by overlapping spectra of different elements or chemical species. An automated program that reduces or eliminates the effects of these potential sources of error in the peak heights and areas calculated from electron spectra has been developed. The program uses linear least-squares fitting of one or more basis spectra to determine the peak height or area for each spectrum recorded during the profile. The program selects the basis spectra from the energy region data set without operator intervention. The number of unique lineshapes is determined by the program, again without operator intervention, and a profile is calculated for each species. The fit method of intensity determination was found to increase the dynamic range by reducing false contributions to the calculated intensities; this observation holds when a single basis spectrum is sufficient to model the data. Contributions from overlapping spectra are resolved if an example of each pure species lineshape is available in the regional data set. The method is demostrated by practical example.
